Physiological changes induced by the aging process is dynamic and progressive, reducing the adaptability and independence of older people and may be influenced by genetic and environmental factors. Thus the aim of this thesis was to investigate the association between polymorphism of the ACE gene ID and the phenotypes of muscular strength and blood pressure of 62 elderly Brazilian (67.35 ± 5.66 years) during a 16-week program of supervised training. The elderly women were stratified by age, with the group 1 (G1, n = 34) <70 years and group 2 (G2 n = 28) ≥ 70 years, and in three groups by ACE, ACE-II (n = 8) ACE- DD (n = 35) and ACE-ID (n = 19). The level of muscle strength was evaluated by the method of maximum repetitions and measures of blood pressure (BP) were measured before and after training (PAPré1 and PAPós1) and before and after each training session (PAPre2 and PAPós2), in place of training. DNA samples were isolated from peripheral blood leukocytes polymorphism and insertion / deletion (ID) of the ACE gene (rs1800795) was genotyped by polymerase chain reaction (PCR) plus PCR-confirmatory. The genotype distribution of the polymorphism ID attended the prerogatives of Hardy-Weitíherg. There was variation in power levels before and after training and the age between groups (t-test) and the ACE polymorphism (ANOVA) (p <0.05). Depending on the results it was concluded that resistance training helps to reduce SBP and increased muscle strength of upper and lower limbs when considering the age and ACE polymorphism. The venlafaxine, 1-[2-dimethylamino-1-(4-methoxyphenyl)-ethyl] cyclohexanol, is a antidepressant drug of second generation. This is one of the most potent reuptake inhibitor of serotonin and noradrenaline, and its therapeutic effect is attributed to this activity. Venlafaxine is biotransformed in the liver to O-desmethylvenlafaxine (desvenlafaxine), N, O-desmethylvenlafaxine, N-desmethylvenlafaxine. Enzymes CYP2D6, CYP2C19 and CYP2C9 metabolize venlafaxine and major metabolite is the O-desmethylvenlafaxine. This is pharmacologically active and contributes significantly to the pharmacological effect of venlafaxine, as is found in plasma at high concentrations. The investigation of the formation of degradation products is of great importance, because the products formed may be less active, more active or toxic. Among manufacturing processes available, direct compression is indicated for OLZ because this drug is moisture sensitive. Before drug developement, preformulation investigations involving physical and chemical properties of drug and excipient combined and separated must be performed. Thus, drug solubility profile, thermal analysis techniques, mainly DSC and TG, and X rays graphics are used to verify drug behavior. The objective of this work was preformulation study of OLZ, drug development and characterization of OLZ tablets to accomplish pharmaceutical equivalence. DSC and TG tests were performed, as well as X rays diffraction and solubility test in different media. Media used in solubility assay were: water; HCl 0,1 mol/L; and USP buffers pH 2.5; pH 4.5; pH 6.8 and pH 7.4. This test duration was 48h. OLZ showed to be more soluble in acidic pH, HCl medium, in which it reached the concentration of 20,62 mg/mL, without saturation. In buffers pH 2.5 and 4.5 saturation solubility was 2,06 mg/mL and 5,19 mg/mL, respectively; in pH 6.8 and pH 7.4 OLZ reached only 221,45 μg/mL and 79,06 μg/mL, respectively. Progesterone is a steroid hormone used to treat uterin disfunction, such as hemorrhage and secondary amenorrhea, contraception, hormone replacement therapy in postmenopausal women, as well as in luteal phase support cases in assisted reproduction techniques. However, it presents low oral bioavailability because of its weak water solubility and intense hepatic metabolism. So, progesterone administration is carried out, mainly, through micronized drug dispersed in oils and surfactant formulations, in soft gelatin capsules, trying to improve oral bioavailability. However, there are no official compendia notifications about dissolution methods used to evaluate progesterone release from these dosage forms. Thus, this study proposes progesterone containing soft gelatin capsules dissolution method development