The aim of this study was to evaluate the tissue compatibility of a silorane-based resin system (FiltekTM Silorane) and a methacrylate-based nanoparticle resin (FiltekTM Supreme XT) after implantation in the subcutaneous connective tissue of isogenic mice. One hundred and thirty five male isogenic BALB/c mice were randomly assigned to 12 experimental and 3 control groups, according to the implanted material and the experimental period of 7, 21 and 63 days. At the end of each period, the animals were killed and the tubes with the surrounding tissues were removed and processed for microscopic analysis. Samples were subjected to a descriptive and a semi-quantitative analyses using a 4-point scoring system (0-3) to evaluate the collagen fiber formation and inflammatory infiltrate. Data were statistically analyzed using the Kruskal Wallis test (?=0.05). The results showed that there was no significant difference between the experimental and control groups considering the three evaluation periods (p>0.05). Color stability of silorane-based composites submitted to accelerated artificial ageing-An in situ study

Objectives: To assess the in situ color stability, surface and the tooth/restoration interface degradation of a silorane-based composite (P90, 3M ESPE) after accelerated artificial ageing (AAA), in comparison with other dimethacrylate monomer-based composites (Z250/Z350, 3M ESPE and Esthet-X, Dentsply). Methods: Class V cavities (25 mm(2) x 2 mmdeep) were prepared in 48 bovine incisors, which were randomly allocated into 4 groups of 12 specimens each, according to the type of restorative material used. After polishing, 10 specimens were submitted to initial color readings (Easyshade, Vita) and 2 to analysis by scanning electronic microscopy (SEM). Afterwards, the teeth were submitted to AAA for 384 h, which corresponds to 1 year of clinical use, after which new color readings and microscopic images were obtained. The values obtained for the color analysis were submitted to statistical analysis (1-way ANOVA, Tukey, p < 0.05). Results: With regard to color stability, it was verified that all the composites showed color alteration above the clinically acceptable levels (Delta E >= 3.3), and that the silorane-based composite showed higher Delta E (18.6), with a statistically significant difference in comparison with the other composites (p < 0.05). The SEM images showed small alterations for the dimethacrylate-based composites after AAA and extensive degradation for the silorane-based composite with a rupture at the interface between the matrix/particle. Conclusion: It may be concluded that the silorane-based composite underwent greater alteration with regard to color stability and greater surface and tooth/restoration interface degradation after AAA. OBJETIVO: caracterizar a interação entre tensão e cinética de polimerização e seus parâmetros determinantes, e avaliar a qualidade da rede polimérica e propriedades mecânicas de compósitos comerciais para dentes posteriores a base de dimetacrilato ou de silorano. A cinética de polimerização foi avaliada sob influência da intensidade da fonte de luz, da temperatura e da umidade. Por fim, o grau de conversão (GC) do compósito a base de silorano foi avaliado por espectroscopia e titulação. MÉTODOS: foram utilizados três compósitos de baixa contração: Kalore (KAL - GC America) e N´Durance (NDUR Septodont), a base de dimetacrilatos; e Filtek P90 (P90 - 3MESPE), a base de silorano. O EsthetX HD (EHD - Dentsply) foi utilizado como controle. A tensão de polimerização (TP) dos espécimes (n = 5; Ø = 5 mm; h = 1 mm, 10 min) foi obtida em máquina universal de ensaios mecânicos. O objetivo deste estudo foi avaliar, in vitro, o comportamento citotóxico e a produção de citocinas induzidos pelos materiais restauradores resinosos contendo novos monômeros KaloreTM (GC FUJI) e FiltekTMSilorane (3M ESPE) em comparação com as resinas compostas convencionais Charisma® (Heraeus-Kulzer) e FiltekTM Z250 (3M ESPE), em cultura de fibroblastos L929 e macrófagos RAW 264.7 de camundongos. As células foram estimuladas com as resinas compostas, fotopolimerizadas ou não, a partir da colocação em contato indireto ou pela extração de seus componentes durante 15, 45 e 120 dias. Após a incubação por 6, 12 e 24 horas, a viabilidade celular foi avaliada pelo Ensaio MTT e a produção de citocinas foi investigada pelo ensaio imunoenzimático (ELISA). Os dados obtidos foram analisados utilizando-se análise de variância de uma via (One-way ANOVA) e pós-teste de Tukey (α=0,05). A produção de TNF-α e IL-6 não foi detectada em fibroblastos da linhagem L929, após 6, 12 e 24 horas de contato indireto com as resinas compostas KaloreTM e FiltekTM Silorane. Compostos orgânicos podem ser liberados dos materiais resinosos, mesmo após sua polimerização, como resultado da presença de monômeros residuais e do processo de degradação do próprio material, podendo ocasionar efeitos citotóxicos, genotóxicos e alergênicos. O objetivo do presente estudo, in vitro, foi avaliar a liberação de compostos orgânicos de dois materiais resinosos, recentemente lançados no mercado, que apresentam inovações em suas formulações (resinas Kalore TM - GC FUJI e FiltekTM Silorane - 3M ESPE), variando a fonte de luz polimerizadora (halógena ou LED), a solução de imersão (água ou saliva artificial) e o pH da solução de imersão (7 ou 4,5). Foram confeccionados 56 corpos de prova da resina Kalore TM e 56 da resina FiltekTM Silorane, sendo 28 polimerizados com luz halógena e 28 com luz LED. Após aleatorização, 7 corpos de prova de cada resina foram armazenados em água com pH neutro, 7 em água com pH ácido, 7 em saliva com pH neutro e 7 em saliva com pH ácido. A leitura dos espectros das soluções foi realizada por meio da espectroscopia de fluorescência após 1, 3, 24, 48, 72, 168, 216, 312, 432, 504 e 672 horas. Após 672 horas, ainda verificou-se a liberação de compostos orgânicos das resinas KaloreTM e FiltekTM Silorane em todas as condições avaliadas. A liberação de compostos orgânicos foi menor nos grupos experimentais polimerizados pela luz LED. A quantidade de compostos orgânicos liberados foi menor nas amostras imersas em saliva. A manutenção da integridade entre a interface dente e restauração é importante para o sucesso clínico de restaurações adesivas, influenciando significativamente na longevidade das mesmas. Assim, o objetivo deste trabalho in vitro, foi avaliar o efeito de diferentes sistemas adesivos na resistência de união à dentina e ao esmalte de uma resina composta a base de silorano, após 24 horas e 6 meses de armazenamento. Para este estudo, setenta terceiros molares humanos hígidos, recém-extraídos e não restaurados, foram usados em um dos dois testes de resistência de união. Para a resistência à microtração à dentina, vinte e oito terceiros molares humanos, sem raízes, tiveram a superfície oclusal planificada a fim de expor a dentina superficial. Os dentes foram aleatoriamente divididos em 8 grupos experimentais (n=7): Adper Single Bond 2 (3M ESPE); Adper Scotchbond Multiuso (3M ESPE); Adper SE Plus (3M ESPE) e sistema adesivo P90 (3M ESPE), sendo os palitos obtidos de cada grupo divididos entre os testes envelhecidos ou não. Após a aplicação do sistema adesivo relacionado com cada um dos grupos, foi aplicada uma camada do adesivo da resina à base de silorano e fotopolimerizado por 10 segundos. O objetivo do estudo foi avaliar a união de cimentos à base de metacrilato à superfície de um compósito à base de silorano, com ou sem prétratamento de superfície, em teste de microtração. No total, 84 discos de resina à base de silorano (3mmX10mm) foram confeccionados, 42 discos tiveram a superfície tratada (ST) com jatos de óxido de alumínio (50μm) e os demais (42) não tiveram a ST. The aim of this study was to investigate the influence of different composite resins - Filtek P90 (silorane-based composite) and Heliomolar (methacrylate-based composite) - on light transmission and decrease in Knoop hardness between the bottom and top of cured specimens. The irradiance of a light-curing unit (LCU) was measured with a power meter (Ophir Optronics; 900 mw/cm2) and spectral distributions were obtained using a spectrometer (USB 2000). Twenty standardized cylindrical specimens (2 mm thick x 7 mm diameter) of each composite resin were obtained by curing using the LCU for 40 s. Light energy transmission through the composite was calculated (n=10). The Knoop hardness number for each surface was recorded as the mean of 3 indentations. The difference in Knoop hardness between the top and bottom (DKH) of the same specimen was calculated (n=10). The irradiance of light that passed through Filtek P90 (272 mW/cm2) was not significantly greater than that the passed through Heliomolar (271 mW/cm2). The DKH of Filtek P90 (25%) was significantly higher than that of Heliomolar (12%). There was a greater degree of subsurface polymerization of the methacrylate-based composite compared to the silorane-based composite.

