O objectivo deste trabalho é de enquadrar a crise de 68 em França no seu contexto épocal, genérico e especificamente francês. Analisam-se assim resumidamente os principais acontecimentos políticos da década, bem com é feita uma panorâmica da principal produção intelectual relevante. Em seguida a uma breve descrição da sucessão dos acontecimentos procede-se a uma análise dos diversos tipos de explicações fornecidas para justificar os factos ocorridos.

Ventilation distribution can be assessed by SPECT with Technegas. This study was undertaken in piglets with different degrees of ventilation inhomogeneity to compare PET using (68)Ga-labeled pseudogas or ""Gallgas"" with Technegas. Methods: Twelve piglets were studied in 3 groups: control, lobar obstruction, and diffuse airway obstruction. Two more piglets were assessed for lung volume (functional residual capacity). Results: In controls, SPECT and PET images showed an even distribution of radioactivity. With lobar obstruction, the absence of ventilation of the obstructed lobe was visible with both techniques. In diffuse airway obstruction, SPECT images showed an even distribution of radioactivity, and PET images showed more varied radioactivity over the lung. Conclusion: PET provides detailed ventilation distribution images and a better appreciation of ventilation heterogeneity. O 68Ga é um radionuclídeo promissor para a medicina nuclear, decaindo por emissão de pósitrons com abundância de 89%, apresentando tempo de meia vida física de 68 minutos, o que é compatível com a farmacocinética de muitas biomoléculas e substratos de baixo peso molecular. Outra característica importante é a sua disponibilidade por um sistema de gerador, onde o radionuclídeo pai, 68Ge (t1/2 = 270,95 dias) é adsorvido em uma coluna e o filho, 68Ga, é eluído na forma iônica 68Ga3+. O desenvolvimento dos geradores de 68Ge/68Ga teve início na década dos anos 60, mas o seu uso clínico começou a ser aceitável e relevante apenas recentemente. O método de separação do 68Ge do 68Ga mais utilizado é o sistema cromatográfico de troca iônica, devido sua praticidade de operação, mas outros sistemas de geradores já foram propostos, como de extração por solvente e técnica de evaporação. Atualmente, os geradores de 68Ge/68Ga são disponíveis comercialmente tanto com colunas utilizando matrizes inorgânicas preparadas com TiO2 ou SnO2 como também usando resina orgânica. The measurement by serological means of the impact of the Hong Kong/68 influenza virus on a population

In order to obtain evidence on the size of the impact of the Hong Kong/68 variant of influenza A2 virus on the population of São Paulo, Brazil, serum samples taken in 1967 before this variant appeared and during successive years after it appeared were examined for their antibody content. Haemagglutination-inhibition tests performed on a total of 2726 serum samples from adults showed a sharp decrease in 1969 of the proportion of sera without antibody to the Hong Kong/68 variant and a corresponding mercase in the proportion with high titres. It was concluded that about three-quarters of the adult population became infected at some time after the variant appeared, the majority in the first year of prevalence.

Gammaherpesviruses cause important infections of humans, in particular in immunocompromised patients. Recently, murine gammaherpesvirus 68 (MHV-68) infection of mice has been developed as a small animal model of gammaherpesvirus pathogenesis. Efficient generation of mutants of MHV-68 would significantly contribute to the understanding of viral gene functions in virus-host interaction, thereby further enhancing the potential of this model. To this end, we cloned the MHV-68 genome as a bacterial artificial chromosome (BAC) in Escherichia coli. During propagation in E. coli, spontaneous recombination events within the internal and terminal repeats of the cloned MHV-68 genome, affecting the copy number of the repeats, were occasionally observed. The gene for the green fluorescent protein was incorporated into the cloned BAC for identification of infected cells. BAC vector sequences were flanked by loxP sites to allow the excision of these sequences using recombinase Cre and to allow the generation of recombinant viruses with wild-type genome properties. Murine gammaherpesvirus 68 (gamma HV-68; also referred to as MHV-68) is a gammaherpesvirus which infects murid rodents. Previous studies showed that CD8 T cells are important for controlling gamma HV-68 replication during the first 2 weeks of infection and suggested a role for B cells in latent or persistent gamma HV-68 infection. To further define the importance of B cells and CD8 T cells during acute and chronic gamma HV-68 infection, we examined splenic infection in mice with null mutations in the transmembrane domain of the mu-heavy-chain constant region (MuMT; B-cell and antibody deficient) or in the beta2-microglobulin gene (beta2 -/-; CD8 deficient). Immunocompetent mice infected intraperitoneally with gamma HV-68 demonstrated peak splenic titers 9 to 10 days postinfection, cleared infectious virus 15 to 20 days postinfection, and harbored low levels of latent virus at 6 weeks postinfection. Beta2-/- mice showed peak splenic gamma HV-68 titers similar to those of normal mice but were unable to clear infectious virus completely from the spleen, demonstrating persistent infectious virus 6 weeks postinfection. These data indicate that CD8 T cells are important for clearing infectious gamma HV-68 from the spleen. The murine gammaherpesvirus 68 (MHV-68 or γHV-68) model provides many advantages for studying virus-host interactions involved in gammaherpesvirus replication, including the role of cellular responses to infection. We examined the effects of cellular cyclooxygenase-2 (COX-2) and its by-product prostaglandin E2 (PGE2) on MHV-68 gene expression and protein production following de novo infection of cultured cells. Western blot analyses revealed an induction of COX-2 protein in MHV-68-infected cells but not in cells infected with UV-irradiated MHV-68. Luciferase reporter assays demonstrated activation of the COX-2 promoter during MHV-68 replication. Two nonsteroidal anti-inflammatory drugs, a COX-2-specific inhibitor (NS-398) and a COX-1-COX-2 inhibitor (indomethacin), substantially reduced MHV-68 protein production in infected cells. Inhibition of viral protein expression and virion production by NS-398 was reversed in the presence of exogenous PGE2. Global gene expression analysis using an MHV-68 DNA array showed that PGE2 increased production of multiple viral gene products, and NS-398 inhibited production of many of the same genes. These studies suggest that COX-2 activity and PGE2 production may play significant roles during MHV-68 de novo infection.

