Nos processos contínuos há uma forte participação dos profissionais da manutenção e da engenharia na operação do sistema de produção. Manutenção e engenharia atuam diretamente na operação quando especificam parâmetros de controle do processo, intervêm em eventos previstos ou não, resolvem problemas de grande complexidade tecnológica e logística, e modificam processos e equipamentos para obter melhores resultados. Por outro lado, é a operação que detém o conhecimento de como realizar manobras, da inércia das instalações para responder a comandos, dos efeitos práticos do encadeamento e desdobramento de manobras, da alteração de parâmetros de controle, ou da indisponibilidade de equipamento, influenciando decisivamente o espectro de ação da engenharia e da manutenção. Não obstante, mesmo as mais avançadas e contemporâneas abordagens para projeto organizacional não apresentam critérios integrados de projeto para essas três decisivas funções, que se interpenetram. Diante do crescente desenvolvimento da tecnologia da informação e da comunicação, o sistema educacional vem gradualmente modificando as estratégias de ensino-aprendizagem, adaptando para tanto, os tradicionais procedimentos e técnicas didáticas aos instrumentos tecnológicos fornecidos pelos ambientes virtuais de aprendizagem. O processo de avaliação e acompanhamento da aprendizagem nestes ambientes requer o uso de novas estratégias de ensino que congregue os recursos tecnológicos e a intervenção positiva do professor/tutor. Propõe-se um modelo de design instrucional estendido do "Integrative Learning Design Framework On-line" (ILDF) sob o enfoque da taxonomia dos objetivos educacionais de Bloom e do ciclo de aprendizagem vivencial de Kolb adaptado por Belhot para uma disciplina de pós-graduação em Engenharia de Produção. Os artigos científicos são o principal veículo de comunicação para divulgação dos resultados obtidos por milhares de pesquisadores ao redor do mundo. Entretanto, escrever os artigos científicos em publicações de alto impacto é uma tarefa complexa para autores que têm o inglês como segunda língua. Por meio da aprendizagem baseada em exemplos e em experiência, as habilidades de leitura, interpretação e redação de textos científicos podem ser desenvolvidas por intermédio da prática, facilitada por meio de exemplos de trechos de texto reutilizáveis. Para auxiliar na redação de artigos, várias ferramentas computacionais foram desenvolvidas para dar suporte nas várias etapas de concepção do artigo. As ferramentas existentes, que aplicam este tipo de suporte, são todas voltadas para outros temas de pesquisa, incompatíveis com os temas da Engenharia de Produção, que por sua vez possui maneiras particulares de redigir e estruturar o texto. The Sulfur-Iodine Cycle for the thermochemical production of hydrogen offers many benefits to traditional methods of hydrogen production. As opposed to steam methane reforming - the most prevalent method of hydrogen production today - there are no carbon dioxide emissions. Compared to other methods of hydrogen production, the efficiency of the cycle is excellent. Due to the high temperatures necessary for the cycle, which are generally greater than 8500C, several of the Generation IV nuclear reactor concepts are attractive thermal energy sources. However, the high temperature and corrosive reaction conditions of the cycle, involving reactions including the decomposition of H2SO4 at 400-9000C, present formidable corrosion challenges. The conversion of sulfuric acid to sulfur dioxide was the focus of this study. The alloying of structural materials to platinum has been proposed as a solution to this problem. A catalytic loop to test the materials was constructed. Sulfuric acid was pumped over the material at 903+20C. The sulfur dioxide production of the catalyst was measured as a means of quantifying the efficiency of the system as a function of temperature.; (cont.) The maximum possible production of the material was calculated by using a mass balance. A gas chromatograph was used to calculate the actual production of sulfur dioxide. The results of the experiment show that an molecular conversion efficiency of 10% is attained when operating at 900C while using 800H + 5%Pt as a catalyst. Recent advances in tissue engineering, combining an extracellular matrix (ECM)-like vehicle with therapeutic molecules, cells and/or genes has yielded promising results for brain injury repair. The purpose of this thesis was to develop a collagen scaffold for the non-viral delivery of the gene encoding for Glial Cell-Derived Neurotrophic Factor (GDNF); hence to provide a local, long-term release and overexpression of GDNF via transfection of cells seeded into the scaffold or endogenous cells. The first part of the thesis aimed to investigate the in vitro transfection of marrow stromal stem cells (also referred to as mesenchymal stem cells, MSCs) in monolayer with plasmid GDNF ([mu]gDNF). Several parameters were evaluated: the choice of a transfer reagent (GenePorter2 versus Lipofectamine 2000), the doses of plasmid incorporated in the liposomes (ranging from 0.2[mu]g to 2[mu]g), the post-transfection medium (Medium 1: DMEM low glucose, 20% FBS and 1% antibiotic versus Medium 2: DMEM low glucose, 20% FBS, 1% antibiotic and 10ng/ml FGF-2) and the culture environment during transfection (static versus dynamic). The purpose of this thesis was to provide a broad survey of bioprocess options for typical drug production vehicles in the biotechnology industry. This goal was accomplished by comparing the growth and monoclonal antibody production by HPCHO Chinese hamster ovary cells and IB4 hybridoma cells in batch, fed-batch, and three perfusion systems: the stirred ceramic membrane reactor (SCMR), the alternating tangential flow (ATF) hollow fiber membrane system, and the external spin filter (ESF) system. The batch experiments for each cell line were used as base case scenarios for the comparisons of cell growth and