A case of extramedullary plasmacytoma of the third eyelid gland in a 7-year-old American Cocker Spaniel is reported. An enlargement of the third eyelid gland, abundant mucopurulent discharge, mild hyperemia and corneal pigmentation in the OD was present. Excisional biopsy of the mass revealed the gland was infiltrated and partially destroyed by a uniform population of neoplastic plasma cells. The neoplastic cells were positive for CD138, Ki-67 and lambda light chain. CD20, CD3, kappa light chain and cytokeratin were negative. Twelve months following surgery, no recurrence was observed. To the authors` knowledge, this is the first extramedullary plasmacytoma of the third eyelid gland reported in dogs.

O prolapso da glândula da terceira pálpebra (“olho de cereja”) é o distúrbio primário mais comum deste anexo oftálmico. Caracteriza-se pelo edema e inflamação da glândula, resultando em exposição desta no canto naso medial do olho. Esta condição afeta cães e gatos e sua etiologia envolve a fraqueza das bandas de tecido conectivo que unem a membrana nictitante aos tecidos periorbitais. Devido significativa colaboração da glândula à produção lacrimal, esta não deve ser excisada, o que pode levar ao desenvolvimento de ceratoconjuntivite seca. O tratamento de escolha é a cirurgia, na qual a glândula é reduzida, voltando à sua posição anatômica. Scrapie é uma encefalopatia espongiforme transmissível (EET) que causa lesões cerebrais degenerativas em ovinos e caprinos. Caracteriza-se pelo acúmulo, no tecido encefálico e linforreticular, da forma anormal da proteína priônica (PrPSc) que provoca a morte maciça de neurônios e células gliais, além de vacuolização intensa no tecido afetado. Esse trabalho descreve a utilização da técnica de imuno-histoquímica (IHQ) para proteína priônica em tecido linforreticular de biópsias de terceira pálpebra e mucosa retal, como método diagnóstico de scrapie em ovinos. Realizaram-se exames de IHQ para scrapie em amostras de uma propriedade de origem de um ovino com diagnóstico dessa enfermidade. Utilizaram-se anticorpos monoclonais antipríon para diagnóstico ante mortem pela técnica de IHQ. Nas 318 amostras de biópsias analisadas, encontrou-se 19 resultados positivos para PrPSc nos folículos de terceira pálpebra e não foi obtida marcação no tecido linfático de mucosa retal em nenhuma das amostras coletadas. Realizaram-se 18 necropsias dos animais positivos anteriormente por biópsia e 21 necropsias de ovinos parentes dos positivos de scrapie. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES); As glândulas lacrimal e superficial da terceira pálpebra atuam produzindo o componente aquoso do filme lacrimal. Nesta pesquisa, estudaram-se aspectos morfológicos e morfométricos do parênquima e do estroma de ambas as glândulas em cães mestiços, hígidos, adultos (machos ou fêmeas). As glândulas lacrimal e superficial da terceira pálpebra de 14 cães foram colhidas, totalizando 56 amostras. Foram estudadas, à estatística, as variáveis macroscópicas e morfométricas, comparando-as quanto ao tipo glandular (lacrimal e superficial da terceira pálpebra) e quanto ao dimorfismo sexual (macho e fêmea). Às glândulas lacrimais foram significativamente maiores comparativamente as superficiais da terceira pálpebra. Foram evidenciados diferenças morfométricas expressivas quanto aos vasos e ductos interlobulares, ducto interlobular, infiltração linfocitária e parênquima secretório entre os dois tipos glandulares. As glândulas lacrimais dos machos foram significativamente maiores comparativamente às fêmeas, assim como as glândulas superficiais da terceira pálpebra apresentaram-se mais espessas. The present experiment was carried out in ten animals with third eyelid prolapsed gland. All of the prolapsed glands were surgically excised. The age of the dogs ranged from two month to ten years old, and the breeds were Beagle (20%), Teckel (10%), Pinscher (10%), Poodle (10%) and other mongrel dogs. Aspirative citology was made in all the removed glands, after that they were fixed in 10% formalin and stained with Hematoxiline and Eosine. It was observed a focal mononuclear inflamatory infiltrate in the conjunctive and periacinal tissues. Ductal dilatation and metaplasia were noted on cells of the ducts. Cyitologic examination showed similar inflamatory infiltrate to histopatologic findings. The glands showed a chronic adenitis. The absence of histo and cytologic significance changes justify the tissue function observed during surgical excision.

