OBJETIVO: Analisar as percepções acerca da atividade profissional de um grupo de manipuladores de alimentos de um hospital público. MÉTODOS: A pesquisa, de cunho qualitativo, foi realizada em um serviço de nutrição e dietética de um hospital público de ensino da Cidade de São Paulo. Desenvolveu-se a partir de corpus obtido por técnica do grupo focal realizada com 15 funcionárias, sendo 8 vinculadas ao lactário e 7 à cozinha dietética. Os depoimentos foram gravados, transcritos e analisados sob o ponto de vista do conteúdo e das representações sociais. Posteriormente, a validação da análise foi realizada com as participantes. A caracterização socioeconômica foi realizada com uso de um formulário auto-aplicável. RESULTADOS: O trabalho é percebido como crucial para a sobrevivência, porém de grande responsabilidade social, além de implicar em rotina exaustiva. Além disso, constitui instância que possibilita apoio e relacionamento entre as trabalhadoras, embora não lhes proporcione o status que julgam merecer. O trabalho é representado como instância simultânea de sofrimento e apoio; embora importante, tanto para elas mesmas, quanto para o grupo social. PO artigo demonstra as interações entre algumas bases de dados internacionais na disponibilização de periódicos da área de Ciência da Informação. Para isso, foram investigadas seis bases de dados com acesso a texto completo dentro da área de Ciência da Informação. Foram elas: Web of Science, Scopus, Library, Information Science & Technology Abstracts, Information Science & Technology Abstracts, Library Literature Information Science – full text e Library Information Science Abstract. No desenvolvimento da pesquisa foram utilizados conceitos, princípios e equações da ‘teoria ingênua dos conjuntos’, o que permitiu observar as bases de dados como conjuntos compostos por elementos, aqui considerados os títulos de periódicos, bem como conferir consistência aos resultados. A análise dos dados foi dividida em três etapas, a saber: a) a primeira priorizou o cruzamento entre as duas maiores bases de dados utilizadas para levantamento bibliográfico (Web of Science e Scopus), seguido do cruzamento com a mais utilizada na Ciência da Informação, a Library and Information Science Abstracts; b) a segunda etapa tratou somente do cruzamento das bases especialistas da área; c) a terceira e última etapa tratou da interseção de todas as bases de dados do estudo. No Brasil, como em diferentes partes do mundo, observa-se que a epidemia da aids atinge de forma crescente o segmento feminino, apesar das ações para deter sua disseminação. Uma das limitações das intervenções de controle foi a utilização do conceito de risco para nortear as ações preventivas à transmissão do Human Immunodeficiency Virus (HIV), uma vez que não incorporavam a possibilidade de resposta social no seu enfrentamento e eram direcionadas para uma parcela da população, excluindo outros grupos sociais, gerando estigma e discriminação. O conceito de vulnerabilidade surgiu posteriormente ao conceito de risco, tendo como pressuposto que todas as pessoas são vulneráveis ao HIV e que a infecção é determinada por três planos interdependentes - dimensão individual, social e programática. Este trabalho teve como objetivo identificar os elementos da vulnerabilidade feminina ao HIV/AIDS, por meio de uma revisão sistemática da literatura denominada metassíntese. Recent data demonstrate that the magnitude of the heat loss that occurs from the respiratory tract during exercise correlates with the degree of post-exertional obstruction that develops in asthmatics. Respiratory heat loss relates directly to the minute ventilation and heat capacity of the inspired gas and inversely to its water content and temperature. Because it has been shown that inhaling 100% oxygen during exercise blunts the obstructive response, we wondered if this effect could be accounted for by differing values of heat exchange with air and oxygen breathing. To examine this question, we studied 10 asthmatics by measuring multiple aspects of pulmonary mechanics before and after four bouts of exhausting leg work during which the subjects inhaled either air or oxygen conditioned to provide widely differing thermal burdens on their airways. Under all inspired gas conditions, oxygen breathing produced significantly less obstruction than air. Minute ventilation was also significantly less with oxygen as was the total heat lost. Two studies were carried out to test the hypothesis that the fall and recovery of nasal resistance after exercise in asthmatic and non-asthmatic subjects are related to the development of bronchoconstriction after exercise. In study 1 nasal resistance (posterior rhinomanometry) and specific airway resistance (sRaw) were measured before challenge and one, five, 10 and 30 minutes after four minutes of exhausting legwork exercise in nine asthmatic subjects and nine age matched healthy subjects. One minute after exercise there was a reduction in nasal resistance of 49% (SD 15%) from baseline in the healthy subjects and of 66% (17%) in the asthmatic subjects. This response and the subsequent return of nasal resistance to baseline values did not differ significantly between the two groups despite a substantial difference in the change in sRaw, an increase of 74% (45%) in the asthmatic subjects 10 minutes after exercise, and no change in the non-asthmatic subjects. To produce a flat retina after an operation demands an unrestricted view of the retina during the operation, and to achieve such a view most retinal surgeons would unhesitatingly recommend the binocular indirect ophthalmoscope and scleral depression. Once the retina and all its breaks are clearly in view, the keynote of the ensuing surgery should be simplicity and safety. The former requires that the intended operation should be the least complicated available, and, should the surgical intention be frustrated, that each step should blend with the next in orderly progression, the surgeon exhausting the possibility of one step before moving on to the next and being able to recognise when the possibilities are exhausted. The latter requires experience in deciding which is in fact the least complicated operation. In general the cavity of the globe should not be entered unless the eye stands to lose more than it gains by remaining inviolate. Paracentesis, fluid release, and intravitreal air all have their place, and to avoid them gains us nothing if the retina remains detached.

