As temperaturas de transformações de fases são essenciais para trabalhar e caracterizar as ligas de NiTi, podendo-se assim, aproveitar de maneira eficiente suas propriedade de memória de forma e superelasticidade. O objetivo deste trabalho foi caracterizar as temperaturas de transformações de fases (As, Af, Ms, Mf, Rs, Rf) e a superfície de chapas e fios quanto a sua morfologia e a molhabilidade da liga NiTi submetidos a diferentes tratamentos térmicos para aplicação como órtese coronária. Tratamentos térmicos influenciam diretamente nas temperaturas de transformações de fases e na superfície do material, portanto os fios e chapas foram submetidos a diferentes tratamentos térmicos com diferentes tempos e temperaturas adquirindo diferentes colorações. Os fios foram submetidos a tratamentos térmicos de têmpera e temperaturas de 530 e 570°C, sendo analisados por DSC (Differencial Scanning Calorimeter), microscopia óptica e microdureza. Quando comparados com o fio sem tratamentos térmico mostraram mudanças nas temperaturas de transformações de fases. O fio com tratamento térmico de 570ºC apresentou as melhores temperaturas para aplicação como material biomédico. Constituem uma fração de variação genética importante, que interagem, modulam ou direcionam a expressão gênica. MicroRNA são pequenos RNA de aproximadamente 20 nucleotídeos que regulam a expressão gênica pós-transcricional. O objetivo desta Tese foi analisar a expressão de microRNA com relação às variações estruturais do número de cópias dos respectivos genes em humanos, as regiões de CNV-miRNA. Foram selecionadas 11 regiões de provável polimorfismo no número de cópias além de duas regiões invariáveis como controle positivo, que foram avaliadas por triagem molecular em PCR quantitativa em uma amostra de 103 indivíduos da população brasileira. Autophosphorylation of JAK2 on Tyrosines 221 and 570 Regulates Its Activity

The tyrosine kinase JAK2 is a key signaling protein for at least 20 receptors in the cytokine/hematopoietin receptor superfamily and is a component of signaling by insulin receptor and several G-protein-coupled receptors. However, there is only limited knowledge of the physical structure of JAK2 or which of the 49 tyrosines in JAK2 are autophosphorylated. In this study, mass spectrometry and two-dimensional peptide mapping were used to determine that tyrosines 221, 570, and 1007 in JAK2 are autophosphorylated. Phosphorylation of tyrosine 570 is particularly robust. In response to growth hormone, JAK2 was rapidly and transiently phosphorylated at tyrosines 221 and 570, returning to basal levels by 60 min. Analysis of the sequences surrounding tyrosines 221 and 570 in JAK2 and tyrosines in other proteins that are phosphorylated in response to ligands that activate JAK2 suggests that the YXX[L/I/V] motif is one of the motifs recognized by JAK2. Experiments using JAK2 with tyrosines 221 and 570 mutated to phenylalanine suggest that tyrosines 221 and 570 in JAK2 may serve as regulatory sites in JAK2, with phosphorylation of tyrosine 221 increasing kinase activity and phosphorylation of tyrosine 570 decreasing kinase activity and thereby contributing to rapid termination of ligand activation of JAK2.

A peptide corresponding to a surface loop in the C-terminal domain of chicken ovotransferrin (residues 570-584) was made by solid-phase synthesis and used to immunize rabbits. A 15-amino acid-residue disulphide-linked loop occurs in both domains of all five transferrins for which the sequence is available and lies on the opposite side of the iron-binding site from the interdomain cleft. Polyclonal antibodies to the peptide were specific for non-reduced holo-ovotransferrin and the C-terminal domain, as shown by e.l.i.s.a. and immunoblotting. The antibody did not inhibit binding of ovotransferrin to receptors on chick-embryo reticulocytes but was able to bind ovotransferrin bound to the cellular receptors at 0 degree C. The loop composed of residues 570-584 appears to be remote from the transferrin receptor-binding site.

In purple membrane added with general anesthetics, there exists an acid-base equilibrium between two spectral forms of the pigment: bR570 and bR480 (apparent pKa = 7.3). As the purple 570 nm bacteriorhodopsin is reversibly transformed into its red 480 nm form, the proton pumping capability of the pigment reversibly decreases, as indicated by transient proton release measurements and proton translocation action spectra of mixture of both spectral forms. It happens in spite of a complete photochemical activity in bR480 that is mostly characterized by fast deprotonation and slow reprotonation steps and which, under continuous illumination, bleaches with a yield comparable to that of bR570. This modified photochemical activity has a correlated specific photoelectrical counterpart: a faster proton extrusion current and a slower reprotonation current. The relative areas of all photocurrent phases are reduced in bR480, most likely because its photochemistry is accompanied by charge movements for shorter distances than in the native pigment, reflecting a reversible inhibition of the pumping activity.