monoclonal antibody production. The fed-batch experiments for each cell line did not vastly improve the cell growth and monoclonal antibody production over the batch cases. The SCMR perfusion system greatly improved the cell growth and monoclonal antibody production for each cell line. The maximum viable cell concentration of the IB4 hybridoma cells in the SCMR experiment was over seven times that found in the batch experiment and the monoclonal antibody production in the SCMR experiment was over ten times that found in the batch experiment.; (cont.) Explorations and optimizations through the genomic space are a daunting undertaking given the complexity and size of the possible search space. To approach this problem, systematic and combinatorial approaches were employed for the engineering of cellular phenotype in Escherichia coli. Initially, a computational method based on global cellular stoichiometry was employed to identify single and multiple gene knockout targets for lycopene production in E. coli. These targets led to substantial increases in lycopene production, but were limited in scope due to the nature of these models. Therefore, these approaches and targets were complemented with combinatorial searches to identify unknown and regulatory targets. When combined, these searches led to further increases of lycopene production and allowed for the visualization of the resulting metabolic landscape. A more exhaustive search was conducted in the background of eight genotypes which resulted in the formulation of the gene knockout search network. This network enables the investigation into how phenotype optimization is biased by search strategy.; (cont.) Developing a cyclical schedule for producing multiple items on a single processor under stochastic demand that minimizes total setup and inventory holding costs is an important problem. This problem is faced in many continuous production environments such as chemical or petrochemical production, where the costs of switching production from one item over to another are very high. Due to high changeover costs, each item is produced in large batches and its demand is fulfilled from the finished goods inventory. Holding this inventory incurs inventory carrying costs. Therefore, good production planning policies are required to determine when and how much of each item should be produced so that the total cost of setup changeovers and holding inventory is minimized, while ensuring that sufficient inventory is available to meet customer demand which varies over time. In this thesis, I present the work done to develop production planning policies for a large chemical manufacturing company that operates in the environment described above. This research investigates the production output and inventory distribution of a manufacturing system that produces multiple part types on flexible machines while incorporating CONWIP inventory controls. The production machines, which are separated by individual storage areas for each part type, are modeled as unreliable and operate according to a strict priority sequence. The maximum total inventory of each part type in the entire system is limited a preset number of tokens in the system. While varying the token levels and the demand for finished parts, the average production output for each part type and the average inventory level for each buffer is monitored to understand their behaviors. The relationship between the inventory distributions and the production rates is also investigated. In the transportation sector, the current dependence on petroleum to satisfy large transportation fuel demand in the US is unsustainable. Oil resources are finite, and causing heavy US reliance on oil imports. Therefore, the development of alternative transportation fuels that do not depend on oil is becoming increasingly necessary. Our research investigates the feasibility of producing gasoline synthetically from nuclear hydrogen and two carbon sources: carbon dioxide emissions and municipal solid waste. These synthetic fuels have the potential to satisfy the large demand for gasoline, while reducing CO2 emissions. The nuclear hydrogen is produced through High Temperature Steam Electrolysis (HTSE), with heat and electricity provided by a supercritical CO2 cooled gas fast reactor. Through this study, we determine the suitable components for gasoline production from CO2 emissions and MSW. The feasibility of these methods of gasoline production was assessed by performing material and energy balances for the involved processes, determining preliminary cost estimates, and evaluating production scale and environmental impact. Rising natural gas prices and growing concern over CO₂ emissions have intensified interest in alternative methods for producing hydrogen. Nuclear energy can be used to produce hydrogen through thermochemical and/or electrochemical processes. This thesis investigates the feasibility of high temperature steam electrolysis (HTSE) coupled with an advanced gas-cooled fast reactor (GFR) utilizing supercritical carbon dioxide (S-CO₂) as the coolant. The reasons for selecting this particular reactor include fast reactor uranium resource utilization benefits, lower reactor outlet temperatures than helium-cooled reactors which ameliorate materials problems, and reduced power conversion system costs. High temperature steam electrolysis can be performed at conditions of 8500C and atmospheric pressure. However, compression of the hydrogen for pumping through pipes is unnecessary if electrolysis takes place at around 6 MPa. The reactor coolant at 6500C is used to heat the steam up to temperatures ranging between 2500C and 3500C, and the remaining heat is provided by thermal recuperation from product hydrogen and oxygen. This thesis describes the engineering tasks associated with production of Design Squad, a reality-based television program on WGBH. In Design Squad a client gives two teams of high school students an engineering