How cellular factors regulate gammaherpesvirus lytic replication is not well understood. Here, through functional screening of a cellular kinase expression library, we identified mitogen-activated protein kinase kinase kinase 8 (MAP3K8/Tpl2) as a positive regulator of murine gammaherpesvirus 68 (MHV-68 or γHV-68) lytic gene expression and replication. Tpl2 enhances MHV-68 lytic replication by upregulating lytic gene expression and promoter activities of viral lytic genes, including RTA and open reading frame 57 (ORF57). By screening a cellular transcription factor library, we identified the Fos AP-1 transcription factor as a downstream factor that is both necessary and sufficient for mediating the enhancement of MHV-68 lytic replication by Tpl2. In addition, Tpl2 stimulates the promoter activities of key viral lytic genes, including RTA and ORF57, in an AP-1-dependent manner. We identified an AP-1-responsive element on the MHV-68 RTA promoter as the cis element mediating the upregulation of RTA promoter activity by Tpl2. Pluronic F-68, an 80% hydrophilic member of the Pluronic family of polyethylene-polypropylene-polyethylene tri-block copolymers, protects non-neuronal cells from traumatic injuries and rescues hippocampal neurons from excitotoxic and oxidative insults. F-68 interacts directly with lipid membranes and restores membrane function after direct membrane damage. Here, we demonstrate the efficacy of Pluronic F-68 in rescuing rat hippocampal neurons from apoptosis after oxygen–glucose deprivation (OGD). OGD progressively decreased neuronal survival over 48 h in a severity-dependent manner, the majority of cell death occurring after 12 h after OGD. Administration of F-68 for 48 h after OGD rescued neurons from death in a dose-dependent manner. At its optimal concentration (30 μm), F-68 rescued all neurons that would have died after the first hour after OGD. This level of rescue persisted when F-68 administration was delayed 12 h after OGD. Numerous studies have confirmed that Pluronic F-68 can effectively protect mammalian cell cultures grown in intensely agitated bioreactors from the damaging effects of hydrodynamic shear. Like mammalian cells, plant cells exposed to intense shear conditions are also severely damaged. However, the possibility of shear-protection for plant cells by Pluronic F-68 (PF-68) is seldom addressed. This work addressed this deficiency and investigated the shear-protective properties of PF-68 on wild carrot cultures (Daucus carota), using a two-level, three-factor (23) factorial design. The aim of the experiments was to determine whether PF-68 could provide similar protection for plant cells compared to animal cells, using a novel shearing apparatus [Chem. Eng. Res. Des. 79 (2001) A8]. The three factors involved in the study were impeller speed (300, 400 rpm), exposure time (3, 4 h) and the concentration of the proposed shear-protective agent—PF-68 (0.05, 0.1%, w/v). Herpesviruses are characterized as having two distinct life cycle phases: lytic replication and latency. The mechanisms of latency establishment and maintenance, as well as the switch from latency to lytic replication, are poorly understood. Human gammaherpesviruses, including Epstein-Barr virus (EBV) and human herpesvirus-8 (HHV-8), also known as Kaposi's sarcoma-associated herpesvirus (KSHV), are associated with lymphoproliferative diseases and several human tumors. Unfortunately, the lack of cell lines to support efficient de novo productive infection and restricted host ranges of EBV and HHV-8 make it difficult to explore certain important biological questions. Murine gammaherpesvirus 68 (MHV-68, or γHV68) can establish de novo lytic infection in a variety of cell lines and is also able to infect laboratory mice, offering an ideal model with which to study various aspects of gammaherpesvirus infection. Here we describe in vitro studies of the mechanisms of the switch from latency to lytic replication of MHV-68. An MHV-68 gene, rta (replication and transcription activator), encoded primarily by open reading frame 50 (ORF50), is homologous to the rta genes of other gammaherpesviruses, including HHV-8 and EBV. HHV-8 and EBV Rta have been shown to play central roles in viral reactivation from latency. We first studied the kinetics of MHV-68 rta gene transcription during de novo lytic infection. Murine gammaherpesvirus 68 (MHV-68 [also referred to as γHV68]) is phylogenetically related to Kaposi's sarcoma-associated herpesvirus (KSHV [also referred to as HHV-8]) and Epstein-Barr virus (EBV). However, unlike KSHV or EBV, MHV-68 readily infects fibroblast and epithelial cell lines derived from several mammalian species, providing a system to study productive and latent infections as well as reactivation of gammaherpesviruses in vivo and in vitro. To carry out rapid genome-wide analysis of MHV-68 gene expression, we made DNA arrays containing nearly all of the known and predicted open reading frames (ORFs) of the virus. RNA obtained from an MHV-68 latently infected cell line, from cells lytically infected with MHV-68 in culture, and from the lung tissue of infected mice was used to probe the MHV-68 arrays. Using a tightly latent B-cell line (S11E), the MHV-68 latent transcription program was quantitatively described. Using BHK-21 cells and infected mice, we demonstrated that latent genes are transcribed during lytic replication and are relatively independent of de novo protein synthesis. Murine gammaherpesvirus 68 (MHV-68) is closely related to Epstein-Barr virus (EBV) and Kaposi's sarcoma-associated herpesvirus (KSHV) and provides a small-animal model with which to study the pathogenesis of gammaherpesvirus (γHV) infections. To completely explore the potential of the MHV-68 system for the investigation of γHV microRNAs (miRNAs), it would be desirable to know the number and expression patterns of all miRNAs encoded by MHV-68. By deep sequencing of small RNAs, we systematically investigated the expression profiles of MHV-68 miRNAs in both lytically and persistently infected cells. In addition to the nine known MHV-68 miRNAs, we identified six novel MHV-68 miRNA genes and analyzed the expression levels of all MHV-68 miRNAs. Furthermore, we also characterized the cellular miRNA expression signatures in MHV-68-infected versus noninfected NIH 3T3 fibroblasts and in 12-O-tetradecanoyl-phorbol-13-acetate (TPA)-treated versus nontreated S11 cells. We found that mmu-mir-15b and mmu-mir-16 are highly upregulated upon MHV-68 infection of NIH 3T3 cells, indicating a potential role for cellular miRNAs during MHV-68 infection. Objetivou-se evidenciar a intensidade da ação da variante Hong Kong/68 do virus da influenza A2 sobre a população de São Paulo, Brasil. Para tanto, foram coletados soros em 1967, antes do aparecimento dessa variante e durante anos seguintes à sua presença, para investigação de seu conteúdo em anticorpos. As provas de inibição da hemaglutinação, levadas a efeito em 2726 amostras de adultos, mostraram acentuada declínio em 1969, relativo à proporção de soros sem anticorpos para Hong Kong/68 e correspondente aumento na proporção com títulos elevados. 68, this happenning sprouted simultaneously in many countries with singular features, and it was remarked by the forgetfulness and the normalization of the society and politics which followed its irruption. It comes bach through celebration, in regular intervals, which are only defined by decimal criterion. Every time that the celebration of the year of 1968 occurs, it empties the dramatization which coverd its owm occurence in 1968, and what is actually celebrated are the ideological appropriations of the happening and the aspects which are assimilated in the present.; 68, acontecimento que irrompeu simultaneamente em vários países, com contornos singulares, foi marcado pelo esquecimento e pela normalização da sociedade e da política que se seguiram à sua irrupção. Retorna pela via da comemoração, em intervalos regulares de tempo, definidos pelo único critério da numeração decimal. Os eventos comemorativos de 68, a cada vez que se repetem, esvaziam o acontecimento da dramaticidade que o revestiu na sua própria atualidade, e o que se comemora então são as apropriações ideológicas do acontecimento, os seus aspectos assimiláveis pelo presente.

