The objective of this study was to evaluate the nutritional traits and in vitro digestibility of silages from different corn cultivars harvested at two cutting heights. It was evaluated 11 cultivars (Dina 766, Dina 657, Dina 1000, P 3021, P 3041, C 805, C 333, AG 5011, FO 01, Dina co 9621 and BR 205) harvest 5 cm above ground (low) and 5 cm below the intersection of the first ear (high). It was used a random block design (three blocks), arranged in a 11 × 2 factorial scheme. Silages from plants harvested at high cutting height presented average content of dry matter significantly superior to silages from plants harvested at low height. Cultivars FO 01, AG 5011, Dina co 9621 and Dina 766 presented greater content of crude protein than cultivars C 805, P 3041 and P 3021, which presented the lowest contents of this nutrient. The raise in the cut height increased in vitro dry matter true digestibility coefficients and in vitro dry matter digestibility of silage evaluated. BACKGROUND: Cellulose and hemicellulose are quantitatively the most important structural carbohydrates present in ruminant diets. Rumen micro-organisms produce enzymes that catalyse their hydrolysis, but the complex network formed by structural carbohydrates and lignin reduces their digestibility and restricts efficient utilisation of feeds by ruminants. This study aimed to produce two enzymatic extracts, apply them in ruminant diets to determine the best levels for ruminal digestibility and evaluate their effects on in vitro digestibility. RESULTS: In experiment 1 a two-stage in vitro technique was used to examine the effects of different enzymatic levels of Aspergillus japonicus and Aspergillus terricola on tropical forages. Enzyme addition had minor effects on corn silage at the highest enzymatic level. In experiment 2 an in vitro gas production (GP) technique was applied to determine apparent in vitro organic matter digestibility and metabolisable energy. The addition of enzymes in GP showed interesting results. Good data were obtained using sugar cane and Tifton-85 hay supplemented with extracts of A. japonicus and A. terricola respectively. CONCLUSION: Overall, the study suggests that addition of crude extracts containing exogenous fibrolytic enzymes to ruminant diets enhances the effective utilisation of ruminant feedstuffs such as forages. O estilo de vida contemporâneo tem aumentado a demanda por produtos alimentícios que proporcionem benefícios à saúde como os alimentos funcionais. Entretanto, a incorporação de diversos compostos bioativos nas matrizes alimentícias é dificultada pela característica hidrofóbica da maioria destes compostos, que também desfavorece a absorção no intestino. Este é o caso de muitos flavonoides como a quercetina, e sistemas de encapsulação como as micropartículas lipídicas sólidas são capazes de diminuir estes problemas. O presente trabalho de Mestrado teve como objetivo avaliar a digestibilidade in vitro de micropartículas lipídicas sólidas produzidas com óleo de babaçu como fonte de triacilglicerois de cadeia média (MCTs) e triestearina, e a bioacessibilidade da quercetina encapsulada nestes sistemas lipídicos. Foram produzidas dispersões variando-se a concentração de tensoativo (de 2,0 a 4,0%) e de óleo de babaçu (1,2, 2,0 e 2,8%). Os sistemas produzidos apresentaram diâmetro médio que variou de 700 nm a 1800 nm para as diferentes formulações e estabilidade físico-química durante 30 dias. Os resultados obtidos nos ensaios de digestibilidade in vitro indicam uma maior tendência de desestabilização dos sistemas produzidos com mais óleo de babaçu na fase gástrica. A germinação das sementes de grão-de-bico foi acompanhada por um período de 6 dias, no qual pequenas variações nos teores de nitrogênio e globulina total foram registradas. A globulina majoritária (tipo 11 S) apresentou maiores variações após o quarto dia de germinação. A natureza e distribuição da fração globulina majoritária isolada na cromatografia em Sepharose CL-6B mostrou pequenas modificações ao final do período de germinação. A eletroforese em gel de poliacrilamida com dodecilssulfato de sódio do pico eluído na cromatografia em Sepharose CL-6B demonstra modificações nas bandas de proteínas entre os pesos moleculares de 20 e 30 kDa e acima de 60 kDa, indicando degradação protéica durante o período. Atividade proteolítica foi detectada na fração albumina da semente que aumentou até o quarto dia, seguido de queda até o sexto dia de germinação, quando da utilização de globulina total isolada da semente e caseína