Adult stem cells in various tissues are relatively quiescent. The cell cycle inhibitor p21cip1/waf1 (p21) has been shown to be important for maintaining hematopoietic stem cell quiescence and self-renewal. We examined the role of p21 in the regulation of adult mammalian forebrain neural stem cells (NSCs). We found that p21-/- mice between post-natal age 60-240 d have more NSCs than wild-type (+/+) controls due to higher proliferation rates of p21-/- NSCs. Thereafter, NSCs in p21-/- mice decline and are reduced in number at 16 mo relative to p21+/+ mice. Similarly, both p21-/- and p21+/+ NSCs display self-renewal in vitro; however, p21-/- NSCs display limited in vitro self-renewal (surviving a few passages, then exhausting). Thus, p21 contributes to adult NSC relative quiescence, which we propose is necessary for the life-long maintenance of NSC self-renewal because NSCs may be limited to a finite number of divisions.

1. Exercise seems to change the extracellular potassium concentration far beyond the narrow limits seen in resting subjects. To examine alterations in plasma potassium concentration during exercise, twenty healthy, well-trained men ran on the treadmill at 6 deg inclination with catheters inserted in the femoral vein and artery. 2. During 1 min exhausting exercise plasma potassium concentration rose in parallel in the vein and artery, reaching peak post-exercise values of 8.34 +/- 0.23 mmol l-1 and 8.17 +/- 0.29 mmol l-1. After 3 min recovery the potassium concentration was 0.50 +/- 0.05 mmol l-1 below pre-exercise values. Both the rise of plasma potassium concentration during exercise and the decline during recovery followed exponential time courses with a half-time of 25 s. 3. Exercise at reduced intensity showed that the peak post-exercise potassium concentration was linearly related to the exercise intensity. Individual resting, peak and nadir values were proportionally related. 4. The increased potassium concentration during exercise can be explained in full by the electrical activity in the exercising muscles. Repeated 1 min exhausting exercise bouts revealed no relationship between potassium concentration and plasma pH nor glycogen break-down. 5. OBJECTIVE: To compare the maximum energy cost of running (MECR) estimated from the speed of a mile run and the maximum oxygen consumption (VO2max) with a field step test. METHODS: MECR was defined as the maximum VO2, derived from the equation VO2 = 5.3 mph + 3.9 (ml.min-1.kg-1), when the subject performs a maximal, exhausting run and attains maximum speed. The equation is based on a straight line relation between speed and VO2. In this study MECR was obtained from a mile run and was compared with VO2max by extrapolation using a step test. Both tests were performed in the field. RESULTS AND CONCLUSIONS: The variation in the MECR with VO2max was within +/- 10%. It therefore appears that the speed of a mile run is a good criterion of VO2max or estimated MECR for the measurement of physical fitness, and for the selection, recruitment, and assignment of an individual in field sports.