and validation aiming to analyze two pharmaceutical specialties (called A and B) in vitro dissolution profile commercially available. The Brazilian Savanna is recognized as the richest in the world, sheltering more than 6.500 plant species already cataloged, and medicinal uses are attributed to over 220. Species of Myrtaceae family are used in folk medicine in gastrointestinal disorders, infectious diseases and hemorrhagic states, many are known by the presence of essential oils. The objective of this work was the pharmacognostic study of leaves and bark of species Myrcia tomentosa (Aubl.) DC. in order to establish quality parameters and to determine the chemical composition of essential oil (leaves, bark and flowers), as well as to assess the occurrence of seasonal variation between the chemical constituents of essential oil from leaves and to evaluate the antimicrobial activity of leaf essential oil, ethanolic crude extract and fractions from leaf and bark extracts. The Pterodon emarginatus Vogel, known as sucupira branca (Fabaceae) has been shown to possess larvicidal activity against (Aedes aegypti), antiparasitic effect (Leishmania amazonensis), fungicide, bactericide, antioxidant, antiinflammatory, antinociceptive, and antiproliferative activity among several different human cancer cell lines that may be related to presence of diterpenes, in particular, vouacapanes derivatives. This work aimed to define and characterize the powder, the crude and standardized dry extract of the fruit of sucupira . After crushing the fruits, showed 1.2% volatile. The chromatographic (TLC) indicated the presence of chemical markers vouacapanes (6α,7β-dihydroxyvouacapan-17β-oic acid - Rf = 0.16; 6α,7β-dihydroxyvouacapan-17β-oate methyl ester - Rf = 0.44; 6α-hydroxyvouacapan-7β-17β-lactone - Rf = 0.71) and lupeol (Rf = 0.78). In vibration bands in the infrared region coinciding with the five main bands of vouacapanes (-OH, C-H , C=O, C=C, C-O). The concentration of total terpenes of 9.3%. Then to obtain the crude extract was held percolation in 95% ethanol PA. The crude extract showed pH 5.1, relative density of 0.87, viscosity of 25.43 mPas, solid 15.2%, total terpenes of 17.1% and the same chromatographic profile and in the infrared spectrum obtained for the powder plant. In this work, standardized spray-dried extracts of Rosmarinus officinalis L.(Lamiaceae), were obtained. The work started with the obtainment and characterization of the R. officinalis dried and powder leaves. A hydroalcoholic extract was obtained by percolation using as solvent ethanol at 80 % (v/v). The drying experiments followed a Box-Behnken (33) factorial design in order to obtain dry powders with optimized properties. In this way, the influence of several in-process parameters e. g. extract feed rate, drying air inlet temperature and spray nozzle airflow rate, on the dried extracts properties were investigated. The herbal raw material as well as both hydroalcoholic and dried extracts were analyzed by several physic-chemical and physical techniques. Additionally, the in vitro antioxidant activity from both hydroalcoholic and dried extracts were assessed by DPPH free radical scavenging test. The study of bioconversion of biologically active compounds by microorganisms, mainly filamentous fungi and their possible glycosylated derivatives have been extensively explored and have brought promising results. Microbial models of animal metabolism, based on the similarity of the hepatic metabolism and microbial enzyme, became an alternative for the elucidation of metabolic routes of both drugs as well as new drug candidates. In this context, the objective of this study was to explore the ability of filamentous fungi to promote the bioconversion of entacapone and perform the identification, purification and characterization of derivatives formed. We selected eight filamentous fungi Absidia blakesleana ATCC 26617, Aspergillus candidus ATCC 1009, Beauveria bassiana ATCC 7159, Beauveria sp, Cunninghamella echinulata ATCC 9244, Cunninghamella echinulata ATCC 9245, Cunninghamella elegans ATCC 26169 and Rhizopus arrhizuz ATCC 11145, six of them proved to be effective in the metabolism of entacapone, Cunninghamella echinulata ATCC 9245 being the most promising. Palicourea