Incorporation of silver nanoparticles into composite resins is recommended for their reported antibacterial properties, but this incorporation can affect the wettability of such materials. Therefore, this study evaluated the effect of nano-silver addition to silorane-based and methacrylate-based composites on their contact angle. Nano-silver particles were added to Z250 (methacrylate-based) and P90 (silorane-based) composites at 0.5% and 1% by weight. The control group had no additions. SEM-EDX analysis was performed to confirm the homogeneity of the nano-silver distribution. Seventy-two composite discs were prepared and standardized to the identical surface roughness values, and then distributed randomly into 6 groups containing 12 samples each (N = 12). Two random samples from each group were observed by atomic force microscopy. Distilled water contact angle measurements were performed for the wettability measurement. Two-way ANOVA, followed by the Tukey-HSD test, with a significance level of 5%, were used for data analysis. It was observed that wettability was significantly different between the composites (p = 0.0001), and that the addition of nano-silver caused a significant reduction in the contact angle (p = 0.0001). We investigated the effect of sodium ascorbate (SA) on the microtensile bond strengths (MTBSs) of different composites to bovine enamel after vital bleaching with hydrogen peroxide (HP) or carbamide peroxide (CP). Thirty bovine incisors were randomly divided into five groups and treated with no bleaching application (control), 35% HP alone, 35% HP + 10% SA for 10 minutes (HP + SA), 16% CP alone, or 16% CP + 10% SA for 10 minutes (CP + SA). Specimens were restored with Silorane adhesive and Filtek Silorane composite (designated as S / group) or with Clearfil SE bond and Filtek Supreme XT (designated as F / group). Composite build-up was created on the enamel. Sectioned specimens (n = 10 per group; 1 mm2; cross-sectional area) were created and stressed in a universal testing machine at 1 mm/min crosshead speed. The application of 10% SA immediately after bleaching with 16% CP or 35% HP increased the enamel MTBS, regardless of the adhesive / composite resin used. The aim of the present study was to evaluate the influence of different surface treatments on bond strength of repair resin-based silorane through microtensile test. 160 silorane blocks were randomly distributed among 40 experimental groups (n=4) resulting from the interaction of three factors under study: surface treatment, adhesive system and resin used to repair. The blocks were stored for 9 days in deionized water at 37°C prior to surface treatment and repair. Repair was performed, after the use of a bonding agent: (Ad 1) Silorane bond or (Ad 2) Adper Scotchbond, with Filtek P90 (P90) or Filtek Z350 XT (Z350 XT) on one of the 10 treatment protocols: control (C), silane (Si), phosphoric acid (Pa), Pa plus Si (PaSi), hydrofluoric acid (Ha), Ha plus Si (HaSi), sandblasting (S), S plus Si (SSi), diamond bur (Db), Db plus Si (DbSi). Blocks were sectioned into beams and after age by storage for six months, subjected to microtensile testing and subsequent analysis in a stereoscope to define the failure types. Additionally, 10 blocks of Filtek P90 composite resin were made for surface analysis by SEM, after completion of surface treatment. Hay pocos estudios valorando los efectos biol??gicos de estos nuevos sistemas restauradores basados en resinas de tipo Silorane, y los que hay, est??n realizados b??sicamente in vitro, en cultivo de c??lulas. Nos hemos planteado evaluar la compatibilidad tisular, in vivo, frente a un nuevo sistema restaurador basado en resinas de tipo Silorane y frente a una resina en base a metacrilato. Para ello valoramos la respuesta tisular de este nuevo composite implant??ndolo en tejido subcutaneo de ratones isog??nicos y aplic??ndolo en cavidades profundas en dientes de perros.

