OBJETIVO: Analisar a acurácia do diagnóstico de dois protocolos de imunofluorescência indireta para leishmaniose visceral canina. MÉTODOS: Cães provenientes de inquérito soroepidemiológico realizado em área endêmica nos municípios de Araçatuba e de Andradina, na região noroeste do estado de São Paulo, em 2003, e área não endêmica da região metropolitana de São Paulo, foram utilizados para avaliar comparativamente dois protocolos da reação de imunofluorescência indireta (RIFI) para leishmaniose: um utilizando antígeno heterólogo Leishmania major (RIFI-BM) e outro utilizando antígeno homólogo Leishmania chagasi (RIFI-CH). Para estimar acurácia utilizou-se a análise two-graph receiver operating characteristic (TG-ROC). A análise TG-ROC comparou as leituras da diluição 1:20 do antígeno homólogo (RIFI-CH), consideradas como teste referência, com as diluições da RIFI-BM (antígeno heterólogo). RESULTADOS: A diluição 1:20 do teste RIFI-CH apresentou o melhor coeficiente de contingência (0,755) e a maior força de associação entre as duas variáveis estudadas (qui-quadrado=124,3), sendo considerada a diluição-referência do teste nas comparações com as diferentes diluições do teste RIFI-BM. OBJETIVO: Avaliar a eficácia do dispositivo endoscópico temporário de exclusão duodeno jejunal (DEED) na redução da resistência à insulina e do risco cardiovascular em pacientes obesos mórbidos portadores de diabetes tipo 2 (DMT2), utilizando a relação Triglicerídeos/colesterol HDL (TG/HDL), porcentagem de perda de peso e controle glicêmico. MÉTODO: Neste estudo foram incluídos 54 pacientes implantados com o DEED e acompanhados por um período de 6 meses. Todos apresentavam uma relação TG/HDL com valor acima de 3,5, sugerindo uma maior resistência insulínica e um perfil lipídico compatível com um maior risco cardiovascular. O valor da relação inicial foi comparado com o valor obtido após 6 meses do implante do dispositivo, com o objetivo de avaliar se houve redução desse valor, indicando melhora na resistência insulínica e redução do risco cardiovascular. Também foi avaliada a melhora dos níveis de hemoglobina glicada (HbA1c) e a perda de peso obtida com o uso do dispositivo. Latex collected from natural rubber trees forming membranes can be used as biomaterials in several fields being the temperature a key parameter. Thermogravimetry (TG) coupled to Fourier transform infrared spectroscopy (FTIR) is a useful technique to investigate the thermal degradation of both latex and cast films (membranes), wich were obtained from Hevea brasiliensis (RRIM 600 clone) and used without stabilization. The membranes were prepared by casting the latex onto a glass substrate at 65 degrees C for 6 h. The thermal degradation was followed by FTIR spectra acquisition along the process, allowing the identification of the gaseous components evolved upon the thermal treatment. According to TG measurements, the main processes of thermal degradation of the latex and membranes occur at three temperature intervals for both.

Solid-state compounds of yttrium and lanthanide chelates of ethylenediaminetetraacetic acid have been synthesized. Simultaneous thermogravimetry and differential scanning calorimetry (TG-DSC), theoretical and experimental infrared spectroscopy (FTIR), elemental analysis, complexometry and TG-DSC coupled to FTIR were used to characterize and to study the thermal decomposition of these compounds. The results provided information about the composition, dehydration, thermal stability, thermal decomposition and identification of gaseous products evolved during the thermal decomposition of these compounds. The theoretical and experimental spectroscopic data suggest the possible modes of coordination of the ligand with the lanthanum and terbium metal ions. © 2013 Akadémiai Kiadó, Budapest, Hungary.