Purpose: To detect the occurrence and expression of the suppressor gene p53 and of the oncogene c-Myc in eyelid tumors of dogs using the PCR, RT-PCR, PCR-ELISA and RT-PCR-ELISA techniques. These genes have not been described in dog eyelid tumors before. Methods: Nine samples of eyelid or third eyelid epithelial tumors were obtained from the archives of the Department of Veterinary Pathology. Tumor diagnosis was confirmed by evaluation of hematoxylin-eosin stained sections, and immunohistochemistry for cytokeratin AE1/AE3 and vimentin V9. A canine mammary tumor was used for positive control. Agarose gel electrophoresis, PCR-ELISA and RT-PCR-ELISA were used to detect p53 and c-Myc genes. Results: The occurrence of p53 was detected in most of the eyelid tumors and third eyelid tumors studied (88.8%, n = 8) and was expressed in 75% of the positive samples, as indicated by ELISA. The c-Myc gene was found in 77.7% (n = 7) of the samples and was expressed in eight samples. Conclusions: Eyelid and third eyelid tumors of dogs express both the p53 and the c-Myc genes as shown by PCR and RT-PCR. However, PCR ELISA and RT-PCR ELISA were more efficient in assessing occurrence and expression of these genes because they identified amplified products that were not detected by agarose gel electrophoresis. © 2010 American College of Veterinary Ophthalmologists.

Pós-graduação em Cirurgia Veterinária - FCAV; Com o trabalho, propõe-se apresentar um modelo experimental de prolapso da glândula da terceira pálpebra em cães, valendo-se, para tal, da ressecção do retináculo glandular periorbitário e de estudos macro e microscópicos, bem como das intercorrências clínicas, quanto à produção lacrimal. As variáveis foram estudadas em bulbos dos olhos com glândulas prolapsadas, em outros com glândulas sepultadas cirurgicamente e em bulbo do olhos que passaram por exérese glandular. Mediante análise dos resultados constatou-se que a produção lacrimal diminuiu, comparativamente às glândulas normais não prolapsadas, notadamente naqueles casos em que as glândulas prolapsadas não foram sepultadas. Glândulas prolapsadas apresentaram-se maiores, quanto ao comprimento, à largura e à espessura. Microscopicamente, glândulas prolapsadas não sepultadas cirurgicamente, exibiram alterações inflamatórias glandulares e ductais em maior intensidade. The lacrimal and third eyelid superficial glands produce the aqueous component of the preocular tear film. In this research, morphologic and morphometric assessments of the parenchyma and stroma of both lacrimal glands of healthy adult mongrel dogs were performed. Both lacrimal and third eyelid glands of fourteen dogs were collected, summing fifty-six samples. The macroscopic and morphometric data were statistically analyzed, according to the glandular type (lacrimal and third eyelid superficial glands) and sexual dimorphism (male or female). The lacrimal glands were significantly larger and longer than the superficial glands of the third eyelid. Expressive morphometric differences of interlobular duct, lymphocytic infiltration, interlobular vessels and secretory parenchyma between the two glandular types were encountered. The lacrimal glands from the male subjects were significantly larger than those from female ones, as well as the superficial glands of the third eyelid were thicker. The higher lymphocyte infiltration and poorer secretor parenchyma in female dogs may be one of the reasons for the higher incidence of keratoconjunctiviti sicca (KCS) in such canine population.

A 3-year-old Rhodesian Ridgeback was presented with conjunctivitis, enlargement of the third eyelid and a dorsotemporal deviation of the right eye. A mass within the third eyelid was detected and excised. The histopathologic examination showed a malignant peripheral nerve sheath tumor, which most likely is a neurofibrosarcoma based on immunohistochemistry.