HIV infection selectively targets CD4+ effector memory T (TEM) cells, resulting in dramatic depletion of CD4+ T cells in mucosal effector sites in early infection. Regeneration of the TEM cell compartment is slow and incomplete, even when viral replication is controlled by antiretroviral therapy (ART). Here, we demonstrate that IL-15 dramatically increases in vivo proliferation of rhesus macaque (RM) CD4+ and CD8+ TEM cells with little effect on the naive or central memory T (TCM) cell subsets, a response pattern that is quite distinct from that of either IL-2 or IL-7. TEM cells produced in response to IL-15 did not accumulate in blood. Rather, 5-bromo-2′-deoxyuridine (BrdU) labeling studies suggest that many of these cells rapidly disperse to extralymphoid effector sites, where they manifest (slow) decay kinetics indistinguishable from that of untreated controls. In RMs with uncontrolled SIV infection and highly activated immune systems, IL-15 did not significantly increase CD4+ TEM cell proliferation, but with virologic control and concomitant reduction in immune activation by ART, IL-15 responsiveness was again observed. These data suggest that therapeutic use of IL-15 in the setting of ART might facilitate specific restoration of the CD4+ T cell compartment that is the primary target of HIV with less risk of exhausting precursor T cell compartments or generating potentially deleterious regulatory subsets.

The influence of a regimen designed to lower the muscle glycogen content on the capacity to perform a single brief isometric contraction has been studied. Eight male subjects performed a single exhausting isometric contraction of the knee extensor muscles at a tension corresponding to 60% of maximum voluntary contraction (MVC). This was followed by prolonged cycling exercise at a work rate equivalent to approximately 75% of maximum oxygen uptake in order to reduce the muscle glycogen content. A diet low in carbohydrate was consumed for the remainder of this day in order to retard the resynthesis of muscle glycogen. The isometric contraction at 60% of MVC was repeated on the following day. Endurance time on the first day was 53.8 +/- 8.4 s (mean +/- SD); this was reduced (45.8 +/- 12.1 s; p less than 0.02) on the second day. From previously published data on rates of muscle glycogen utilisation during isometric exercise, it seems probable that insufficient glycogen is available in the muscle under the low carbohydrate condition to enable maximum performance to be achieved.

In rice (Oryza sativa), the presence of a dominant Badh2 allele encoding betaine aldehyde dehydrogenase (BADH2) inhibits the synthesis of 2-acetyl-1-pyrroline (2AP), a potent flavor component in rice fragrance. By contrast, its two recessive alleles, badh2-E2 and badh2-E7, induce 2AP formation. Badh2 was found to be transcribed in all tissues tested except for roots, and the transcript was detected at higher abundance in young, healthy leaves than in other tissues. Multiple Badh2 transcript lengths were detected, and the complete, full-length Badh2 transcript was much less abundant than partial Badh2 transcripts. 2AP levels were significantly reduced in cauliflower mosaic virus 35S-driven transgenic lines expressing the complete, but not the partial, Badh2 coding sequences. In accordance, the intact, full-length BADH2 protein (503 residues) appeared exclusively in nonfragrant transgenic lines and rice varieties. These results indicate that the full-length BADH2 protein encoded by Badh2 renders rice nonfragrant by inhibiting 2AP biosynthesis. The BADH2 enzyme was predicted to contain three domains: NAD binding, substrate binding, and oligomerization domains. Event Related Potentials Reveal that Increasing Perceptual Load Leads to Increased Responses for Target Stimuli and Decreased Responses for Irrelevant Stimuli

Lavie (1995) have suggested that perceptual processing is influenced by perceptual load. Specifically, relevant information receives additional processing in high load situations exhausting the available capacity. On the other hand, irrelevant information receives less processing with increasing load on a relevant task, as there is a reduced amount of residual processing available. Rees et al. (1997) provided the first physiological evidence for this model, showing this pattern in a functional magnetic resonance imaging study. Likewise, Handy et al. (2001) offered supporting evidence measuring event related potentials (ERPs). Both of these studies presented irrelevant information in peripheral vision. Here we manipulated load while using the identical stimuli and the same task (a peripheral gap judgment task) with centrally presented irrelevant stimuli. ERPs show the pattern predicted by Lavie and colleagues, specifically for the N1 component. This work offers further evidence that visual attention modulates relatively early processing of perceptual information. Specifically, increasing load resulted in stronger N1 responses to relevant information and weaker N1 responses to irrelevant information.

