Os estudos sobre a conversão in vitro de meristemas apicais radiculares em gemas caulinares de plantas do gênero Catasetum vêm contribuindo para uma melhor compreensão dos processos de competência, indução e determinação celular no processo de desenvolvimento, necessitando, no momento, de aprofundamento em estudos moleculares. Para tanto, a utilização de plantas transgênicas representa uma ferramenta de trabalho importante. Além disso, os métodos de transformação genética acenam como uma alternativa eficaz para o melhoramento de plantas de interesse econômico com ciclos reprodutivos longos, como as orquídeas. Desta forma, o objetivo deste projeto foi estabelecer um protocolo para transformação genética de Catasetum pileatum, utilizando tecidos meristemáticos como explantes alvos. O carcinoma adenóide cístico pode raramente sofrer desdiferenciação, um fenômeno também referido como transformação para alto grau. Contudo, alguns casos de carcinoma adenóide cístico foram descritos mostrando transformação para adenocarcinomas que não são pobremente diferenciados, indicando que a transformação para alto grau pode não refletir necessariamente um estágio mais avançado da progressão tumoral, mas sim uma transformação em uma outra forma histológica, que pode abranger um amplo espectro de carcinomas em termos de agressividade. Inicialmente, investigamos a expressão das proteínas reguladas pela hipóxia (HIF-1α, VEGF, GLUT-1 e CD105), dado que a hipóxia contribui para a agressividade tumoral e, pode, também, promover um fenótipo desdiferenciado em certos tipos de câncer. Abelson murine leukemia virus (Ab-MLV) encodes the v-Abl protein tyrosine kinase and induces transformation of immortalized fibroblast lines and pre-B cells. Temperature-sensitive mutations affecting the kinase domain of the protein have demonstrated that the kinase activity is absolutely required for transformation. Despite this requirement, mutations affecting other regions of v-Abl modulate transformation activity. The SH2 domain and the highly conserved FLVRES motif within it form a phosphotyrosine-binding pocket that is required for interactions between the kinase and cellular substrates. To understand the impact of SH2 alterations on Ab-MLV-mediated transformation, we studied the Ab-MLV mutant P120/R273K. This mutant encodes a v-Abl protein in which the βB5 arginine at the base of the phosphotyrosine-binding pocket has been replaced by a lysine. Unexpectedly, infection of NIH 3T3 or pre-B cells with P120/R273K revealed a temperature-dependent transformation phenotype. At 34°C, P120/R273K transformed about 10-fold fewer cells than wild-type virus of equivalent titer; at 39.5°C, 300-fold fewer NIH 3T3 cells were transformed and pre-B cells were refractory to transformation. Jaagsiekte sheep retrovirus (JSRV) is the causative agent of ovine pulmonary adenocarcinoma, a transmissible lung cancer of sheep. The envelope of JSRV may have oncogenic properties, since it can morphologically transform mouse NIH 3T3 cells and other fibroblast lines. Recently, we found that the cytoplasmic tail of the envelope transmembrane (TM) protein is necessary for transformation, and in particular a consensus binding motif (YXXM) for phosphatidylinositol 3-kinase (PI3K) is important. Moreover, JSRV-transformed cells show phosphorylation (activation) of Akt/protein kinase B, a downstream target of PI3K. In these studies, we directly tested for the involvement of PI3K in transformation by JSRV. Contrary to expectations, four different experiments indicated that PI3K is not necessary for JSRV-induced transformation: (i) cotransfection with a dominant negative truncated form of the PI3K regulatory subunit (Δp85) did not affect transformation frequency, (ii) cells stably expressing Δp85 showed the same frequencies of transformation as parental NIH 3T3 cells, (iii) fibroblasts established from double-knockout mice lacking PI3K p85α and p85β could be transformed with JSRV envelope, and (iv) incubation of cells with the PI3K inhibitor LY294002 did not specifically inhibit transformation...