An experimental study was made of 20 dogs in order to compare various surgical techniques used to correct eversion of the third eyelid, namely resection of most of the cartilage, resection of the central portion of the cartilage, and cartilage homotransplantation. An analysis was made of histological results obtained 45 days after operation, the most satisfactory result being recorded for homotransplantation of the third eyelid cartilage.

Ovine scrapie is a member of the transmissible spongiform encephalopathies (TSEs), a heterogeneous family of fatal neurologic disorders characterized by deposition of an abnormal isoform (prion protein [PrP] PrP-Sc) of a cellular sialoglycoprotein in neural tissue. PrP-Sc is detectable in some lymphoid tissues of infected sheep months or years before development of clinical disease. Detection of PrP-Sc in these tissues is the basis for live-animal testing. In this study, we characterize the performance of a preclinical diagnostic test for ovine scrapie based on a monoclonal antibody (MAb)-based immunohistochemistry assay of nictitating membrane (“third eyelid”)-associated lymphoid tissue. The results of third eyelid immunohistochemistry assay agreed with the scrapie status of the sheep for 41 of 42 clinical suspects with confirmed scrapie and 174 of 175 sheep without scrapie. Third eyelid sampling agreed with the scrapie status for 36 of 41 clinically normal sheep positive for PrP-Sc immunostaining of brain tissue, including 27 sheep with positive biopsy specimens that progressed to clinical disease with confirmed scrapie 3 to 20 months after biopsy. This report is the first characterization of the histology and ultrastructure of the barred owl conjunctiva. The inferior eyelid was dominated by a large disk-shaped plate covered by a non-keratinized stratified squamous or cuboidal epithelium of variable thickness. The apical surface of the plate epithelium varied from flat to long microvilli or even short cytoplasmic extensions similar to those seen in the third eyelid. All specimens had a few goblet cells filled with mucous secretory granules in the plate region. The underlying connective tissue was a dense fibroelastic stroma. Eosinophils were surprisingly common in the epithelial layer and underlying connective tissue in the plate and more distal orbital mucosal region. The orbital mucosa contained goblet cells with heterogeneous glycosylation patterns. The leading edge and marginal plait of the third eyelid are designed to collect fluid and particulate matter as they sweep across the surface of the eye. The palpebral conjunctival surface of the third eyelid was covered by an approximately five-cell-deep stratified squamous epithelium without goblet cells. The bulbar surface of the third eyelid was a bilayer of epithelial cells whose superficial cells have elaborate cytoplasmic tapering extensions reaching out 25 μm. Narrow cytofilia radiated outwards up to an additional 15–20 μm from the cytoplasmic extensions. Lectin labeling demonstrated heterogeneous glycosylation of the apical membrane specializations but only small amounts of glycoprotein-filled secretory granules in the third eyelid.

