A Rare Case of Trisomy 15pter-q21.2 Due to a De Novo Marker Chromosome

Supernumerary marker chromosomes (sSMC) may or may not be associated with an abnormal phenotype, depending on the presence of euchromatin, on their chromosomal origin and whether they are inherited. Over 80% of sSMCs are derived from acrocentric chromosomes and half of them include the short arm of chromosome 15. Generally, they appear as bisatellited isodicentric marker chromosomes, most of them are symmetric. These chromosomes are normally originated de novo and are associated with mild to severe intellectual disability but not with physical abnormalities. We report on a patient with an SMC studied using classical and molecular cytogenetic procedures (G and C banding, NOR staining, painting and centromeric fluorescent in situ hybridization (FISH), BAC-FISH, and SKY). The MLPA technique and DNA polymorphic markers were used in order to identify its parental origin. The marker chromosome, monosatellited and monocentric, was found to be derived from a maternal chromosome 15 and was defined as 15pter-q21.2. Este projeto objetivou desenvolver um marcador molecular ligado ao gene de resistência a Tobacco mosaic virus (TMV), em vista da necessidade de aprimorar os métodos de melhoramento de plantas para atender crescentes demandas de produtividade. O outro objetivo deste trabalho foi a avaliação de uma população segregante F2 e de retrocruzamento (RC1F1) a Meloidogyne incognita raça 3, oriunda do cruzamento das cultivares comerciais Coker 176 (C176) e Coker 371 Gold (C371G). Para o desenvolvimento do marcador ligado ao gene de resistência a TMV, o gene N, foram desenvolvidos iniciadores específicos para regiões conservadas (TIR, NBS e LRR) deste gene com base em sua seqüência. Estes iniciadores foram utilizados para amplificar um marcador cuja ligação ao referido gene foi confirmada em 200 indivíduos de população segregante F2 oriunda do cruzamento entre uma linhagem resistente (Coker176) e outra suscetível ao vírus (Kentucky326). A proporção entre o número de plantas resistentes e suscetíveis (154:46) não diferiu estatisticamente daquela esperada no caso de segregação de um gene dominante de resistência, que seria de 3:1. Rearranjos cromossômicos envolvendo a presença de cromossomos marcadores extranumerário são achados citogenéticos freqüentes em pacientes que apresentam deficiência mental, alterações de crescimento, dismorfias e/ou malformações. A presença desse material é responsável por trissomia ou tetrassomia parcial de determinadas regiões cromossômicas, causando quadros clínicos distintos e inespecíficos. A variabilidade fenotípica está relacionada principalmente com os diferentes graus de mosaicismo, os genes presentes na região adicional, o cromossomo de origem, entre outros fatores. Sendo assim, a caracterização desse material cromossômico é de importância fundamental para a determinação do prognóstico e do aconselhamento genético dos pacientes e suas famílias. O presente estudo teve como objetivo a análise de cromossomos marcadores extranumerários por meio de técnicas de citogenética convencional e molecular. Foram selecionados onze pacientes que são acompanhados pelo o Serviço de Genética Médica do Hospital das Clínicas da Faculdade de Medicina de Ribeirão Preto USP, todos com diagnóstico citogenético convencional por bandeamentos GTG de cromossomo marcador extranumerário. Para determinação da origem e caracterização dos cromossomos marcadores foram aplicadas as técnicas de Cariótipo Espectral (SKY) e de Hibridação in situ Fluorescente (FISH). Este trabalho analisa filmes do cineasta Chris Marker que indagaram sobre a Unidade Popular do Chile nos anos 1970. Entre as produções abordadas, estão duas que foram remontadas a partir de documentários chilenos feitos durante o governo de Salvador Allende: La première année (1973) e On vous parle