O objetivo deste estudo foi avaliar a resistência de união e grau de conversão de um compósito a base de silorano em associação a sistemas adesivos autocondicionantes e convencionais. Para a realização do teste de microtração, foram utilizados 50 terceiros molares humanos sem raiz e sem esmalte oclusal e com a superfície dentinária exposta. As amostras foram submetidas ao desgaste da superfície dentinária com lixas de carbeto de silício de granulação 600 para a padronização da lama dentinária e divididas aleatoriamente em cinco grupos de acordo com o sistema adesivo aplicado (n=10): 3 sistemas autocondicionantes G1 - P90; G2 - Adper Easy One Bond; G3 - Adper SE Bond; e dois convencionais G4 - Adper Single Bond 2; G5 - Adper Scotchbond Multi-Uso Plus. Em seguida, foram aplicados os sistemas adesivos e a resina composta a base de silorano de acordo com as instruções do fabricante. Após o procedimento restaurador, as amostras foram seccionadas, obtendo-se palitos com largura de 0,7 - 1mm. Methyl methacrylate used in bone cements has drawbacks of toxicity, high exotherm, and considerable shrinkage. A new resin, based on silorane/oxirane chemistry, has been shown to have little toxicity, low exotherm, and low shrinkage. We hypothesized that silorane-based resins may also be useful as components of bone cements as well as other bone applications and began testing on bone cell function in vitro and in vivo. MLO-A5, late osteoblast cells, were exposed to polymerized silorane (SilMix) resin (and a standard polymerized bisGMA/TEGDMA methacrylate (BT) resin and compared to culture wells without resins as control. A significant cytotoxic effect was observed with the BT resin resulting in no cell growth, whereas in contrast, SilMix resin had no toxic effects on MLO-A5 cell proliferation, differentiation, nor mineralization. The cells cultured with SilMix produced increasing amounts of alkaline phosphatase (1.8-fold) compared to control cultures. Compared to control cultures, an actual enhancement of mineralization was observed in the silorane resin-containing cultures at days 10 and 11 as determined by von Kossa (1.8–2.0 fold increase) and Alizarin red staining (1.8-fold increase). Microtensile Bond Strength of Methacrylate and Silorane Resins to Enamel and Dentin

The aim of this study was to evaluate the microtensile bond strength (µTBS) of two substrates (enamel and dentin) considering two study factors: type of composite resin [methacrylate-based (Filtek Supreme) or silorane-based (Filtek LS)] and aging time (24 h or 3 months). Twenty human molars were selected and divided into 2 groups (n=10) considering two dental substrates, enamel or dentin. The enamel and dentin of each tooth was divided into two halves separated by a glass plate. Each tooth was restored using both tested composite resins following the manufacturer's instructions. The samples were sectioned, producing 4 sticks for each composite resin. Half of them were tested after 24 h and half after 3 months. µTBS testing was carried out at 0.05 mm/s. Data were analyzed by three-way ANOVA and Tukey's HSD tests at α=0.05. Significant differences between composite resins and substrates were found (p<0.05), but no statistically significant difference was found for aging time and interactions among study factors. The methacrylate-based resin showed higher µTBS than the silorane-based resin. The µTBS for enamel was significantly higher than for dentin, irrespective of the composite resin and storage time. Three months of storage was not sufficient time to cause degradation of the bonding interaction of either of the composite resins to enamel and dentin.