Control of scrapie, an ovine transmissible spongiform encephalopathy or prion disorder, has been hampered by the lack of conventional antemortem diagnostic tests. Currently, scrapie is diagnosed by postmortem examination of the brain and lymphoid tissues for PrPSc, the protein marker for this group of disorders. For live, asymptomatic sheep, diagnosis using tonsil or third-eyelid lymphoid tissue biopsy and PrPSc assay has been described. To evaluate the feasibility and efficacy of third-eyelid testing for identification of infected flocks and individual infected sheep, 690 sheep from 22 flocks were sampled by third-eyelid lymphoid tissue biopsy and immunohistochemistry. Sheep were further evaluated for relative genetic susceptibility and potential contact exposure to scrapie. Third-eyelid testing yielded suitable samples for 80% of the sheep tested, with a mean of 18.1 lymphoid follicles (germinal centers) per histologic section. Three hundred eleven of the sheep were sampled through passive surveillance programs, in which only sheep with potential contact with an infected sheep at a lambing event were tested, regardless of their scrapie susceptibility genotype. In addition, 141 genetically susceptible sheep with no record of contact with an infected animal at a lambing event were sampled through a targeted active surveillance program. The purpose was to evaluate the effectiveness of beta-radiation with strontium-90 as single modality treatment of canine third eyelid squamous cell carcinoma (SCC). Nine dogs diagnosed with third eyelid SCC were treated with strontium-90. Radiation therapy was administered in four fractions of 100cGy per site every four days and at a depth of 0.2cm (Strontium-90 build' up) in each fraction. Radiation with beta therapy was well tolerated in all animals with no occurrence of radiation induced cataracts. In all cases, there were increased signs of conjunctival inflammation around the mass, which subsided with topical anti-inflammatory. Two dogs required surgical treatment for local tumor recurrence at 150 days and 352 days. In the remaining seven cases, disease free interval ranged from 1239 days to 2555 days. The objective of this research was to present an experimental model for the prolapse of the third eyelid gland in male, adult mongrel dogs by the use of a periorbital glandular support conjunctiva tissue resection followed by macro and microscopic assessments. Length, thickness and width of normal glands were statistically studied at baseline and then again after a period of 30 days of exposition. In relation to the experimental model, 84.21% of glands, which underwent the resection of the conjunctiva support tissue, support tissue, remained protusioned. There were significant differences between normal and protusioned glands. Protusioned glands were larger in respect to length, thickness and width. For the microscopy studies, two experimental groups were established: non-treated protusioned glands (GI); and buried protusioned glands (GII). Glandular, conjunctiva tissue, and duct alterations were studied in both groups. Through the results obtained by the Schirmer's test, it was evidenced that the lachrymal production decreased when compared to the normal ones not protusioned, especially in those cases where the protusioned glands were not buried.

Keratoconjunctivitis sicca is characterized by qualitative or quantitative abnormalities in the aqueous component of the tear film and is frequent in dogs with distemper. Twenty-eight third eyelid glands of end-stage dogs that died from distemper were analyzed by light microscopy. Keratoconjunctivitis sicca was diagnosed based on ophthalmic findings regarding alterations in the ocular surface and by the Schirmer tear test. The excised glands were fixed in 10% buffered formalin and processed for embedding in paraffin. The blocks were cut into 4µm sections and the sections were stained with hematoxylin-eosin and Masson's trichrome. Analysis revealed tubuloacinar gland atrophy, thickening of connective tissue, and mild to intense inflammatory exudation mainly characterized by the presence of plasma cells and histiocytes and by compensatory hypertrophy of some glands. The glandular lumen was hypertrophied and contained shedded epithelial cells, polymorphonuclear cells and red blood cells, in addition to mucus.

v.16, n.4, p. 538-547 out./dez. 2015.; Canine visceral leishmaniasis (CVL) is a disease with multisystemic injuries, and among the various tissues affected, the third eyelid (TE) is often involved. This ocular adnexa has been studied for the leishmaniasis pathogenesis elucidation and for diagnosis advance. This study aimed to carry out the parasite immunodetection and evaluate histologic lesions in TE of Leishmania (leishmania) chagasi naturally infected dogs. Twenty-six TE samples from infected dogs were submitted to histologic evaluation and to immunohistochemistry for Leishmania sp. The main lesion observed in the third eyelid conjunctiva (TEC) was lymphoplasmacytic inflammation, with Mott cells and parasitized histiocytes. Additionally, loss of epithelial stratification, ulceration, goblet cells thinning or hyperplasia were often found. The same inflammatory pattern was observed in the third eyelid lacrimal gland (TELG), often accompanied by acinar atrophy and secretory ducts dilatation. The present experiment was carriedout in ten animals with third eyelid prolapsed gland.All of the prolapsed glands were surgically excised.The age of the dogs ranged from two month toten years old, and the breeds were Beagle (20%),Teckel (10%), Pinscher (10%), Poodle (10%) andother mongrel dogs. Aspirative citology was madein all the removed glands, after that they were fi-xed in 10% formalin and stained with Hematoxilineand Eosine. It was observed a focal mononuclearinflamatory infiltrate in the conjunctive and periacinaltissues. Ductal dilatation and metaplasia werenoted on cells of the ducts. Cyitologic examinationshowed similar inflamatory infiltrate to histopatologicfindings.