Jaagsiekte sheep retrovirus (JSRV) is an exogenous retrovirus of sheep that induces a contagious lung cancer, ovine pulmonary adenocarcinoma. We previously showed that the gene encoding JSRV envelope protein (Env) appears to function as an oncogene, since it can transform mouse NIH 3T3 cells. The cytoplasmic tail of the Env transmembrane protein (TM) is necessary for the transformation. However, previous experiments did not exclude the involvement of the Env surface protein (SU) in transformation. In this study, we created a series of nested deletion mutants through the SU domain and assessed their ability to transform rodent fibroblasts. All SU deletion mutants downstream of the predicted signal peptide were unable to transform murine NIH 3T3 or rat 208F cells. Transport to the plasma membrane of selected deleted Env proteins was confirmed by confocal immunofluorescence microscopy of hemagglutinin-tagged versions. Additional sequential SU deletion mutants lacking 50-amino-acid (aa) blocks throughout SU also were unable to transform. Furthermore, minimal insertion mutants of two amino acids (Leu/Gln) at various positions in SU also abolished transformation. These data indicate that domains in SU facilitate efficient JSRV transformation. This could reflect a necessity of SU for appropriate configuration of the Env protein or independent activation by SU of a signaling pathway necessary for transformation. Complementation between SU and TM mutants for transformation supported the latter hypothesis. Cotransfection with ΔGP Y590F (mutant in the TM cytoplasmic tail) with ΔGP SUΔ103-352 (lacking most of SU) resulted in efficient transformation. The resulting transformants showed evidence for the presence and expression of both mutant plasmids.

Jaagsiekte sheep retrovirus (JSRV) is the causative agent of ovine pulmonary adenocarcinoma (OPA), a transmissible lung cancer of sheep. The virus can induce tumors rapidly, and we previously found that the JSRV envelope protein (Env) functions as an oncogene, because it can transform mammalian and avian fibroblast cell lines. (N. Maeda, Proc. Natl. Acad. Sci. USA 98:4449-4454, 2001). The molecular mechanisms of JSRV Env transformation are of considerable interest. Several reports suggested that the phosphatidylinositol 3-kinase/Akt pathway is important for transformation of mammalian fibroblasts but not for chicken fibroblasts. In this study, we found that Akt/mTOR is involved in JSRV transformation of mouse NIH 3T3 fibroblasts, because treatment with the mTOR inhibitor rapamycin reduced transformation. We also found that H/N-Ras inhibitor FTI-277 and MEK1/2 inhibitors PD98059 and U0126 strongly inhibited JSRV transformation of NIH 3T3 fibroblasts, suggesting that the H/N-Ras-MEK-mitogen-activated protein kinase (MAPK) p44/42 pathway is necessary for the transformation. In RK3E epithelial cells, the MEK1/2 inhibitors also eliminated transformation, but FTI-277 only partially inhibited transformation. It was noteworthy that p38 MAPK inhibitors enhanced JSRV transformation in both fibroblasts and epithelial cells. Treatment of transformed cells with p38 inhibitors both increased levels of phospho-MEK1/2 and phospho-p44/42 and induced rapid enhancement of the transformed phenotype. Human adenovirus E1A makes extensive connections with the cellular protein interaction network. By doing so, E1A can manipulate many cellular programs, including cell cycle progression. Through these reprogramming events, E1A functions as a growth-promoting oncogene and has been used extensively to investigate mechanisms contributing to oncogenesis. Nevertheless, it remains unclear how the C-terminal region of E1A contributes to oncogenic transformation. Although this region is required for transformation in cooperation with E1B, it paradoxically suppresses transformation in cooperation with activated Ras. Previous analysis has suggested that the interaction of E1A with CtBP plays a pivotal role in both activities. However, some C-terminal mutants of E1A retain CtBP binding and yet exhibit defects in transformation, suggesting that other targets of this region are also necessary. To explore the roles of these additional factors, we performed an extensive mutational analysis of the C terminus of E1A. We identified key residues that are specifically required for binding all known targets of the C terminus of E1A. Agrobacterium-mediated sorghum transformation frequency has been enhanced significantly via medium optimization using immature embryos from sorghum variety TX430 as the target tissue. The new transformation protocol includes the addition of elevated copper sulfate and 6-benzylaminopurine in the resting and selection media. Using Agrobacterium strain LBA4404, the transformation frequency reached over 10% using either of two different selection marker genes, moPAT or PMI, and any of three different vectors in large-scale transformation experiments. With Agrobacterium strain AGL1, the transformation frequencies were as high as 33%. Using quantitative PCR analyses of 1,182 T0 transgenic plants representing 675 independent transgenic events, data was collected for T-DNA copy number, intact or truncated T-DNA integration, and vector backbone integration into the sorghum genome. A comparison of the transformation frequencies and molecular data characterizing T-DNA integration patterns in the transgenic plants derived from LBA4404 versus AGL1 transformation revealed that twice as many transgenic high-quality events were generated when AGL1 was used compared to LBA4404. This is the first report providing molecular data for T-DNA integration patterns in a large number of independent transgenic plants in sorghum.