coriacea (Cham.) K. Schum, known as douradinha or douradinha-docampo, is a plant of Cerrado in the form of sub-shrubs with yellow flowers, belonging to the Rubiaceae family. Its leaves are used in preparations of home remedies in the form of teas, to treat kidney stones, inflammation / infection of the urethra, kidneys, bladder, and as a diuretic. This study aimed to evaluate the acute toxicity in rats and mice, as well as the diuretic activity in rats of the crude aqueous extract of dried leaves Palicourea coriacea (EABLPC). The acute oral toxicity EABLPC was investigated according to OECD 423. To evaluate the diuretic activity three groups of rats received doses of EABLPC (80, 160 and 320 mg / kg), one group received only saline solution, and another received furosemide (20 mg / kg) administered by gavage for eight days, being made volume measurement and evaluation of urinary concentrations of electrolytes (Na+, K+ and Cl-), creatinine and urea in urine and blood. Vernonanthura ferruginea (Less.) H. Rob. (Asteraceae), a typical plant of the Brazilian savannah, popularly known as assa-peixe, presented analgesic, antiedematogenic, antioxidant and antiulcer activities in scientific studies. But in literature there are no works related to the production of dried extracts of this plant species. In this work, standardized spray-dried extracts of V. ferruginea were obtained. The work started with the obtainment and characterization powder of the leaves of the V. ferruginea. A hydroalcoholic extract was obtained by percolation using as solvent ethanol 95 % (v/v). The drying experiments followed a 23 + 2 factorial design. The influence of several in-process parameters e. g. extract feed rate, drying air inlet temperature and drying adjuvant ratio, on the dried extracts properties were investigated. Thromboembolic disorders produce the majority of diseases affecting the heart, and are the third leading cause of death worldwide. Venous thromboembolism is a thromboembolic disorder that occurs in the venous circulation, is a disease of high prevalence and remains a major cause of morbidity and mortality in hospitalized patients undergoing major surgery, the fact that affect patients from the very young, disease free prior to the more elderly mean that it is a problem with major socioeconomic burden by as much as by absenteeism in hospital patients in productive age. The present work aimed to do the study of chemical variability, evaluating biological activities and microencapsulate the essential oil of the fruits of Pterodon emarginatus Vogel (Fabaceae). Chapter 1 presents the study of the chemical variability of essential oil from fruits of sucupira of 11 individual of the Brazilian savanna, collected from five different populations, and were identified as its chemical composition by gas chromatography coupled to mass spectrometry (GC/MS), used for data analysis to multivariate statistical tool that indicated compounds β caryophyllene and α copaene as the main discriminant of the samples studied, both of which are of greater significance. Chapter 2 describes the antimicrobial activity and chemical composition of total oil of the fruits of sucupira, four samples being analyzed, three of the city of Jussara GO and one of region of Jaciara MT. Na busca por novos agentes antineoplásicos os produtos de origem natural têm um papel de destaque, uma vez que cerca de 25% dos fármacos prescritos no mundo são obtidos de plantas e aproximadamente 49% dos fármacos desenvolvidos nas últimas três década foram obtidos a partir de produtos naturais. Eugenia dysenterica DC. pertence à família Myrtaceae e é conhecida p pula mente c m cagaita , sendo empiricamente utilizada no Brasil para o tratamento de distúrbios gastrointestinais e diabetes. Neste trabalho foram investigados a citotoxicidade e os mecanismos de indução de apoptose do extrato padronizado hidroalcóolico das folhas da E. dysenterica DC. sobre células leucêmicas HL60 e K562, atividade antitumoral in vivo em camundongos com tumor ascítico de Ehrlich (TAE), bem como a atuação do extrato frente a células basais, como fibroblastos e eritrócitos, além do perfil de toxicidade oral aguda em camundongos. Os estudos de citotoxicidade foram realizados por meio dos ensaios de exclusão do azul