Compósitos mesoporosos de ZrO2-CeO2 estão sendo desenvolvidos devido às suas excelentes propriedades morfológicas e estruturais, necessárias para várias aplicações, que incluem sensores de gás, catálise automotiva e ânodos de células a combustível de óxido sólido. Nesse trabalho foi desenvolvido um novo método de síntese sol-gel com template cooperativo utilizando o polímero tribloco P-123 e os cloretos de Zr/Ce como precursores dos óxidos. Foram sintetizados ZrO2-x(mol)%CeO2 com x = 50, 70 e 90% de CeO2, uma vez que esses materiais apresentam melhores características para aplicações catalíticas. Dois processos de calcinação diferentes foram testados (até 540 e 400ºC). O NiO (60% m/m) foi impregnado para que o material obtenha a condutividade eletrônica necessária para aplicação em ânodos de SOFC. Os resultados de difração de raios X indicaram sistemas cuja fase cristalográfica predominante é a cúbica tipo fluorita (a fase tetragonal é minoritária). Fase única cúbica foi obtida para 90% de CeO2 após a calcinação até 400ºC. Dentre as características morfológicas dos materiais calcinados até 540ºC, os resultados de adsorção/dessorção de N2, imagens de microscopia eletrônica e espalhamento de raios X a baixos ângulos apresentaram aglomerados cristalinos de ZrO2-CeO2...

Biologia.; Embora muitos metais, como o cobre, sejam essenciais para os organismos, o seu uso industrial e na agricultura tem levado a um aumento de sua concentração na natureza. Uma vez em altas concentrações, este metal pode tornar-se tóxico e ameaçar a saúde de organismos marinhos. Além disso, em corpos aquáticos, é freqüentemente relatada a presença de compostos protetores, como o selênio. Desta forma, este trabalho foi dividido em duas partes: na primeira, mexilhões Perna perna foram expostos a diversas concentrações de cobre por 96 horas, a fim de se ter uma estimativa da toxicidade deste metal. A concentração letal 50% em 96 horas foi de 245 μgl-1 (CL50,96h) e a maior concentração sem efeito foi a de 80 μgl-1 (CL0,96h). Após esta etapa, os animais foram expostos à 30, 60, 180 e 540 μgl-1 de cobre, a fim de determinar danos oxidativos em brânquias. Houve um aumento nos níveis de glutationa total para todas as concentrações testadas, exceto para 30 μgl-1. A atividade da enzima glutationa redutase foi menor nos animais expostos a 540 μgl-1, e a atividade da tioredoxina redutase foi menor em todas as concentrações exceto na de 60 μgl-1. O uso de misturas de glyphosate, em tanque, para manejo de espécies de plantas daninhas de difícil controle tem sido prática comum entre os agricultores brasileiros. Desta forma, este trabalho teve como objetivo avaliar a eficácia e seletividade de misturas, em tanque, de herbicidas com glyphosate para o controle de trapoeraba (Commelina benghalensis L.), erva-de-touro (Tridax procumbens L.) e capim-carrapicho (Cenchrus echinatus L.) na cultura da soja RR®. O experimento foi conduzido em Maracaí, São Paulo, no período de novembro de 2006 a março de 2007, utilizando-se o cultivar CD-214RR® e delineamento experimental de blocos ao acaso, com 21 tratamentos e quatro repetições. The photochemistry of merocyanine 540 (MC 540), a sensitizing dye that binds preferentially to leukemia and electrically excitable cells, has been investigated. MC 540-mediated photooxidation of histidine, arachidonate, and unsaturated phospholipid vesicles was assessed by spin label oximetry and shown to involve type II (singlet oxygen) chemistry. The dye was also shown to be a potent sensitizer of lipid peroxidation in a natural cell membrane, the erythrocyte ghost. Inhibition by azide, stimulation by 2H2O, and identification of the cholesterol product 5 alpha-cholest-6-ene-3 beta,5-diol in this system, all were consistent with singlet oxygen intermediacy. Finally, MC 540 was found to be considerably more phototoxic to K-562 leukemia cells in 2H2O than in H2O. We conclude that singlet oxygen plays a major role in the phototherapeutic effects of this dye.