Obesity is one of the leading causes of health care issues all over the world. A large slice of the worldwide population develops co-morbidities as a consequence of this condition, such as type 2 diabetes mellitus (T2DM), atherosclerosis, fatty liver, cardiovascular diseases and even some cancers, conditions that are associated with reduced life expectancy. The inefficiency of current therapies that aim to treat obesity leads to further investigation on novel therapeutic targets for this condition, including modulation of white adipose tissue (WAT). In WAT, glucose and lipid metabolism converge into triglyceride/ glycerol + free fatty acid (TG/ (GL + FFA)) cycle, which is considered a futile cycle since it doesn’t generate products while consuming energy substrates. The decreased activity of this cycle in obesity, decreases substrates oxidation and increases free fatty acids (FFA) storage in WAT. If coupled to increased mitochondrial β-oxidation, modulation of TG/ (GL+FFA) cycling provides an attractive therapeutic target to stimulate fat mobilization and prevent WAT dysfunction. Dissertação para obtenção do Grau de Mestre em Engenharia Química e Bioquímica; In the present work a stochastic model to be used for analyzing and predicting experimental data from simultaneous thermogravimetric (TG) and differential scanning calorimetry (DSC) experiments on the thermal and catalytic degradation of high-density polyethylene (HDPE) was developed. Unlike the deterministic models, already developed, with this one it’s possible to compute the mass and energy curves measured by simultaneous TG/DSC assays, as well as to predict the product distribution resulting from primary cracking of the polymer, without using any experimental information. For the stochastic model to predict the mass change as well as the energy involved in the whole process of HDPE pyrolysis, a reliable model for the cracking reaction and a set of vaporization laws suitable to compute the vaporization rates are needed. In order to understand the vaporization process, this was investigated separately from cracking. For that, a set of results from TG/DSC experiments using species that vaporize well before they crack was used to obtain a global correlation between the kinetic parameters for vaporization and the number of C-C bonds in the hydrocarbon chain. The best fitting curves were chosen based on the model ability to superimpose the experimental rates and produce consistent results for heavier hydrocarbons. Samples of epoxy resin (EP) based on the diglycidyl ether of bisphenol A (DGEBA), cured with nadic methyl anhydride (NMA) and mercaptan (CAPCURE 3-800), or amine-phenol (CAPCURE EH-30), respectively, SE 4 and SE 5, were prepared in the stoichiometric ratio determined by the supplier. The curing behaviour of the epoxy systems SE 4 and SE 5 was followed by DSC (Differential Scanning Calorimetry). These SE and their ingredients were analyzed using FTIR transmission techniques (KBr pellets, pyrolysis without control and controlled pyrolysis-FT-IR, the CONTROLPIR/FT-IR) for characterizing the curing agents (CA). The temperature range used for samples pyrolysis was found from TG (thermogravimetry). Thus, the FT-IR bands of liquid pyrolysate obtained by CONTROLPIR/FT-IR were evaluated, in comparison to the reference spectrum of CA. The characterization of CA was also possible, by the analysis of such SE with the TG/FT-IR technique.

No presente trabalho foram desenvolvidos comprimidos de captopril revestidos com hidroxipropilmetilcelulose (HPMC), Opadry®, polivinilpirrolidona (PVP), Eudragit® E e goma laca. Foi realizado estudo termoanalítico do fármaco e das formulações através de termogravimetria (TG) e calorimetria exploratória diferencial (DSC). Através da análise das curvas DSC verificou-se que não houve a ocorrência de interação entre o fármaco e os excipientes lactose, celulose microcristalina, croscarmelose sódica, Aerosil® e talco, utilizados na formulação do comprimido. Através desta técnica detectou-se a possibilidade de interação entre captopril e estearato de magnésio. De acordo com os resultados obtidos através de DSC não foram observadas alterações na cristalinidade do fármaco decorrentes dos processos de compressão e revestimento. A termogravimetria foi utilizada para o estudo da cinética de degradação do captopril e dos comprimidos. Os parâmetros cinéticos foram determinados através do método de Ozawa. Os resultados demonstraram que não houve alteração da estabilidade térmica do captopril na forma de comprimido. A formulação revestida com HPMC foi a que apresentou maior estabilidade térmica, quando comparada às demais formulações de revestimento.