design task for them to complete over two days of building. In order to facilitate production of this program an extensive amount of engineering preparation work is required. Tasks for each episode include concept brainstorming, prototyping of challenge solutions, material procurement, and on-set mentoring. These development and preparation activities require creative brainstorming, teamwork, and communication across a multi-disciplinary team. The MIT based engineering support team has created methods and guidelines to ensure efficient knowledge generation during prototyping that utilize sample builds, proofs of concept, and sketching. Techniques have also been developed for mentoring and guiding the cast members on set. Forward metabolic engineering (FME) is a rational approach to cellular engineering, relying on an understanding of the entire metabolic network to direct perturbations for phenotype improvement. Conversely, inverse metabolic engineering (IME) uses a global, combinatorial approach to identify genetic loci that are important for a given phenotype. These two approaches complement each other in a strain improvement program. FME and IME approaches were applied to poly-3-hydroxybutyrate (PHB)production in Synechocystis PCC6803 [IME] and recombinant E. coli [FME] in this thesis.IME was appropriate for Synechocystis, where metabolic regulation of the native PHB pathway was not well understood. A high throughput screening method was established by developing a staining protocol that quantitatively related nile red fluorescence to PHB content, while maintaining cell viability for both organisms. This was combined with fluorescence activated cell sorting (FACS) to screen for high PHB mutants. A Synechocystis insertion mutagenesis library was screened to identify gene disruptions that increased PHB. Two gene disruptions in proline biosynthesis and an unknown function were identified and characterized.An analogous IME study in E. coli did not find increased PHB mutants...

This Master of Science Thesis deals with applying DEA (Data Envelopment Analysis) to the academic performance evaluation of graduate programs in Brazil, exploring it on a Mechanical and Production Engineering Program 2001-2003 data. The data used is that of the national assessment carried by CAPES, the governmental body in charge for graduate program assessment and certification. It is used the CCR output oriented DEA model, the CCR-Output with Assurance Region, and Window Analysis. The main findings are first that the CCR has the concerning problem of zero values of weights of outputs that is not appropriate in a sense that a graduate program has the higher efficiency score zeroing some output (e.g., number of academic papers published). Secondly, the Assurance Region method proved useful. Third, the Window Analysis also gave some light to the consistency of the performance in the time frame analysed. Also, the analysis results in the understanding that the Mechanics and Production Engineering should not be assessed jointly like currently applied by CAPES and rather should be assessed in its own field separately. Finally, the result of the DEA analysis showed some serious inconsistencies with the CAPES method. Graduate programs considered excellent has got low performance score and vice versa. As field research, the method of collection of data if constituted of the elaboration of an analysis protocol and, to arrive itself at the final diagnosis, it operation the data through the statistical method, with the accomplishment of descriptive analyses. O objetivo deste artigo é retratar parte do trabalho avaliativo desenvolvido em disciplinas de um curso de Engenharia de Produção, nas quais foram utilizados diversos conteúdos multimídia, via Internet, como ferramenta de apoio ao processo de ensino-aprendizagem. Este trabalho relata desde a fase de desenvolvimento dos conteúdos, passando pela descrição dos meios utilizados e o trabalho do professor e formadores, culminando no relato dos resultados obtidos nas notas finais dos alunos. Os resultados obtidos dão forte evidência de que o blended-learning tende a ser um aliado de extrema importância em disciplinas da Engenharia de Produção e no ensino superior em geral. As análises realizadas evidenciam ganhos de qualidade, os quais são comprovados pelos relatos dos alunos e pela qualidade medida através do desempenho dos mesmos.; The scope of this paper is to describe a section of the evaluation work developed in Production Engineering disciplines from UNIFEI, which uses many multimedia contents through the web as a complementary form of education. This report starts describing the development of the course and goes until the ways employed and the activities of the teacher and collaborators, ending with the analysis of the students’ final grades. A gestão ambiental tem emergido como uma questão estratégica para muitas organizações. A partir deste tema geral, surgem outros, como por exemplo, Ecoeficiência e Produção Mais Limpa, que também vêm ganhando destaque e importância. Porém, tanto no meio acadêmico quanto no empresarial há certa carência de conhecimento frente, principalmente, aos dois primeiros conceitos, Ecoeficiência e Produção Mais Limpa, sobretudo no que se refere às aplicações dos mesmos. Nesse contexto, o presente trabalho objetiva realizar uma análise, sobre a produção acadêmica que aborda e discute os termos Ecoeficiência e Produção Mais Limpa em quatro periódicos da Engenharia de Produção. Os seguintes indicadores foram analisadas neste estudo: número total de publicações encontradas; número de artigos publicados por periódicos; definições utilizadas pelos autores das publicações para os termos; número de palavras-chaves; número total de artigos relacionados aos dois conceitos, bem como quantificação das referências desses artigos. Conclui-se que sete publicações dos quatro periódicos abordam a Produção Mais Limpa e nenhum artigo trata do conceito de Ecoeficiência.