In vitro incubation of leukemic bone marrow with the lipophilic fluorescent dye merocyanine 540 and simultaneous exposure to light reduced the concentration of L1210 leukemia cells by about 5 orders of magnitude but spared enough normal pluripotent hematopoietic stem cells to allow hematopoietic reconstitution of lethally irradiated syngeneic mice. This simple and rapid procedure may find an application in the purging of tumor cells from autologous bone marrow grafts.

Photodynamic inactivation of Staphylococcus aureus planktonic and biofilm cells by a phtotosensitizer, merocyanine 540 (MC 540), was investigated. For the planktonic experiments, MC 540 binding efficiency to bacterial cells was found to increase with both increasing MC 540 concentration and increasing incubation time, but the binding became saturated following 10 min of incubation. The antimicrobial activity was enhanced with an increasing light dose, but an increase in the light dose could not further improve the antimicrobial activity if the maximum excitation level attainable was less than the necessary minimum threshold level. Complete inactivation was achieved when the excitation level of MC 540 was somewhere above the threshold level. The relationship between antimicrobial activity and the excitation level of MC 540 revealed that the more MC 540 was excited, the more S. aureus cells were killed. For the biofilm experiments, the antimicrobial activity was enhanced with an increase in the light dose. No viable cells were detected when organisms were exposed to 15 μg of MC 540 per ml and a light dose of 600 J/cm2 or to 20 μg of MC 540 per ml and a light dose of 450 J/cm2. 1. The fluorescent dye Merocyanine-540 and the two weakly fluoresecnet dyes Merocyanine-rhodanine and Merocyanine-oxazolone are shown to respond as optical probes of membrane potential in heart muscle. 2. In frog hearts stained with Merocyanine-540, the absorption at 540 nm decreases by 0.1-1.0% and increase at 570 nm excitation wave-length, the fluorescence increases by 1-2%. The time course of all three optical measurements follows the kinetics of the action potential. 3. Merocyanine-rhodanine exhibits potential-dependent optical responses through a 0.5% decrease in absorption at 750 nm, and Merocyanine-oxazolone has a 1.0% decrease in absorption at 720 nm. Their optical responses have a signal-to-noise ratio of 100/1 and 500/1, respectively. 4. The action spectrum of Merocyanine-rhodanine is triphasic in frog heart with an increase in transmittance from 780 to 700, a decrease from 700 to 600, and increase from 600 to 450 nm. Merocyanine-oxazolone shows only increases in transmittance during membrane depolarization. 5. The optical responses of these probes are linear with respect to changes in membrane potential. 6. The effects of merocyanine 540 on the electrical properties of lipid bilayer membranes have been investigated. The alterations this dye was found to produce in the intrinsic conductances of these membranes were minimal, but it profoundly altered the conductances produced by extrinsic permeant species. These alterations were much larger for neutral membranes than for negatively charged ones. The dye increased the conductances mediated by positively charged permeant species and decreased those by negatively charged permeant species, suggesting that it produces a negative electrostatic potential on the membrane; it also altered the kinetics and the voltage dependencies of permeation by these charge carriers. The magnitudes of dye-mediated conductance changes were much larger for positively charged permeants than for negatively charged ones; also, changes in ionic strength altered these dye effects in opposite directions from those predicted by the Stern equation, and the dependence of the conductance alteration on dye concentration was steeper than that predicted by this equation. Finally, only very small changes in liposome zeta potentials were induced by the dye. Promoter hypermethylation, mutations, and loss of heterozygosity in the CDKN2A gene as well as polymorphisms at the 3′-untranslated region were determined in vertical growth phase melanomas. Methylation-specific polymerase chain reaction in soluti and in situ showed that 19% of the cases were hypermethylated at the CDKN2A promoter region, and some of these cases were heterogeneous with both methylated and unmethylated tumor cells. Methylation was associated with increased tumor cell proliferation by Ki-67 expression (P = 0.01) and decreased patient survival (P = 0.025). Point mutations in CDKN2A were found in 4% of the cases, whereas 90% had loss of heterozygosity at one or more of 4 markers studied. Furthermore, presence of the 540 C>T polymorphism at the 3′-untranslated region of CDKN2A (23%) was associated with improved survival in multivariate analysis (hazard ratio, 2.6; P = 0.02). Our results suggest that promoter methylation of the CDKN2A gene is present in a subgroup of the tumors and associated with increased tumor cell proliferation and reduced survival. Further, the 540 C>T polymorphism might define a distinct subgroup of low-grade vertical growth phase melanomas. These findings support a significant role of the CDKN2A gene in melanoma progression.