Einleitung: Es konnte gezeigt werden, dass der aus Sojabohnen gewonnene Bowman-Birk Protease Inhibitor Zellen in vivo und in vitro vor chemikalien- und strahleninduzierter Karzinogenese schützt. In den folgenden Experimenten wurde die Rolle von BBI und P-Tyrosin im Prozess der neoplastischen Zelltransformation untersucht. Methoden: Hierfür wurde das in vitro Zellsystem C3H 10T1/2 (Mausfibroblasten) verwendet. Die Transformationsassays wurden nach der von Reznikoff et al. etablierten Methode durchgeführt. Ergebnisse: Neoplastische C3H 10T1/2 Zellen konnten anhand ihres Phänotyps erkannt werden, denn diese formten 3D-Cluster, so genannte Foci. Je nach Behandlung wurden quantitative Unterschiede bezüglich der Transformationsereignisse gefunden: Generell waren C3H 10 T1/2 Zellen sehr sensitiv gegenüber neoplastischer Zelltransformation. Es wurde beobachtet, dass die spontane Transformationsrate von Kontrollzellen in hohen Passagen (mit Beginn von Passage 20) anstieg. Behandlung mit chemischen oder physikalischen Agenzien erhöhte die Transformationsfrequenz: Verglichen mit der Foci-Anzahl, die aus unbehandelten Kontrollzellen entstanden waren, zeigte sich eine vermehrte Anzahl an Foci in bestrahlten oder mit Methylcholanthren behandelten Zellen. Structural transformation refers to the reallocation of economic activity across the broad sectors agriculture, manufacturing, and services. This review article synthesizes and evaluates recent advances in the research on structural transformation. We begin by presenting the stylized facts of structural transformation across time and space. We then develop a multi-sector extension of the one-sector growth model that encompasses the main existing theories of structural transformation. We argue that this multi-sector model serves as a natural benchmark to study structural transformation and that it is able to account for many salient features of structural transformation. We also argue that this multi-sector model delivers new and sharper insights for understanding economic development, regional income convergence, aggregate productivity trends, hours worked, business cycles, wage inequality, and greenhouse gas emissions. Approved for public release; distribution in unlimited.; The thesis studies the Israel Defense Forces' (IDF) transformation in the context of the U.S. military transformation. The thesis argues that the uniqueness of the U.S. military transformation does not prevent other militaries from applying the relevant concepts and considers the IDF as a good candidate for such a demonstration. Therefore, the thesis explores the U.S. military transformation as a model to be benefited in the IDF's continuing transformation. The thesis also studies IDF from many perspectives to identify the transformational imperatives and the relevance of the current IDF transformational efforts. Main areas of interest are the Israeli security environment, societal transformation, and the features of the IDF's transformational efforts. During the analysis, the thesis discovers weak points concerning, both, the U.S. military transformation and the IDF's current organization. Source transformation systems are special-purpose programming languages, or in some cases suites of languages, that are designed for the analysis and transformation of computer languages. They enable rapid prototyping of programming languages, source code renovation, language-to-language translation, design recovery, and other custom analysis techniques. With the emergence of these systems a serious problem is evident: expressing a parser for common computer languages is sometimes very difficult. Source transformation systems employ generalized parsing algorithms, and while these are well suited for the kind of agile parsing techniques in use by transformation practitioners, they are not well suited for parsing languages that are context-dependent. Traditional deterministic parser generators do not stumble in this area, but they sacrifice the generalized parsing abilities that transformation systems depend on. When it is hard to get the input into the system as a correct and accurate parse tree the utility of the unified transformation environment is degraded and more ad hoc approaches become attractive for processing input. This thesis is about the design of a new computer language transformation system with a focus on enhancing the parsing system to support