du Chili: ce que disait Allende (1973). Além disso, mais três filmes desse realizador se dedicaram ao tema: La Spirale (1976), Lambassade (1974) e O fundo do ar é vermelho (1977). Por meio do estudo dessa filmografia, identificaram-se quais foram as estratégias narrativas utilizadas e qual foi o discurso político sobre a chamada experiência chilena que essas estratégias elaboraram. A reflexão sobre o corpus permitiu verificar que o interesse por esse processo, visto desde a França, emergiu do espaço deixado pelas frequentes desilusões diante de uma referência política anterior, igualmente vinda da América Latina e que havia motivado os debates da esquerda francesa na década de 1960: a Revolução Cubana. The objective of this paper was to determine the association of a SNP in the µ-calpain gene at position 316 with growth and quality of meat traits of steers grown on pasture. Fifty-nine Brangus and 20 Angus steers were genotyped for CAPN1 316. Warner Bratzler shear force was measured in l. lumborum samples after a 7-day aging period. A multivariate analysis of variance was performed, including shear force (WBSF), final weight (FW), average daily gain (ADG), backfat thickness (BFT), average monthly fat thickness gain (AMFTG), rib-eye area (REA), and beef rib-eye depth (RED) as dependent variables. The CAPN1 316 genotype was statistically significant. Univariate analyses were done with these variables. The marker genotype was statistically significant (p < 0.05) for WBSF (kg: CC: 4.41 ± 0.57; CG: 5.58 ± 0.20; GG: 6.29 ± 0.18), FW (kg: CC: 360.23 ± 14.71; CG: 381.34 ± 5.26; GG: 399.23 ± 4.68), and ADG (kg/d: CC: 0.675 ± 0.046; CG: 0.705 ± 0.016; GG: 0.765 ± 0.014) Shear force, final weight and average daily gain were significantly different according to the CAPN1 316 marker genotypes. The objectives of this study were to verify the use of LW7 marker in identifying maintainer lines (B-lines) and restorer lines (R-lines) in grain sorghum and sweet sorghum, and to identify B-lines in the F2, BC1F2 and BC2F2 generations. Twenty five accessions of sorghum were evaluated, and LW7 marker correctly identified accessions which presented male sterility gene (rf4) in Suphan Buri1 and 03B cultivars; moreover, these genotypes did not show 779 bp band. The cross between Suphan Buri1 and a male-sterile line (A-line) 03A resulted in a sterilized male, confirming the usefulness of the marker in breeding programs. B-lines in the F2, BC1F2 and BC2F2 generations were identified by LW7 marker. The segregation ratio of 3:1 for male fertility and male sterility in the progenies of the three generations supported the one-gene model of Mendelian segregation. The use of marker assisted selection was successful for line development of sweet sorghum with male sterility.

YLM, a codominant polymerase chain reaction (PCR) marker linked to Yd2, could substantially improve the precision and efficiency of barley yellow dwarf virus (BYDV) resistance breeding. The aim of this study was to assess the effectiveness of YLM in a marker-assisted introgression programme and to quantify associations between the presence of Yd2 and other agronomic and quality traits. The Yd2 gene was introgressed into a BYDV-susceptible background through two cycles of marker-assisted backcrossing. BC₂ F₂-derived lines, either carrying or not carrying the YLM allele associated with resistance, were compared in the presence and absence of BYDV. The YLM marker was shown to be effective in the introgression of Yd2. Lines carrying the YLM allele associated with resistance produced significantly fewer leaf symptoms and showed a reduction in yield loss when infected with BYDV. There were no deleterious effects associated with the introgression of Yd2 on grain yield, grain size or malting quality. The implications of marker-assisted selection for Yd2 on barley improvement are discussed.