This study evaluated the temperature variation in the pulp chamber during photoactivation of two restorative composite resins (Filtek P90 silorane-based composite and Heliomolar methacrylate-based composite) with either a quartz-tungsten-halogen (QTH) or light-emitting diodes (LED) light-curing unit (LCU) and using dentin thicknesses (0.5 and 1.0 mm). Standardized cavities (2x2x2 mm) were prepared in 80 bovine incisors, which were randomly assigned to 8 groups according to the photoactivation method and dentin thickness. Filtek P90 and Heliomolar (both in shade A3) were used with their respective adhesive systems (P90 self-etch primer / P90 adhesive bond and Excite adhesive). All experiments were carried out in a controlled environment (37°C). The temperature variations (°C) were recorded using a digital thermometer attached to a K-type thermocouple. The results were analyzed statistically by ANOVA and Tukey's test (α=0.05). For composite/dentin thickness interaction, temperature increase was significantly higher in 0.5 mm dentin thickness (40.07°C) compared with 1.0 mm dentin thickness (39.61°C) for Filtek P90. For composite/LCU interaction, the temperature increase was significantly higher for Filtek P90 (39.21°C - QTH and 40.47°C - LED) compared with Heliomolar (38.40°C - QTH and 39.30°C - LED). The silorane-based composite promoted higher temperature increase in the pulp chamber than the methacrylate-based composite.

AIM: To evaluate the impact of different light-curing times on dentin microtensile bond strength of two restorative systems after 24 h and 6 months of water storage. METHODS: Standardized Class II preparations were performed in 56 freshly-extracted human molars (n = 7), restored with methacrylate- or silorane-based restorative systems, and light-cured using a light-emitting diode at 1390 mW/cm2 by the recommended manufacturers' time or double this time. After storage for 24 h at 37 oC, the teeth were sectioned to yield a series of 0.8-mm thick slices. Each slab was trimmed into an hourglass shape of approximately 0.64 mm2 area at the gingival dentin-resin interface. Specimens were tested using universal testing machine at crosshead speed of 0.5 mm/min until failure, after 24 h and 6 months of storage. Data were statistically analyzed by three-way ANOVA and Tukey's test (α = 0.05). RESULTS: The highest bond strength values were recorded for the groups restored with methacrylate system (p<0.001) as well as for extended light-curing time (p = 0.0034). INTRODUCTION: Modern composites are exceptional physically and aesthetically, but success is dependent on effective bonding to tooth structures, possibly enhanced by phosphoric acid pre-etching. AIMS: To determine the effect pre-etching of tooth surfaces has on the shear bond strengths of silorane-based and of methacrylate-based composite resin systems METHOD: To 120 recently extracted human molars and pre-molars, composite resin stubs were bonded with Silorane System Adhesive or Scotchbond Universal, to pre-etched or to non-pre-etched tooth surfaces, using an Ultradent jig. After thermocycling, each specimen was debonded under a shear force, in an Instron Universal testing machine (crosshead speed 0.5mm/minute. RESULTS: Data were subjected to an analysis of variance and when significant, to pairwise comparisons. The silorane-based system achieved significantly higher mean shear bond strengths (MPa) on pre-etched surfaces of: uncut enamel (15.204 vs 9.424), cut enamel (21.352 vs 12.110) and dentine (19.787 vs 8.278). Methacrylate-based bonds to pre-etched uncut enamel were significantly stronger (28.898 vs 18.451). Pre-etching cut enamel enhanced the bond (20.548 vs 16.384, not significant).