The phosphatidylinositide 3-kinase pathway is frequently deregulated in human cancers and inhibitors offer considerable therapeutic potential. We previously described the promising tricyclic pyridofuropyrimidine lead and chemical tool compound PI-103. We now report the properties of the pharmaceutically optimized bicyclic thienopyrimidine derivatives PI-540 and PI-620 and the resulting clinical development candidate GDC-0941. All four compounds inhibited phosphatidylinositide 3-kinase p110α with IC50 ≤ 10 nmol/L. Despite some differences in isoform selectivity, these agents exhibited similar in vitro antiproliferative properties to PI-103 in a panel of human cancer cell lines, with submicromolar potency in PTEN-negative U87MG human glioblastoma cells and comparable phosphatidylinositide 3-kinase pathway modulation. PI-540 and PI-620 exhibited improvements in solubility and metabolism with high tissue distribution in mice. Both compounds gave improved antitumor efficacy over PI-103, following i.p. dosing in U87MG glioblastoma tumor xenografts in athymic mice, with treated/control values of 34% (66% inhibition) and 27% (73% inhibition) for PI-540 (50 mg/kg b.i.d.) and PI-620 (25 mg/kg b.i.d.), respectively. Melioidosis is an occupationally and recreationally acquired infection important in Southeast Asia and northern Australia. Recently cases have been reported from more diverse locations globally. The responsible bacterium, Burkholderia pseudomallei, is considered a potential biothreat agent. Risk factors predisposing to melioidosis are well recognised, most notably diabetes. The Darwin prospective melioidosis study has identified 540 cases of melioidosis over 20 years and analysis of the epidemiology and clinical findings provides important new insights into this disease. Risk factors identified in addition to diabetes, hazardous alcohol use and chronic renal disease include chronic lung disease, malignancies, rheumatic heart disease, cardiac failure and age ≥50 years. Half of patients presented with pneumonia and septic shock was common (21%). The decrease in mortality from 30% in the first 5 years of the study to 9% in the last five years is attributed to earlier diagnosis and improvements in intensive care management. Of the 77 fatal cases (14%), all had known risk factors for melioidosis. This supports the most important conclusion of the study, which is that melioidosis is very unlikely to kill a healthy person, provided the infection is diagnosed early and resources are available to provide appropriate antibiotics and critical care where required.

The lipophilic dye merocyanine 540 (MC540) was used to model small molecule-membrane interactions using micropatterned lipid bilayer arrays (MLBAs) prepared using a 3D Continuous Flow Microspotter (CFM). Fluorescence microscopy was used to monitor MC540 binding to fifteen different bilayer compositions simultaneously. MC540 fluorescence was two times greater for bilayers composed of liquid-crystalline (l.c.) phase lipids (1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC), 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), 1-stearoyl-2-oleoyl-sn-glycero-3-phosphocholine (SOPC), and 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC)) compared to bilayers in the gel phase (1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC)). The effect cholesterol (CHO) had on MC540 binding to the membrane was found to be dependent on the lipid component; cholesterol decreased MC540 bindingin DMPC, DPPC and DSPC bilayers while having little to no effect on the remaining l.c. phase lipids. MC540 fluorescence was also lowered when 1,2-dioleoyl-sn-glycero-3-phospho-L-serine (sodium salt) (DOPS) was incorporated into DOPC bilayers. The increase in the surface charge density appears to decrease the occurrence of highly fluorescent monomers and increase the formation of weakly fluorescent dimers via electrostatic repulsion. This paper demonstrates that MLBAs are a useful tool for preparing high density reproducible bilayer arrays to study small molecule-membrane interactions in a high-throughput manner.