Marker assisted selection (MAS) has become a valuable asset to plant breeders as it provides an alternative strategy to introgress valuable alleles into breeding material. As part of its integration into mainstream breeding, finding reliable low cost, high throughput screening techniques is an important aspect of most implementation programs. With recent advancements in molecular marker techniques and the growing availability of commercial genotyping services, there are now a number of options available to breeders for the implementation of MAS. This paper explores the value of three genotyping methods for marker assisted breeding in barley, namely: the traditional approach using conventional PCR and ethidium bromide stained polyacyrlamide gels, a new technology employing multiplex PCR and fluorescent-based marker detection on an automated DNA fragment analyser, and outsourcing to a commercial service provider. A doubled haploid population screened using these methods are compared in relation to the cost, speed, convenience and reliability of each approach.

A number of useful marker-trait associations have been reported for wheat. However the number of publications detailing the integrated and pragmatic use of molecular markers in wheat breeding is limited. A previous report by some of these authors showed how marker-assisted selection could increase the genetic gain and economic efficiency of a specific breeding strategy. Here, we present a practical validation of that study. The target of this breeding strategy was to produce wheat lines derived from an elite Australian cultivar ‘Stylet’, with superior dough properties and durable rust resistance donated from ‘Annuello’. Molecular markers were used to screen a BC1F1 population produced from a cross between the recurrent parent ‘Stylet’ and the donor parent ‘Annuello’ for the presence of rust resistance genes Lr34/Yr18 and Lr46/Yr29. Following this, marker-assisted selection was applied to haploid plants, prior to chromosome doubling with cochicine, for the rust resistance genes Lr24/Sr24, Lr34/Yr18, height reducing genes, and for the grain protein genes Glu-D1 and Glu-A3. In general, results from this study agreed with those of the simulation study. Genetic improvement for rust resistance was greatest when marker selection was applied on BC1F1 individuals. Introgression of both the Lr34/Yr18 and Lr46/Yr29 loci into the susceptible recurrent parent background resulted in substantial improvement in leaf rust and stripe rust resistance levels. Pre-harvest sprouting (PHS) is a complex trait controlled by multiple genes with strong interaction between environment and genotype that makes it difficult to select breeding materials by phenotypic assessment. One of the most important genes for pre-harvest sprouting resistance is consistently identified on the long arm of chromosome 4A. The 4AL PHS tolerance gene has therefore been targeted by Australian white-grained wheat breeders. A new robust PCR marker for the PHS QTL on wheat chromosome 4AL based on candidate genes search was developed in this study. The new marker was mapped on 4AL deletion bin 13-0.59-0.66 using 4AL deletion lines derived from Chinese Spring. This marker is located on 4AL between molecular markers Xbarc170 and Xwg622 in the doubled-haploid wheat population Cranbrook × Halberd. It was mapped between molecular markers Xbarc170 and Xgwm269 that have been previously shown to be closely linked to grain dormancy in the doubled haploid wheat population SW95-50213 × Cunningham and was co-located with Xgwm269 in population Janz × AUS1408. This marker offers an additional efficient tool for marker-assisted selection of dormancy for white-grained wheat breeding. Comparative analysis indicated that the wheat chromosome 4AL QTL for seed dormancy and PHS resistance is homologous with the barley QTL on chromosome 5HL controlling seed dormancy and PHS resistance. Wheat production and productivity in the past witnessed a remarkable growth. However, this growth rate could not be sustained during the last decade, causing concern among world wheat community. Marker-assisted selection (MAS), which is being practiced for improvement of a variety of traits in wheat around the world, may at least partly help in providing the desired solution. Marker-trait associations are now known for a number of simple, but difficult-to-score traits, so that MAS has been found useful for improvement of several of these important economic traits. Breeding strategies including marker-assisted backcrossing, forward