This study aims to assess the effects of Achyranthes bidentata polysaccharides (ABPS) on the expression of bcl-2 and bax in hepatic tissues after exhaustive exercise in order to provide theoretical support for the application of ABPS in the field of sports nutrition. Thirty male Sprague-Dawley rats were randomized into three groups, each consisting of 10 rats: Normal control group (NCG), Exhausting exercises control group (EECG), ABPS treated group (ATG). ABPS were fed orally by gastric intubation to rats of ABPS treated group (ATG) once daily for 7 days. Control animals (EECG and NCG) received the same amount of isotonic sodium chloride solution. Exhaustive exercise was performed on a rodent treadmill. The SP (streptavidin peroxidase) method for immunohistochemical staining was adopted to test the protein expression of bax and bcl-2 in the hepatic tissues of the rats. Exhausting exercises increased bax protein expression of hepatic tissues of rats and bax/bcl-2 ratio dramatically, but a decreased bcl-2 protein expression. In the rats fed ABPS orally by gastric intubation, the bax protein expression and bax/bcl-2 ratio obviously decreased, while bcl-2 protein expression increased. The result indicated that bax and bcl-2 co-regulated the exercise-induced hepatocyte apoptosis. Feeding ABPS orally by gastric intubation to rats can inhibit the hepatocyte apoptosis in exhaustive exercise.

Whole exome sequencing has become a pivotal methodology for rapid and cost-effective detection of pathogenic variations in Mendelian disorders. A major challenge of this approach is determining the causative mutation from a substantial number of bystander variations that do not play any role in the disease etiology. Current strategies to analyze variations have mainly relied on genetic and functional arguments such as mode of inheritance, conservation, and loss of function prediction. Here, we demonstrate that disease-network analysis provides an additional layer of information to stratify variations even in the presence of incomplete sequencing coverage, a known limitation of exome sequencing. We studied a case of Hereditary Spastic Paraparesis (HSP) in a single inbred Palestinian family. HSP is a group of neuropathological disorders that are characterized by abnormal gait and spasticity of the lower limbs. Forty-five loci have been associated with HSP and lesions in 20 genes have been documented to induce the disorder. We used whole exome sequencing and homozygosity mapping to create a list of possible candidates. After exhausting the genetic and functional arguments, we stratified the remaining candidates according to their similarity to the previously known disease genes. The purpose of the study was to investigate the modulatory effect of changes of subthalamic nucleus (STN) activity on the development of central fatigue during exhausting exercise, and reveal the possible mechanism that might affect STN activity from the perspective of neurotransmitters. Rats were randomly divided into electrophysiology and microdialysis study groups. For electrophysiological study, electrical activity in sensorimotor cortex and STN were simultaneously recorded before, during and 90min after the exhausting exercise. For microdialysis study, extracellular fluid of STN was continuously collected with a microdialysis probe and glutamate (Glu), gamma-aminobutyric acid (GABA) levels were subsequently detected with high performance liquid chromatography (HPLC). The behavioral studies showed that rats ran well initiatively with the treadmill exercise in the beginning, 45 ± 11.5min later, movement capacity reduced obviously (which was termed as ‘early fatigue’). Correspondingly, STN activity increased significantly compared with rest condition (p < 0.05), while, cortex activity decreased significantly (p < 0.05). Myosin light chain 2 (MLC2) is a small protein in the myosin complex, regulating muscle contractile function by modulating Ca2+ sensitivity of myofilaments. MLC2 can be modified by phosphorylation and O-GlcNAcylation, two reversible and dynamic posttranslational modifications. The slow isoform of MLC2 (sMLC2) is dephosphorylated in soleus muscle during in situ loaded shortening contractions, which correlates with reduction in shortening capacity. Here, we hypothesize that exhausting in vivo treadmill running induces dephosphorylation of MLC2 in slow twitch soleus, but not in fast twitch EDL muscle, and that there are reciprocal changes in MLC2 O-GlcNAcylation. At rest, both phosphorylation and O-GlcNAcylation of MLC2 were lower in slow than fast twitch muscles. One bout of exhausting treadmill running induced dephosphorylation of sMLC2 in soleus, paralleled by reduced levels of the kinase MLCK2 associated to myofilaments, suggesting that the acute reduction in phosphorylation is mediated by dissociation of MLCK2 from myofilaments. O-GlcNAcylation of MLC2 did not change significantly, and seems of limited importance in the regulation of MLC2 phosphorylation during in vivo running. Stress kann in verschiedenen Varianten auftreten, und sich sehr unterschiedlich auf den Organismus auswirken. Im Rahmen dieses Forschungsprojektes wurde ein freiwillig auferlegter physischer Stress in Form von Hochleistungssport untersucht. Bei hohen Trainings- oder Wettkampfbelastungen werden Stressantworten im Immunsystem des Sportlers mobilisiert, die dann sekundäre Auswirkungen auf somatische Parameter zeigen. Diese Auswirkungen können zu Veränderungen in der zellulären Zusammensetzung im peripheren Blut führen. Des Weiteren, kann man eine Veränderung auf Genexpressionsebene beobachten. Im Rahmen der Dissertationsarbeit wurden home-made cDNA Microarrays hergestellt und etabliert. Die Herstellung und Etablierung sind im Abschnitt Methode dieser Arbeit ausführlich beschrieben. In dieser Studie wurde mit home-made cDNA Microarrays die Genexpression bei acht Halbmarathonläufern untersucht. Der eingesetzte cDNA Microarray enthielt ausgewählte Gene des Stress- und Entzündungsgeschehen. Knockdown of BACE1-AS Nonprotein-Coding Transcript Modulates Beta-Amyloid-Related Hippocampal Neurogenesis