Merocyanine 540 (MC540) is used as a photosensitizer for the inactivation of microorganisms. The following is already known about MC540: firstly, MC540 exists in distilled water in both monomeric and dimeric forms, and the addition of salts into a MC540 solution leads to the formation of large aggregates that can be detected by the resonance light scattering technique. Secondly, singlet oxygen can only be photogenerated by MC540 monomers. In the present work, we studied the effect of MC540 in the aggregated state on the rate of photosensitized inactivation ofStaphylococcus aureusandPseudomonas aeruginosa. To this end, bacteria either in MC540-containing distilled water or in a 0.25 M sodium chloride aqueous solution also containing MC540 are irradiated (546 nm). The results show that, in the presence of salt, the aggregation of MC540 greatly increases the efficiency of the MC540-photosensitized inactivation ofP. aeruginosaandS. aureus. In the presence of salt, the rates ofP. aeruginosaandS. aureusinactivation increase by factors of 10 and 30, respectively, in comparison with the rate of inactivation observed in the case of distilled water. Our results suggest that a salt-induced photosensitization mechanism can switch from the singlet oxygen to the free-radical pathway.

The frequency of gall bladder lesions in cholecystectomies is not clear. The purpose of the present study is to report the morphologies and frequency of gall bladder diseases and lesions of 540 cholecystectomies in the last 10 years in our pathology laboratory. The age of patients ranged from 18 years to 93 years with a mean of 64.75 ±14.43 years. Male to female ratio was 213:327. Of these, 518 cases (96%) had gall stones. Eight (1.5%) were acute cholecystitis, 508 (94.1%) were chronic cholecystitis, 12 (2.2%) were adenocarcinomas, 1 (0.2%) was cystadenocarcinoma, and 11 (2.0%) were normal gall bladders. The frequency of histological lesions were as follows: acute gangrenous inflammation (8 cases, 1.5%), Rokitansky-Aschoff sinuses (RAS) (351 cases, 65%), microliths or inspissated bile in RAS (108 cases, 20%), adenomyomatous changes (16 cases, 3.0 %), focal abscess formations (12 cases, 2.2%), focal xanthogranulomatous changes (15 cases, 2.8%), mucosal ulcers (61 cases, 11.3%), cholesterosis (62 cases, 11%), cholesterol polyp (32 cases, 6%), pyloric gland metaplasia (292 cases, 54%), adenoma (7 cases, 1.3%), xanthogranulomatous cholecystitis (5 cases, 1%), invasive adenocarcinoma (12 cases, 2.2%), and cystadenocarcinoma (1 cases, 0.2%). Sphingolipids function as cell membrane components and as signaling molecules that regulate critical cellular processes. To study unacylated and acylated sphingolipids in cells with fluorescence microscopy, the fluorophore in the analog must be located within the sphingoid backbone and not the N-acyl fatty acid side chain. Although such fluorescent sphingosine analogs have been reported, they either require UV excitation or their emission overlaps with that of the most common protein label, green fluorescent protein (GFP). We report the synthesis and use of a new fluorescent sphingolipid analog, borondipyrromethene (BODIPY) 540 sphingosine, which has an excitation maximum at 540 nm and emission that permits its visualization in parallel with GFP. Mammalian cells readily metabolized BODIPY 540 sphingosine to more complex fluorescent sphingolipids, and subsequently degraded these fluorescent sphingolipids via the native sphingolipid catabolism pathway. Visualization of BODIPY 540 fluorescence in parallel with GFP-labeled organelle-specific proteins showed the BODIPY 540 sphingosine metabolites were transported through the secretory pathway and were transiently located within lysosomes, mitochondria, and the nucleus. Potentiation of the Anti-Tumor Effect of Merocyanine 540-Mediated Photodynamic Therapy by Amifostine and Amphotericin B