breeding, MAS involving doubled haploid technology and F2 enrichment have been successfully utilized for this purpose. However, for improvement of complex polygenic traits, newer technologies based on high throughput genotyping associated with new marker systems (e.g., DArT and SNP), and new selection strategies such as AB-QTL, mapping-as-you-go, marker-assisted recurrent selection and genome-wide selection will have to be tried in future. Im Gegensatz zu den meisten somatischen Zellen sind Tumorzellen durch Expression und Aktivität des Enzyms Telomerase charakterisiert. Somit scheint dieses Enzym, das zur Stabilisierung der Telomere beiträgt, als klinisch-diagnostischer Tumormarker prädestiniert. Darüber hinaus wurde für verschiedene Tumorentitäten ein Zusammenhang zwischen Enzymaktivität und klinischer Prognose gezeigt. Unklar ist bislang, inwiefern Telomerase, Telomere und die katalytische Untereinheit der Telomerase (hTERT) als diagnostische bzw. prognostische Marker bei kindlichen Weichteilsarkomen (WTS) und akuter lymphoblastischer Leukämie der T-Zell-Linie (T-ALL) im Kindesalter geeignet sind. Aufgrund dessen wurden im ersten Teil der Arbeit Telomeraseaktivität, Telomerlänge und hTERT-Expression anhand von WTS-Zelllinien und Blasten von T-ALL-Patienten analysiert. Weder bei den WTS noch bei den leukämischen Zellen bestand ein Zusammenhang zwischen Telomerlänge bzw. hTERT-Expression und klinischer Prognose bzw. klinischem Verlauf. Während bei der T-ALL keine Assoziation zwischen Enzymaktivität und dem Verlauf der Erkrankung bestand, zeigte sich bei den WTS ein Zusammenhang zwischen klinischer Prognose und Telomeraseaktivität. Resistance to powdery mildew races 1, 2 and 5 has been evaluated in F2/F3 families from a cross between the resistant line ‘TGR-1551’ and the susceptible Spanish cultivar ‘Bola de oro’. Resistance is controlled by two independent genes, one dominant and one recessive, that both confer resistance to races 1, 2 and 5 of Podosphaera xanthii. Bulked segregant analysis (BSA) and amplified fragment length polymorphism (AFLP) technique have been used for the identification of markers linked to powdery mildew resistance. The AFLP marker E38M82 has been converted into a derived cleaved amplified polymorphic sequences (dCAPS) marker. Genes that encode eukaryotic initiation factors (eIF) 4E and iso(4E) have been associated with the recessively inherited resistance to potyviruses in a number of plant species. Using previously developed degenerate primers, partial eIF4E and eIF(iso)4E gene sequence regions were obtained through polymerase chain reaction of the genomic DNA templates prepared from the Zucchini yellow mosaic virus (ZYMV)-resistant U.S. Plant Introduction (PI) 595203, and the ZYMV-susceptible ‘New Hampshire Midget’ (NHM). A single nucleotide polymorphism (SNP) was identified in the intron II region of the eIF4E sequence between the resistant and susceptible parental genotypes. F2 and BC1 populations were developed using PI 595203 and NHM and each plant was phenotyped for resistance to ZYMV and resistance was found to be conferred by a single recessive gene. A codominant Cleaved Amplified Polymorphic Sequence (CAPS) marker was created using the identified SNP in the eIF4E gene. This marker was linked to the ZYMV resistance locus (zym) at a linkage distance of 7 cM as estimated with the F2 and BC1 populations. The SCAR (Sequence Characterized Amplified Region) 'Sw-421' molecular marker is located at 1.0 cM from the Sw-5 allele, originated from Lycopersicon peruvianum (L.), which confers resistance to the tomato spotted wilt virus (TSWV). The goal of this study was to distinguish resistant homozygotes (Sw-5/Sw-5) and heterozygotes (Sw-5/Sw-5+) from susceptible (Sw-5+/Sw-5+) plants in crossing populations with the Stevens cultivar and advanced backcrossing populations by using 'Sw421' SCAR marker. The amplification of 940 bp and 900 bp bands characterized the resistant homozygotes and susceptible controls, respectively. A two band pattern (900 bp and 940 bp) was observed in heterozygote genotypes (Sw-5/Sw-5+), which confirmed the co-dominant inheritance mechanism of the marker. Fifty seven plants from the isogenic progenies were characterized based on bands pattern: 18 plants (31.6%) were identified as resistant homozygotes, 8 plants (14.0%) as resistant heterozygotes and 31 plants (54.4%) were characterized as susceptible. The SCAR 'Sw-421' marker is an important tool for selection and pyramid resistance alleles, mainly when other resistance sources to the TSWV are available...