generalized context-dependent parsing. We argue for the use of backtracking LR as the generalized parsing algorithm. We present an enhancement to backtracking LR that allows us to control the parsing of an ambiguous grammar by ordering the productions of the grammar definitions. Transformation Induced Plasticity (TRIP) steels have both high strength and high ductility. Retained austenite in the microstructure, upon straining, transforms to martensite and this absorbs energy and improves the work hardening of the steel, giving improved elongation. The transformation can be either stress-assisted or strain-induced and the initiation and the mechanism depend on the composition of, the size and shape of, and the phases surrounding, the austenite grains. It is important to understand the relationship between these variables and the properties of the TRIP steel. The aim of this work was to determine how the microstructure of the TRIP steel affects the transformation. Four experimental microstructures were developed, containing austenite grains with different sizes, shapes, and surrounding phases. Dans le contexte de l’expression moderne de la religiosité, l’étude explore comment les méthodes de l’Ashtanga yoga produisent l’expérience de la transformation chez les individus qui les pratiquent et qui atteint, dans une certaine mesure, leur entourage. Ethnographiquement, le mémoire se concentre sur le rituel principal de l’Ashtanga Vinyasa Yoga (AVY), les séances de style Mysore, telle qu’exercé par la communauté de pratiquants de la Sattva Yoga Shala à Montréal. En tant que performance, le rituel met en branle le potentiel de transformation. La pratique de ce yoga implique une discipline et un apprentissage draconien sur une longue échelle temporelle qui amènent, au fur et à mesure, des changements se répercutant dans/et influençant le vécu quotidien des adeptes. La transformation de modèles consiste à transformer un modèle source en un modèle cible conformément à des méta-modèles source et cible. Nous distinguons deux types de transformations. La première est exogène où les méta-modèles source et cible représentent des formalismes différents et où tous les éléments du modèle source sont transformés. Quand elle concerne un même formalisme, la transformation est endogène. Ce type de transformation nécessite généralement deux étapes : l’identification des éléments du modèle source à transformer, puis la transformation de ces éléments. Dans le cadre de cette thèse, nous proposons trois principales contributions liées à ces problèmes de transformation. La première contribution est l’automatisation des transformations des modèles. Nous proposons de considérer le problème de transformation comme un problème d'optimisation combinatoire où un modèle cible peut être automatiquement généré à partir d'un nombre réduit d'exemples de transformations. Cette première contribution peut être appliquée aux transformations exogènes ou endogènes (après la détection des éléments à transformer). La deuxième contribution est liée à la transformation endogène où les éléments à transformer du modèle source doivent être détectés. Nous proposons une approche pour la détection des défauts de conception comme étape préalable au refactoring. O trabalho teve como objetivo otimizar a metodologia de transformação de plantas de Eucalyptus grandis; uma importante espécie florestal amplamente cultivada no Brasil. Foram estudadas a transformação por agrobiobalística de explantes com acúmulo de gemas axilares e do SAAT ('sonication-assisted Agrobacterium mediated transformation'). Ambas as técnicas causam microferimentos nos explantes aumentando a penetração da Agrobacterium nos tecidos vegetais. Na agrobiobalística o explante é submetido ao bombardeamento com micropartículas seguida por inoculação com a bactéria; enquanto que no SAAT o explante é submetido à sonicação por breves períodos de tempo na presença da bactéria. Os experimentos de transformação foram avaliados pela analise da expressão transitória do gene repórter uidA. Os parâmetros estudados na transformação por agrobiobalística foram: pressão de disparo ( pressão do gás hélio), distância de vôo das micropartículas, estabilidade da expressão do gene uidA e temperatura de co-cultivo. As freqüências mais elevadas da expressão da ß-glucuronidase foram observadas utilizando 1350 PSI de gás hélio, 9,5 cm de distância de vôo dos microprojéteis e temperatura