Leukemia and lymphoma cells are much more sensitive to Merocyanine 540 (MC540)-mediated photodynamic therapy (PDT) than normal pluripotent hematopoietic stem cells and normal granulocyte/macrophage progenitors (CFU-GM). By contrast, most solid tumor cells are only moderately sensitive to MC540-PDT. The limited activity against solid tumor cells has detracted from MC540's appeal as a broad-spectrum purging agent. We report here that non-cytotoxic concentrations of amifostine (Ethyol, Ethiofos, WR-2721) and amphotericin B used either alone or in combination potentiate the MC540-sensitized photoinactivation of leukemia cells, wildtype small cell lung cancer cells, and cisplatin-resistant small cell lung cancer cells. Amphotericin B also enhances the MC540-sensitized photoinactivation of normal CFU-GM whereas amifostine protects CFU-GM against the cytotoxic action of MC540-PDT. The yield of CD34-positive normal hematopoietic stem and progenitor cells is only minimally diminished by pretreatment with amifostine, amphotericin B, or combinations of amifostine plus amphotericin B. Purging protocols that combine MC540-PDT with amifostine or with amifostine plus amphotericin B may offer a simple and effective approach to the purging of autologous stem cell grafts that are contaminated with solid tumor cells or the purging of stem cell grafts from heavily pretreated leukemia patients that contain reduced numbers of normal stem and progenitor cells and...

Combined use of steady-state fluorescence emission and anisotropy of merocyanine 540 to distinguish crystalline, gel, ripple, and liquid crystalline phases in dipalmitoylphosphatidylcholine bilayers

The various lamellar phases of dipalmitoylphosphadtidylcholine bilayers with and without cholesterol were used to assess the versatility of the fluorescent probe merocyanine 540 through simultaneous measurements of emission intensity, spectral shape, and steady-state anisotropy. Induction of the crystalline phase (Lc') by pre-incubation at 4°C produced a wavelength dependence of anisotropy which was strong at 15 and 25°C, weak at 38°C, and minimal above the main transition (>~41.5°C) or after returning the temperature from 46 to 25°C. The profile of anisotropy values across this temperature range revealed the ability of the probe to detect crystalline, gel (Lβ'), and liquid crystalline (Lα) phases. The temperature dependence of fluorescence intensity was additionally able to distinguish between the ripple (Pβ') and gel phases. In contrast, the shape of the emission spectrum, quantified as the ratio of merocyanine monomer and dimer peaks (585 and 621 nm), was primarily sensitive to the crystalline and gel phases because dimer fluorescence requires a highly-ordered environment. This requirement also explained the diminution of anisotropy wavelength dependence above 25°C. Repetition of experiments with vesicles containing cholesterol allowed creation of a phase map. Superimposition of data from the three simultaneous measurements provided details about the various phase regions in the map not discernible from any one of the three alone. The results were applied to assessment of calcium-induced membrane changes in living cells.

Tip of the Red Giant Branch Distance for the Sculptor Group Dwarf ESO 540-032

We present the first VI CCD photometry for the Sculptor group galaxy ESO 540-032 obtained at the Very Large Telescope UT1+FORS1. The (I, V-I) colour-magnitude diagram indicates that this intermediate-type dwarf galaxy is dominated by old, metal-poor ([Fe/H]~-1.7 dex) stars, with a small population of slightly more metal-rich ([Fe/H]~-1.3 dex), young (age 150-500 Myr) stars. A discontinuity in the I-band luminosity function is detected at I_0 = 23.44 \pm 0.09 mag. Interpreting this feature as the tip of the red giant branch and adopting M_I=-4.20\pm 0.10 mag for its absolute magnitude, we have determined a Population II distance modulus of (m - M)_0=27.64 \pm 0.14 mag (3.4 \pm 0.2 Mpc). This distance confirms ESO 540-032 as a member of the nearby Sculptor group but is significantly larger than a previously reported value based on the Surface Brightness Fluctuation (SBF) method. The results from stellar population synthesis models suggest that the application of the SBF technique on dwarf galaxies with mixed morphology requires a detailed knowledge of the underlying stellar composition and thus offers no advantage over a direct distance measurement using the tip of the red giant branch as distance indicator. We produce the surface brightness profiles for ESO 540-032 and derive the photometric and structural parameters.