Chris Marker é um cineasta que realizou documentários políticos pouco convencionais ao longo de quase sessenta anos de carreira. Depois de uma militância ativa e inserida no coração de acontecimentos cruciais na França e na Europa, e de uma carreira literária como editor, ensaísta e romancista, natural que seus filmes, vistos isoladamente, provoquem as mais equivocadas interpretações. Esta pesquisa procura, através de um levantamento de seus filmes e escritos, dos debates que foram se travando sobre a vida nacional francesa e mundial, sempre retomando os fios dos acontecimentos decisivos do período do Front Populaire (1936), da ocupação da França pelos nazistas e das lutas de Resistência buscar uma interpretação para muitos dos aspectos aparentemente desconcertantes de sua filmografia. The penetration of aqueous6Li+ markers into low energy ion irradiated polyimide (PI) foils was examined by the neutron depth profiling technique in combination with a modified tomographic approach. The ion irradiation always leads to an enhancement in marker uptake. After irradiation at low fluence the marker profiles follow the nuclear damage distribution even in three dimensions. At elevated fluences saturation in the marker uptake is clearly seen. The polymer's penetrant uptake can be described well by regular diffusion, with nuclear damage centres acting as saturable traps. These observations are strikingly different from the marker penetration into high-energy heavy-ion irradiated PI.

O marcador molecular SCAR 'Sw-421' é localizado a 1,0 cM do alelo Sw-5, proveniente de Lycopersicon peruvianum (L.), que confere resistência ao vira-çabeça em tomateiro. Contudo, o mesmo ainda não havia sido testado em populações avançadas de tomateiro. O objetivo do trabalho foi distinguir plantas resistentes homozigotas (Sw-5/Sw-5), resistentes heterozigotas (Sw-5/Sw-5+) e suscetíveis (Sw-5+/Sw-5+) pelo marcador SCAR 'Sw-421'. As amplificações de bandas de 940 pb e 900 pb caracterizaram os genótipos resistentes homozigotos e suscetíveis, respectivamente. Duas bandas (900 pb e 940 pb) foram observadas nos genótipos heterozigotos, confirmando a herança codominante do marcador. De 57 plantas das progênies isogênicas avaliadas 18 (31,6%) plantas foram caracterizadas como resistentes, 8 (14,0%) como heterozigotas e 31 (54,4%) plantas suscetíveis. Marker chromosomes are very rare in Klinefelter patients and phenotypic findings are related to the affected chromosomal region. The phenotypic effects of small supernumerary marker chromosomes (sSMC) range from multiple malformations/mental retardation to no effect (ie a normal phenotype). This wide spectrum of phenotypes is due to the origin, structure and gene content of the marker chromosome. The first Klinefelter case with sSMC 9 was published by Liehr et al in 2005. The present case was referred for chromosomal analysis because of dysmorphic features, speech delay and mild mental retardation. Conventional cytogenetic analysis revealed the 47 XXY karyotype in 17 metaphases and the 48 XXY + marker karyotype in eight metaphases. Fluorescence in situ hybridization (FISH) analysis to identify the marker chromosome was performed using the LSI p16 (9p21) Spectrum Orange/CEP 9 SpectrumGreen Probe (Vysis CDKN2A/CEP 9 FISH Probe) and partial trisomy 9 mosaicism was confirmed in this patient. To our knowledge, this is the second case of Klinefelter syndrome with a small supernumerary marker chromosome derived from chromosome 9.