como substratos. Farinha de grão-de-bico, frações albumina e globulina total isoladas não apresentaram aumento na digestibilidade in vitro; entretanto, a fração globulina majoritária isolada foi mais suscetível à hidrólise após germinação.; Chickpea seed germination was carried out over a period of 6 days. Objetivou-se neste estudo avaliar as características nutricionais e a digestibilidade in vitro de silagens de diferentes cultivares de milho colhidas em duas alturas de corte. Foram avaliadas 11 cultivares (Dina 766, Dina 657, Dina 1000, P 3021, P 3041, C 805, C 333, AG 5011, FO 01, Dina co 9621 e BR 205) colhidas 5 cm acima do solo (baixa) e 5 cm abaixo da inserção da primeira espiga (alta). O experimento foi delineado em blocos casualizados (três blocos), arranjados em esquema fatorial 11 × 2. As silagens das plantas colhidas na altura de corte alta apresentaram teor médio de matéria seca significativamente superior às silagens das plantas colhidas na altura de corte baixa. As cultivares FO 01, AG 5011, Dina co 9621 e Dina 766 apresentaram maior teor de proteína bruta, que as cultivares C 805, P 3041 e P 3021, as quais apresentaram os menores teores deste nutriente. A elevação da altura de corte aumentou os coeficientes de digestibilidade verdadeira in vitro da matéria seca e digestibilidade in vitro da matéria seca das silagens avaliadas. O baru (Dipteryx alata Vog.) é uma leguminosa abundante no Cerrado brasileiro, cuja castanha pode ser explorada através do uso sustentável para o aproveitamento das frações proteicas e lipídicas. Este trabalho teve como objetivo estudar as proteínas desta castanha, presentes na farinha desengordurada e no concentrado proteico, quanto as suas propriedades funcionais, ao perfil das frações proteicas e à digestibilidade in vitro. A farinha desengordurada com hexano foi submetida à extração no pH de maior solubilidade das proteínas, obtendo-se o concentrado proteico. O perfil eletroforético das frações proteicas foi avaliado em gel de SDS-PAGE. As propriedades funcionais indicaram a possibilidade de emprego em diversos alimentos, assim como a soja, conferindo capacidade de absorção de água, capacidade de absorção de óleo, propriedades emulsificantes e espumabilidade. As globulinas, seguidas das albuminas, são as frações majoritárias da farinha e do concentrado proteico, respectivamente. A ß-Lactoglubulina (ß-Lg) é uma das proteínas mais antigênicas presente no leite bovino. Tratamentos físicos, químicos ou enzimáticos podem alterar a antigenicidade desta proteína. Em trabalho anterior, verificou-se que o potencial antigênico da ß-Lg é reduzido quando polimerizada pela enzima transglutaminase (TG) na presença de cisteína (Cys). No entanto, o efeito da polimerização sobre o valor nutricional da ß-Lg ainda não é conhecido. O presente estudo teve como objetivo avaliar o efeito da reação de polimerização catalisada pela TG na digestibilidade in vitro e atividade antigênica da ß-Lg antes e após a ação das enzimas gastrintestinais. A ß-Lg polimerizada pela TG (0, 10 ou 25 U g-1), após tratamento térmico ou na presença de agentes redutores Cys (0, 0,1 e 0,25 mol L- 1) ou ditiotreitol (DTT 0,02 mol -1), foi avaliada quanto à digestibilidade in vitro, utilizando as enzimas pepsina e pancreatina. Solid lipid particles have been investigated by food researchers due to their ability to enhance the incorporation and bioavailability of lipophilic bioactives in aqueous formulations. The objectives of this study were to evaluate the physicochemical stability and digestibility of lipid microparticles produced with tristearin and palm kernel oil. The motivation for conducting this study was the fact that mixing lipids can prevent the expulsion of the bioactive from the lipid core and enhance the digestibility of lipid structures. The lipid microparticles containing different palm kernel oil contents were stable after 60 days of storage according to the particle size and zeta potential data. Their calorimetric behavior indicated that they were composed of a very heterogeneous lipid matrix. Lipid microparticles were stable under various conditions of ionic strength, sugar concentration, temperature, and pH. Digestibility assays indicated no differences in the release of free fatty acids, which was approximately 30% in all analises. The in vitro digestibility tests showed that the amount of palm kernel in the particles did not affect the percentage of lipolysis, probably due to the high amount of surfactants used and/or the solid state of the microparticles.