Background. Alzheimer's disease (AD) is a devastating neurological disorder and the main cause of dementia in the elderly population worldwide. Adult neurogenesis appears to be upregulated very early in AD pathogenesis in response to some specific aggregates of beta-amyloid (Aβ) peptides, exhausting the neuronal stem cell pools in the brain. Previously, we characterized a conserved nonprotein-coding antisense transcript for β-secretase-1 (BACE1), a critical enzyme in AD pathophysiology. We showed that the BACE1-antisense transcript (BACE1-AS) is markedly upregulated in brain samples from AD patients and promotes the stability of the (sense) BACE1 transcript. In the current paper, we examine the relationship between BACE1, BACE1-AS, adult neurogenesis markers, and amyloid plaque formation in amyloid precursor protein (APP) transgenic mice (Tg-19959) of various ages. Results. Consistent with previous publications in other APP overexpressing mouse models, we found adult neurogenesis markers to be noticeably upregulated in Tg-19959 mice very early in the development of the disease. Knockdown of either one of BACE1 or BACE1-AS transcripts by continuous infusion of locked nucleic acid- (LNA-) modified siRNAs into the third ventricle over the period of two weeks caused concordant downregulation of both transcripts in Tg-19959 mice. É cada vez mais freqüente a condução de pesquisas científicas orientadas por avaliações qualitativas: pesquisas qualitativas, como são geralmente denominadas. A estratégia de pesquisa discutida neste texto - Estudo de Caso - pede avaliação qualitativa, pois seu objetivo é o estudo de uma unidade social que se analisa profunda e intensamente. Busca-se apreender a totalidade de uma situação e, criativamente, descrever, compreender e interpretar a complexidade de um caso concreto, mediante um mergulho profundo e exaustivo em um objeto delimitado. Estudos epistemológicos têm mostrado que um grande número de pesquisas orientadas por Estudo de Caso apresenta sérias deficiências: análises intuitivas, primitivas e impressionistas, não conseguindo transcenderem a simples relatos históricos, obviamente muito afastados do que se espera de um trabalho científico. São diversas as fontes de erros que comprometem um Estudo de Caso.