de 26ºC durante o co-cultivo. A transformação genética está se tornando uma importante ferramenta, dentro dos programas de melhoramento genético de citros, e uma alternativa para contornar barreiras naturais da espécie, que dificultam o desenvolvimento de novas variedades pelo melhoramento convencional. Entretanto, os protocolos utilizados para transformação genética de citros têm resultado num baixo número de plantas transgênicas. Com o objetivo de estudar fatores que possam influenciar o processo de transformação genética de citros via Agrobacterium tumefaciens analisou-se o efeito do uso de acetoseringona em diferentes etapas do processo, as condições de incubação dos explantes durante e após o período de co-cultivo e o tipo de corte do explante. Foram utilizados segmentos de epicótilo de plântulas germinadas in vitro da variedade de laranja doce ‘Hamlin’ (Citrus sinensis L. Osbeck.) e da variedade citrange ‘Carrizo’ (C. sinensis x Poncirus trifoliata Raf.), inoculados com a estirpe EHA 105 de A. tumefaciens, contendo o plasmídeo p35SGUSINT, com o gene de seleção que codifica a enzima neomicina fosfotransferase II (nptII) e o gene repórter uidA que codifica a enzima b-glucuronidase (gus). El girasol es una especie de la familia Asteraceae (Compositae) de gran importancia económica, considerada hasta hace una década recalcitrante al cultivo in vitro y la transformación genética. En un trabajo previo (Radonic, 2005) se logró establecer el protocolo de selección por enraizamiento en kanamicina con una eficiencia del 0,7 %. En esta tesis se mejoró este protocolo utilizando una construcción portadora de los genes antifúngicos glucanasa y quitinasa, ambos bajo el control del promotor CaMV35S y el enhancer Ω del TMV, obteniéndose una eficiencia de 1,26 %, al aumentar gradualmente la concentración del antibiótico kanamicina en los sucesivos medios de regeneración. También se evaluó la especificidad y estabilidad de dos vectores conteniendo la construcción CaMV35S-secuencia codificante para la β-glucuronidasa (GUS)-interrumpida por un intrón. Las plantas T1 derivadas de los eventos de transformación presentaron un patrón de expresión de tipo no constitutivo, con expresión de GUS localizada exclusivamente en los tricomas de las nervaduras de la cara abaxial de las hojas, siendo necesario la utilización de lupa para su visualización. En las plantas T2 no se pudo detectar la presencia de los transgenes. Raised to Newness of Life: Resurrection and Moral Transformation in Second- and Third-Century Christian Theology

The New Testament contains two important and potentially conflicting understandings of resurrection. One integrates resurrection into salvation, suggesting that it is restricted to the righteous; this view is found most prominently in the Pauline epistles. The other understands resurrection as a prerequisite for eschatological judgment and therefore explicitly extends it to all; this view is found most prominently in the book of Revelation. In the former, moral transformation is part of the process that results in resurrection; in the latter, moral transformation only affects what comes after resurrection, not the event of resurrection itself. The New Testament itself provides no account of how to hold together these understandings of resurrection and moral transformation.

This dissertation is an investigation of the ways in which second- and third-century Christian authors creatively struggled to bring together these two understandings. I select key authors who are not only important in the history of early Christian discussions of resurrection but who also make extensive use of the Pauline epistles. For each author, I investigate not only how they develop or resist the Pauline connection between resurrection and moral transformation but also how they relate that connection to the doctrine of the resurrection of all to face judgment found in Revelation (if they do at all).

The results are remarkably diverse. Irenaeus develops the Pauline connection between resurrection and moral transformation through the Spirit of God but fails to account for the resurrection of those who do not receive that Spirit in this life (although affirming that resurrection nonetheless). Tertullian begins from the model that takes resurrection to be fundamentally a prerequisite for judgment and struggles to account for Paul's connections between resurrection and salvation.