de tripano e redução do tetrazólio (MTT). Os mecanismos de indução de apoptose celular foram investigados por microscopia de luz e de fluorescência e por citometria de fluxo. A investigação do potencial antitumoral in vivo foi feito através da curva de sobrevida utilizando camundongos portadores do TAE tratados com diferentes doses do extrato padronizado hidroalcóolico das folhas da E. dysenterica DC. A bioconversão utiliza o arsenal enzimático presente em sistemas biológicos para modificar quimicamente compostos orgânicos. Este trabalho descreve a utilização de cepas do fungo filamentoso Beauveria bassiana spp. na produção de derivados da naringenina, flavonóide de elevado interesse farmacoterapêutico, e a avaliação das suas potenciais atividades vasodilatadora e antiproliferativa. Realizou-se uma seleção que incluiu, além da cepa de Beauveria bassiana ATCC 7149, diversas cepas de Beauveria bassiana spp. isoladas de amostras de solo do cerrado brasileiro e denominadas: IP3a, IP6, IP8, IP11, IP94, IP98, IP127, IP132, IP147 e IP153. A cepa da coleção ATCC mostrou melhor perfil de produção de derivados e foi utilizada para ensaio em escala semipreparativa. O meio de cultura líquido Peptone Dextrose Soy Meal (PDSM) foi utilizado para a bioconversão de naringenina na concentração de 0,5 mg/mL, a 27ºC e 200 r.p.m. Após 96 horas da adição do substrato, o micélio foi removido por filtração, o sobrenadante extraído com acetato de etila e o micélio com acetona. O uso de plantas medicinais tem sido muito significativo nos últimos anos, sendo incentivado pela Organização Mundial de Saúde (OMS). Synadenium umbellatum Pax, Euphorbiacea (vulgo cola-nota, cancerola, milagrosa) tem o látex usado empiricamente como anti-tumoral e antiinflamatório. Por existir espécies tóxicas nesta família e visando à segurança no uso de extratos vegetais, tal estudo avaliou a toxicidade pré-clinica do látex e do extrato etanólico das folhas (EEF) de S. umbellatum, por via oral, em ratas Wistar. O estudo seguiu diretrizes da OECD (Organisation for Economic Cooperation and Development) para teste de toxicidade aguda (Guideline 423) e de toxicidade subaguda (Guideline 407). Na toxicidade aguda do látex e do EEF, não se observou, na dose de 2000 mg/kg, alterações fisiológicas e comportamentais, e nem a morte dos animais. No entanto, o látex ocasionou congestão e infiltrado leucocitário nos rins, fígado e pulmões, efeitos não observados com o EEF. Na toxicidade subaguda, doses de 50, 100 e 200 mg/kg de EEF não produziram alterações dose-dependentes significativas nos parâmetros laboratoriais e nem alterações fisiológicas, macroscópicas e histopatológicas dos órgãos. The use of medicinal plants in controlling the diseases has shown its potential for the discovery of new medicines. Among the plant species used in the Hospital de Medicina Alternativa (HMA / SES-GO) highlights the vetiver (Vetiveria zizanioides L. Nash), an aromatic plant of the family Poaceae, used as a mild anti-hypertensive and diuretic. In this work it was investigated the chemical composition of the hydroalcoholic extract of vetiver, the freeze-dried aqueous extract and the essential oil from the roots of V. zizanioides. In the hydroalcoholic extract were found terpenoids compounds and / or saponins, flavonoids and phenolic acid. In the freeze-dried aqueous extract was observed the presence of flavonoids and phenols. In the essential oil 38 compounds were identified, and khusimol, palustrol, khusimona, zizanona and epi-α-vetivona were the major constituents. In the assessment of the acute toxicity of the hydroalcoholic extract and the freeze-dried aqueous extract of root of V. zizanioides, it was concluded that these extracts showed no toxicity at a dose of 2000 mg / kg. The Rudgea viburnoides (Cham.) Benth. (Rubiaceae), popularly known as "congonha, congonha-de-Bugre or Bugre" is used in folk medicine as diuretic, hypotensive, anti-rheumatic and depurative of blood (leaves tea). This species is being traded as "porangaba" and used in weight loss programs. This work aimed to study the acute toxicity, the diuretic and antioxidant activities of R. viburnoides. The test for acute toxicity in single dose was performed with the ethanolic crude extract, administered orally, at concentrations