Copyright © Elsevier / INRA, 1996.; In this paper we make the comparison between sources of inocula for an in vitro digestibility method based on that of Tilley and Terry (1963) as modified by Alexander and McGowan (1966). The best results for the prediction of the in vivo digestibility were obtained using the traditional method of rumen liquor from fistulated sheep. However, with the two alternative methods, one using rumen liquor from slaughtered cattle, and another, a sheep faeces suspension as inocula sources, we obtained results significantly correlated with in vivo digestibility at the 5% level, although with lower correlation coefficients. The method that uses rumen liquor from slaughtered cattle is, in our opinion, a valid alternative to the traditional method.

Proteolytic activity was detected in the albumin fraction of the seeds, which increased up to the fourth and then decreased up to the sixth day, when isolated chickpea total globulin and casein were used as substrates. Chickpea flour, isolated albumin and total globulin fractions did not show an increase for in vitro digestibility; however, the isolated major globulin was more susceptible to hydrolysis after germination.

Sapucaia (Lecythis pisonis Camb.) raw nuts collected from Brazil were analyzed to determine the proximate composition, amino acid profile of protein fractions, in vitro protein digestibility and antinutritional factors in order to evaluate their potential as a protein alimentary complement. The nuts contained adequate amounts of essential amino acids, fatty acids and minerals. In the present study, no hemagglutinating or inhibitory activities were observed in any of the samples investigated, indicating low or non-detectable levels of proteinase inhibitors or lectins in the samples. In vitro digestibility of in natura and heated nut globulins by mammalian digestive proteinases was carried out using trypsin + chymotrypsin + peptidase, with resulting mean values of approximately 70.30 and 71.35%, respectively. Taken together, the results suggest that sapucaia nuts may provide a new source of protein to use as a potential nutritional agent.

Baru (Dipteryx alata Vog.) is an abundant legume in the Brazilian Savanna. Its nuts can be exploited sustainably using its protein and lipid fractions. This study aimed to analyze the proteins of the nuts present in the defatted flour and protein concentrate in terms of their functional properties, the profile of their fractions, and the in vitro digestibility. The flour was defatted with hexane and extracted at the pH of higher protein solubility to obtain the protein concentrate. The electrophoretic profile of the protein fractions was evaluated in SDS-PAGE gel. The functional properties of the proteins indicate the possibility of their use in various foods, like soybeans providing water absorption capacity, oil absorption capacity, emulsifying properties, and foamability. Globulins, followed by the albumins, are the major fractions of the flour and protein concentrate, respectively. Digestibility was greater for the concentrate than for the defatted flour.

Solid lipid particles have been investigated by food researchers due to their ability to enhance the incorporation and bioavailability of lipophilic bioactives in aqueous formulations. The lipid microparticles containing different palm kernel oil contents were stable after 60 days of storage according to the particle size and zeta potential data. Their calorimetric behavior indicated that they were composed of a very heterogeneous lipid matrix. Lipid microparticles were stable under various conditions of ionic strength, sugar concentration, temperature, and pH. Digestibility assays indicated no differences in the release of free fatty acids, which was approximately 30% in all analises. The in vitro digestibility tests showed that the amount of palm kernel in the particles did not affect the percentage of lipolysis, probably due to the high amount of surfactants used and/or the solid state of the microparticles.

Cultivars FO 01, AG 5011, Dina co 9621 and Dina 766 presented greater content of crude protein than cultivars C 805, P 3041 and P 3021, which presented the lowest contents of this nutrient. The raise in the cut height increased in vitro dry matter true digestibility coefficients and in vitro dry matter digestibility of silage evaluated. The increase in cut height improved nutritive value of silages by decreasing concentrations of fibrous fractions and increasing in vitro dry matter digestibility.

Effect of crude glycerol on in-vitro ruminal fermentation kinetics

The interest in using crude glycerol in animal feeding has reemerged due to its increasing availability and favorable price resulting from the expansion of biofuel industry. The objective of the present study was to evaluate the effects of substituting corn for crude glycerol at different levels in the diet on ruminal fermentation using in-vitro true digestibility parameters. The experimental treatments consisted of substituting corn for liquid crude glycerol (0; 4; 8 and 12%) in dry matter basis. Diets consisted of 60% alfalfa hay and 40% corn and glycerol substituted the corn in the diet. In addition to the 48 hours traditionally applied in digestibility assays, different in-vitro digestibility times were used (0; 4; 8; 16; 48, 72 and 96 hours) in order to study digestion kinetics. The dietary corn substitution for increasing crude glycerol levels did not affect ammonia nitrogen content, metabolizable energy content, in-vitro digestibility of organic matter and neutral detergent fiber, nor ruminal degradation parameters. However this by-product of biodiesel production may be tested in-vivo as an alternative energy feedstuff in ruminant diets.