Human Tg, the site of synthesis of thyroid hormones, thyroxine (T4) and triiodothyronine (T3), is one of the major autoantigens in autoimmune thyroiditis. The degree of iodination of Tg may have a major impact on its immunological properties by changing its antigenicity with respect to antibody binding. We have previously prepared a panel of MoAbs that bind to different epitopes of the Tg molecule. In the present study, we show that iodination alters the conformation of Tg molecule in such a way that it is recognized differently by different MoAbs. Monoclonal antibody 137C1 recognizes Tg regardless of its iodine content. Monoclonal antibody 42C3 recognizes Tg only if the Tg is iodinated either in vitro or in vivo. Monoclonal antibody 133B1 recognizes both in vivo iodinated Tg and non-iodinated Tg, but this MoAb did not recognize Tg following in vitro iodination. Monoclonal antibody 41A5 recognizes intact Tg and tryptic peptides of normal (in vivo) iodinated and non-iodinated Tg, but did not bind the tryptic peptides of artificially (in vitro) iodinated Tg. From the results of these experiments, we conclude that iodination of Tg by either in vivo or in vitro methods changes its conformation in such a way that some natural epitopes are ‘lost’ and some ‘new’ epitopes are generated. The generation of new epitopes may be important in the generation of autoimmune responses leading to autoimmune disease.

Mast cells were depleted in the peritoneal cavity of WBB6F1-tg/tg mice that did not express a transcription factor, MITF. When acute bacterial peritonitis was induced in WBB6F1-+/+, WBB6F1-W/Wv, and WBB6F1-tg/tg mice, the proportion of surviving WBB6F1-+/+ mice was significantly higher than that of surviving WBB6F1-W/Wv or WBB6F1-tg/tg mice. The poor survival of WBB6F1-W/Wv and WBB6F1-tg/tg mice was attributed to the deficient influx of neutrophils into the peritoneal cavity. The injection of cultured mast cells (CMCs) derived from WBB6F1-+/+ mice normalized the neutrophil influx and reduced survival rate in WBB6F1-W/Wv mice, but not in WBB6F1-tg/tg mice. This was not attributable to a defect of neutrophils because injection of TNF-α increased the neutrophil influx and survival rate in both WBB6F1-W/Wv and WBB6F1-tg/tg mice. Although WBB6F1-+/+ CMCs injection normalized the number of mast cells in both the peritoneal cavity and mesentery of WBB6F1-W/Wv mice, it normalized the number of mast cells only in the peritoneal cavity of WBB6F1-tg/tg mice. Mast cells within the mesentery or mast cells in the vicinity of blood vessels appeared to play an important role against the acute bacterial peritonitis. WBB6F1-tg/tg mice may be useful for studying the effect of anatomical distribution of mast cells on their antiseptic function.