of 2000mg/kg and 5000mg/kg in mice and rats of both sexes by the testo of Class.To test for diuretic activity in rats, it was used the crude ethanolic extract of the leaves at concentrations of 40mg/kg, 80mg/kg and 160mg/kg, and furosemide 20mg/kg as positive control. The determination of total phenols of the crude extract and fractions (hexane, dichloromethane, ethyl acetate and methanol/water) was performed by the method of Butler and Hargerman and the evaluation of antioxidant activity performed by the method of 2,2-diphenyl-1- picryl -hidrazil (DPPH). Tyrosinase (E.C.1.14.18.1) is an enzyme of industrial interest that catalyses the ohydroxylation of monophenols (monophenolase activity) and the oxidation of o-diphenols to reactive o-quinones (diphenolase activity), both reactions using molecular oxygen. Pycnoporus sanguineus (L. ex Fries) Murril, is a white rot fungi capable of producing tyrosinase and widely distributed in nature. It is found in regions of mild climate and in tropical forest. The production and characterization of tyrosinase from P. sanguineus were investigated. The selection of inductors, determination of the luminosity influence, biomass and culture media in the production of tyrosinase and the effect of inhibitors on enzyme activity were determined. The fungus produced intracellular tyrosinase and the higher activity was observed using 0.15% L-tyrosine as inducer, in the presence of light, with inoculum of 10 mycelium discs, medium malt extract broth 2%, incubation at 30°C, and constant agitation of 150 rpm, during 2 days. 6 mmol.L-1 salicylhydroxamic acid (SHAM) and 6 mmol.L-1 phenylthiourea (PTU) inhibited 100% of the tyrosinase activity. 0.1 mmol.L-1 sodium azide inhibited 4.15% of tyrosinase activity, while no inhibition was observed after addition of 0.1 mmol.L-1 of phenylmethanesulfonyl fluoride (PMSF). Before the approval of an active compound, metabolism studies are necessary to ensure its safety, once active metabolites could be synthesized during human biotransformation. The use of eukaryotic microorganisms for the study of drug metabolism has been widely explored, due to its capability of producing metabolites similar to the mammalians, and in silico studies consist in a fast strategy when compared with traditional metabolism studies. In this context, molecular modeling, using docking of molecules of interest in the active site of enzymes involved in drug metabolism, is a useful tool to evaluate the interactions between drug and receptor, because it could predict favorable orientations that could be biotransformated. In this work, sixteen filamentous fungi strains, obtained from collections and isolated from soil in the central Brazil, were evaluated for their capability of the antiretroviral stavudine biotransformation, also complemented by animal metabolism studies and molecular modeling of the most relevant cytochrome P450 isoform of human metabolism: CYP3A4. In search of new antineoplasic agents natural products have played an important role, given that about 40% of available medicines in current treatments have been developed from natural sources, 25% approximately of plants. Synadenium umbellatum belongs to Euphorbiaceae family and is popularly known as "cola-nota", "avelós," "milagrosa", "cancerola", and the latex is empirically used in Brazil for the treatment of cancer. Preliminary studies have shown that the crude extract of the leaves of S. umbellatum has significant antitumor and antiangiogenic activity in vivo. In this study, we investigated the cytotoxicity and the mechanisms of apoptosis induction of S. umbellatum in Ehrlich ascitic tumor (EAT) cells, and the myelotoxic potential of this plant. The cytotoxicity studies were carried out by trypan blue exclusion and reduction of tetrazolium (MTT) tests. The mechanisms of apoptosis induction were investigated by light and fluorescence microscopy, immunocytochemistry and flow cytometry. Investigation of the myelotoxic potential was performed by clonogenic assay of colony forming units of granulocyte-macrophage (CFU-GM). The data were analyzed by analysis of variance (ANOVA) and a posteriori Tukey test (myelotoxicity) and by t test for all other assays. The means were considered statistically significant when P < 0.05.