Em uma area de caatinga do Sub-Medio Sao Francisco, em Petrolina, PE, onde predomina a vegetacao arbustiva-arborea, estao sendo coletadas, em varias epocas do ano, amostras de folhas e ramos finos (simulndo o animal em pastejo) de algumas especies forrageiras, conhecidas como as mais consumidas pelos animais, para determinacao do valor nutritivo. Estas amostras tem sido submetidas a estudos de composicao quimica e digestiblidade in vitro da materia seca (DIVMS) pelo laboratorio do CPATSA. Analises realizadas nas amostras coletadas em 14.05.81, quando as plantas ainda estavam em crescimento, revelaram que estas forrageiras apresentaram elavador niveis de proteina bruta, variando desde 14,86% e 15,35% para moleque duro (Cordia leucocephala) e carqueija (calliandra depauperata) ate 20,,88%; 21,10% e 28,47% para manicoba (Manihoti pseudoglaziovii), mororo (Bauhinia cheilantha) e sete-cascas (Tabebuia spongiosa), respectivamente. 8 pages, 4 tables.-- Available Online Mar 2005.; This study was implemented to evaluate the potential of near-infrared reflectance spectroscopy (NIRS) technology to estimate the chemical composition and in vitro digestibility of botanically complex herbage mixtures characterised, moreover, by a noteworthy variation among samples in the maturity of the forage plants. A total of 107 herbage samples harvested from permanent meadows located in the uplands of Leon (northwestern Spain) were analysed to determine their chemical composition. In addition, the in vitro digestibility of each herbage sample was measured by two different in vitro procedures using buffered rumen fluid. A Bran + Luebbe InfraAlyzer 500 spectrophotometer was used to obtain the near-infrared spectra corresponding to each herbage sample. Prediction equations developed for the estimation of the chemical components showed that NIRS technology could predict these parameters accurately, especially the crude protein and neutral detergent fibre contents (R-adj(2) > 0.95 in both cases). In vitro digestibility parameters could also be predicted with an acceptable degree of accuracy using NIRS technology, particularly the in vitro Tilley and Terry organic matter digestibility (R-adj(2) = 0.925...

The purpose of this study was to produce protein hydrolysates from lactic acid fermentation of three sources of fish wastes: Shrimp by catch (SC), Sphyraena ensis wastes (SB) and mixture of fisheries processing wastes from several species (MixW). MixW were added with several sugar cane molasses concentrations as the carbon source, 180 g.kg-1 of sugar molasses gave the fastest acidification. The maximum concentration of lactic acid (Pmax) was significantly higher with Lactobacillus sp. B2 than that obtained with Lb. plantarum. MixW was selected for scaling up and inoculated with Lactobacillus sp. B2 due to the enhanced lactic acid production and availability. According to microbiological and chemical analyses, the fermented product was well preserved due to the acid produced and the reduction in aW (0.94) that inhibit spoilage microorganisms and putrefaction. The coefficient of protein hydrolysis at 144 h of fermentation was significantly higher (0.94) than the obtained with raw MixW (0.12). The coefficient of protein in vitro digestibility (CPD) was also determined, which was higher, 0.88, than raw MixW (0.69).

Variety effect on composition, kinetics of fermentation and in vitro digestibility of oat (Avena sativa L.) straw and its neutral detergent fibre

Yield, chemical composition, in vitro digestibility and kinetics of fermentation of straw from 18 varieties of oats (Avena sativa L.) were studied. All the straw varieties were grown in three replicates under the same agronomic conditions. Significance differences were observed in the yield of straw (4.4 to 7.5 ton dry matter (DM)/ha) from different varieties. The proportion of seed/straw from these varieties varied from 0.28 to 1.02. Crude protein (CP), neutral detergent fibre (NDF), acid detergent fibre (ADF) and acid detergent lignin (ADL) content varied from 24.2 to 48.1, 626 to 708, 437 to 533 and 52.0 to 92.4 g/kg DM, respectively in the straws. In vitro organic matter digestibility (IVOMD) differed among varieties and varied from 400 to 539 g/kg DM. The mean value of digestible organic matter yield (DOM) was 2.34 ton/ha. A significant difference was observed in the potential gas production (A) and lag time (L) among varieties. The fractional rate of gas production (c, /h) ranged from 0.030 to 0.034. The results emphasized that in any evaluation of oat varieties, kinetics of digestion or fermentation should be taken into consideration as well as yield and digestibility.