May 1968 left remarks and references in many insuspicious areas of human knoledge. In this text we try to explore the relationships wich can be established within its statements, especially those conected to a sexual free set of ideas, and how those themes are dealt by the cinema. Films such as Blow Up, Clockwork Orange, Death in Venice, Last tango in Paris, In the Realm of the Senses and Salo are also analised.; Maio de 68 deixou marcas e referências nos mais insuspeitos campos de atividade e de conhecimento humano. Neste texto, buscamos explorar as relações que podem ser estabelecidas entre algumas de suas formulações – em especial as ligadas a um ideário de liberação e libertação de práticas sexuais – e os temas correlatos, bem como a forma de tratá-los, que invadiram o cinema dito "comercial" na década seguinte. Sob este prisma são analisados filmes como Blow Up, Laranja Mecânica, Morte em Veneza, Último Tango em Paris, Império dos Sentidos e Saló.

In order to obtain evidence on the size of the impact of the Hong Kong/68 variant of influenza A2 virus on the population of São Paulo, Brazil, serum samples taken in 1967 before this variant appeared and during successive years after it appeared were examined for their antibody content. Haemagglutination-inhibition tests performed on a total of 2726 serum samples from adults showed a sharp decrease in 1969 of the proportion of sera without antibody to the Hong Kong/68 variant and a corresponding mercase in the proportion with high titres. It was concluded that about three-quarters of the adult population became infected at some time after the variant appeared, the majority in the first year of prevalence.