Adipose tissue lipoprotein lipase (LPL) is a necessary enzyme for storage of VLDL-TG, but whether it is a rate determining step is unknown. To test this hypothesis we included 10 upper-body obese (UBO), 11 lower-body obese (LBO) and 8 lean women. We infused ex-vivo labeled VLDL-14C-TG and then performed adipose tissue biopsies to understand the relationship between VLDL-TG storage and LPL-activity in femoral and upper-body subcutaneous fat. Both fractional tracer storage and rate of storage of the VLDL-TG tracer were evaluated. VLDL-TG storage was also examined as a function of regional adipose tissue blood flow (ATBF), insulin, VLDL-TG turnover, regional fat mass, FFM, and fat cell size. LPL-activity per adipocyte was significantly greater in obese than lean women but not significantly different per gram lipid. Both VLDL-TG fractional tracer storage per kg lipid and VLDL-TG storage rate per kg lipid were similar in abdominal and femoral fat in all three groups and not significantly different between groups. Self-reactive B cells in BALB/c AM14 transgenic (AM14 Tg) rheumatoid factor (RF) mice are not subject to central or peripheral toleralization. Instead, they remain at a stage of “clonal ignorance”, i.e. they do not proliferate and differentiate into Ab-producing cells. However, the immunoregulatory mechanisms that prevent autoantibody production in these mice remain unclear. In this study, we show that crossing AM14 Tg mice to a mouse strain deficient in Act1, a molecule involved in the regulation of BAFF-R and CD40-signaling in B cells, results in spontaneous activation of AM14 Tg B cells and production of AM14-specific antibodies. Three to five-month old AM14 Tg Act1−/− mice showed significant expansion of AM14 Tg B cells, including a 2–3 fold increase in the spleen and cLNs compared to AM14 Tg Act1+/+ mice. Furthermore, in the presence of endogenous self-Ag (IgHa congenic background), AM14 Tg Act1−/− B cells were spontaneously activated and differentiated into antibody forming cells (AFC). This study was to evaluate the usefulness of serum thymoglobulin (Tg) in adults to assess iodine status through a 5-year cohort study which was conducted in three regions with different levels of iodine intake: mild deficiency, more than adequate, and excess, from 1999 to 2004 in China. A total of 3099 subjects over 14 years old with normal serum levels of Tg in 1999 were eligible, of whom 2448 were followed in 2004. Serum levels of thyroid hormones and thyroid autoantibodies as well as urine iodine were measured, and B-mode ultrasonography of the thyroid was performed. A general linear model was performed to determine the determinant factors of serum Tg. Among subjects with mildly deficient iodine intake, those with more than adequate intake, and those with excessive intake, the baseline levels of serum Tg were substantially different (7.5μg/L, 5.9μg/L, and 6.8μg/L respectively, P<0.01), which were associated with age, sex, the rate of positive TgAb, abnormal thyroid volume, abnormal TSH, and positive personal history of thyroid diseases. Bioidentical hormones are defined as compounds that have exactly the same chemical and molecular structure as hormones that are produced in the human body. It is believed that the use of hormones may be safer and more effective than the non-bioidentical hormones, because binding to receptors in the organism would be similar to the endogenous hormone. Bioidentical estrogens have been used in menopausal women, as an alternative to traditional hormone replacement therapy. Thermal data of these hormones are scarce in literature. Thermal analysis comprises a group of techniques that allows evaluating the physical-chemistry properties of a drug, while the drug is subjected to a controlled temperature programming. The thermal techniques are used in pharmaceutical studies for characterization of drugs, purity determination, polymorphism identification, compatibility and evaluation of stability. L’accumulation de triglycérides (TG) dans les hépatocytes est caractéristique de la stéatose hépatique non-alcoolique (SHNA). Cette dernière se produit dans diverses conditions dont le facteur commun est le métabolisme anormal des lipides. Le processus conduisant à l'accumulation des lipides dans le foie n’a pas encore été totalement élucidé. Toutefois, des lipides s'accumulent dans le foie lorsque les mécanismes qui favorisent leur exportation (oxydation et sécrétion) sont insuffisants par rapport aux mécanismes qui favorisent leur importation ou leur biosynthèse. De nos jours il est admis que la carence en œstrogènes est associée au développement de la stéatose hépatique. Sensitivity of thyroglobulin (Tg) measurement in the follow-up of differentiated thyroid carcinoma (DTC) can be optimized by using a sensitive Tg assay and rhTSH stimulation. We evaluated the diagnostic yield of a sensitive Tg assay and rhTSH stimulated Tg in the detection of recurrences in the follow-up of DTC. Additionally the value of imaging techniques for the localization of recurrences was evaluated. We included 121 disease free patients in long-term follow-up for DTC (median 10 years, range 1–34). Tg during thyroid hormone suppression therapy (Tg-on) and rhTSH stimulated Tg were measured with a sensitive Tg assay. Patients with rhTSH stimulated Tg ≥1.0 ng/ml underwent imaging with neck ultrasound, FDG-PET and post therapy 131I WBS. Sensitive Tg measurement resulted in 3 patients with Tg-on ≥1.0 ng/ml, recurrence could be localized in 2 of them. RhTSH stimulation resulted in Tg ≥1.0 ng/ml in another 17 of 118 patients. Recurrence could be localized in only 1 additional patient (1 out of 118 patients). Recurrence was localized by neck ultrasound in 1 of 3, by FDG-PET in 2 of 3 and by post therapy 131I WBS in 2 of 3 patients. In the detection of recurrences in DTC, rhTSH stimulation had very limited additional value in comparison to Tg-on measurement with a sensitive Tg assay. No presente trabalho foram desenvolvidos comprimidos de captopril revestidos com hidroxipropilmetilcelulose (HPMC), Opadry®, polivinilpirrolidona (PVP), Eudragit® E e goma laca. Foi realizado estudo termoanalítico do fármaco e das formulações através de termogravimetria (TG) e calorimetria exploratória diferencial (DSC). Através da análise das curvas DSC verificou-se que não houve a ocorrência de interação entre o fármaco e os excipientes lactose, celulose microcristalina, croscarmelose sódica, Aerosil® e talco, utilizados na formulação do comprimido. Através desta técnica detectou-se a possibilidade de interação entre captopril e estearato de magnésio. De acordo com os resultados obtidos através de DSC não foram observadas alterações na cristalinidade do fármaco decorrentes dos processos de compressão e revestimento. A termogravimetria foi utilizada para o estudo da cinética de degradação do captopril e dos comprimidos. Os parâmetros cinéticos foram determinados através do método de Ozawa. Os resultados demonstraram que não houve alteração da estabilidade térmica do captopril na forma de comprimido. OBJETIVO: Analisar a acurácia do diagnóstico de dois protocolos de imunofluorescência indireta para leishmaniose visceral canina. MÉTODOS: Cães provenientes de inquérito soroepidemiológico realizado em área endêmica nos municípios de Araçatuba e de Andradina, na região noroeste do estado de São Paulo, em 2003, e área não endêmica da região metropolitana de São Paulo, foram utilizados para avaliar comparativamente dois protocolos da reação de imunofluorescência indireta (RIFI) para leishmaniose: um utilizando antígeno heterólogo Leishmania major (RIFI-BM) e outro utilizando antígeno homólogo Leishmania chagasi (RIFI-CH). Para estimar acurácia utilizou-se a análise two-graph receiver operating characteristic (TG-ROC). A análise TG-ROC comparou as leituras da diluição 1:20 do antígeno homólogo (RIFI-CH), consideradas como teste referência, com as diluições da RIFI-BM (antígeno heterólogo). RESULTADOS: A diluição 1:20 do teste RIFI-CH apresentou o melhor coeficiente de contingência (0,755) e a maior força de associação entre as duas variáveis estudadas (qui-quadrado=124,3), sendo considerada a diluição-referência do teste nas comparações com as diferentes diluições do teste RIFI-BM. El disturbio inicial de la insulinorresistencia (IR) parece centrarse en el tejido adiposo que es un órgano dinámico involucrado en muchos procesos fisiológicos y metabólicos. Expresa y secreta una gran variedad de péptidos activos, adipocitoquinas. En el presente estudio se propuso determinar la relación entre el índice TG/col-HDL, marcadores del metabolismo del tejido adiposo (Adiponectina) y de inflamación (PCRus) con la IR en una población urbana del interior del país. Se evaluaron 176 empleados de los 2 hospitales públicos de Posadas-Misiones, 117 mujeres y 59 hombres, con edades 49,01 ± 9,33 años. Se excluyeron los individuos que presentaban diabetes, hipotiroidismo o hipertiroidismo, enfermedad infecciosa, renal, hepática o neoplasias y embarazadas. Se les realizó una extracción sanguínea con 12 h de ayuno.