The picosecond molecular dynamics in an artificial bacteriorhodopsin (BR) pigment containing a structurally modified all-trans retinal chromphore with a six-membered ring bridging the C11=C12-C13 positions (BR6.11) are measured by picosecond transient absorption and picosecond time-resolved fluorescence spectroscopy. Time-dependent intensity and spectral changes in absorption in the 570-650-nm region are monitored for delays as long as 5 ns after the 7-ps, 573-nm excitation of BR6.11. Two intermediates, J6.11 and K6.11/1, both with enhanced absorption to the red (> 600 nm) of the BR6.11 spectrum are observed within approximately 50 ps. The J6.11 intermediate decays with a time constant of 12 +/- 3 ps to form K6.11/1. The K6.11/1 intermediate decays with an approximately 100-ps time constant to form a third intermediate, K6.11/2, which is observed through diminished 650-nm absorption (relative to that of K6.11/1). No other transient absorption changes are found during the remainder of the initial 5-ns period of the BR6.11 photoreaction. Fluorescence in the 650-900-nm region is observed from BR6.11, K6.11/1, and K6.11/2, but no emission assignable to J6.11 is found. The BR6.11 fluroescence spectrum has a approximately 725-nm maximum which is blue-shifted by approximately 15 nm relative to that of native BR-570 and is 4.2 +/- 1.5 times larger in intensity (same sample optical density). The molecular processes comprising the room temperature bacteriorhodopsin (BR) photocycle are examined through the properties of the photo-induced reverse reaction, K-590 + hν → BR-570 (K → BR). Two sequential pumping pulses, each of 10-ns duration, are used, respectively, to initiate the photocycle via the forward BR-570 + hν → K-590 (BR → K) reaction (532 nm) and to photolytically interrupt the thermal BR photocycle after a 20-ns delay via K → BR (620-700 nm). The ground-state BR-570 population, monitored by 633-nm absorption 200 μs after the photocycle begins, provides a quantitative measure of the efficiency with which K → BR interrupts the photocycle to reform BR-570. The quantum yield (Φ) for K → BR is found to be 1.6 ± 0.1 times larger than that for BR → K which, when compared to a Φ of 0.64 for BR → K, suggests that Φ for K → BR is ≈ 1.0. The significance of such a high efficiency K → BR reaction with respect to mechanistic descriptions of the BR photocycle is discussed.

The fluorescence spectrum of a distinct isometric and conformational intermediate formed on the 10(-11) s time scale during the bacteriorhodopsin (BR) photocycle is observed at room temperature using a two laser, pump-probe technique with picosecond time resolution. The BR photocycle is initiated by pulsed (8 ps) excitation at 565 nm, whereas the fluorescence is generated by 4-ps laser pulses at 590 nm. The unstructured fluorescence extends from 650 to 880 nm and appears in the same general spectral region as the fluorescence spectrum assigned to BR-570. The transient fluorescence spectrum can be distinguished from that assigned to BR-570 by a larger emission quantum yield (approximately twice that of BR-570) and by a maximum intensity near 731 nm (shifted 17 nm to higher energy from the maximum of the BR-570 fluorescence spectrum). The fluorescence spectrum of BR-570 only is measured with low energy, picosecond pulsed excitation at 590 nm and is in good agreement with recent data in the literature. The assignment of the transient fluorescence spectrum to the K-590 intermediate is based on its appearance at time delays longer than 40 ps. The K-590 fluorescence spectrum remains unchanged over the entire 40-100-ps interval. The relevance of these fluorescence data with respect to the molecular mechanism used to model the primary processes in the BR photocycle also is discussed.

Angiotensin II (Ang II) is a pleuripotential hormone that is important in the pathophysiology of multiple conditions including aging, cardiovascular and renal diseases, and insulin resistance. Reactive oxygen species (ROS) are important mediators of Ang II-induced signaling generally and have a well defined role in vascular hypertrophy, which is inhibited by overexpression of catalase, inferring a specific role of H2O2. The molecular mechanisms are understood incompletely. The transcriptional coactivator peroxisome proliferator-activated receptor γ coactivator-1α (PGC-1α) is a key regulator of energy metabolism and ROS-scavenging enzymes including catalase. We show that Ang II stimulates Akt-dependent PGC-1α serine 570 phosphorylation, which is required for the binding of the histone acetyltransferase GCN5 (general control nonderepressible 5) to PGC-1α and for its lysine acetylation. These sequential post-translational modifications suppress PGC-1α activity and prevent its binding to the catalase promoter through the forkhead box O1 transcription factor, thus decreasing catalase expression. We demonstrate that overexpression of the phosphorylation-defective mutant PGC-1α (S570A) prevents Ang II-induced increases in H2O2 levels and hypertrophy ([3H]leucine incorporation). We present the genomic sequence of the human pathogen Legionella pneumophila serogroup 12 strain 570-CO-H (ATCC 43290), a clinical isolate from the Colorado Department of Health, Denver, CO. This is the first example of a genome sequence of L. pneumophila from a serogroup other than serogroup 1. We highlight the similarities and differences relative to six genome sequences that have been reported for serogroup 1 strains.

Asthma is a chronic lung disease that affects people of all ages and is characterized by high morbidity. The mechanisms of asthma pathogenesis are unclear, and there is a need for development of diagnostic biomarkers and greater understanding of regulation of inflammatory responses in the lung. Post-transcriptional regulation of cytokines, chemokines, and growth factors by the action of microRNAs and RNA-binding proteins on stability or translation of mature transcripts is emerging as a central means of regulating the inflammatory response. In this study, we demonstrate that miR-570-3p expression is increased with TNFα stimuli in normal human bronchial epithelial cells (2.6 ± 0.6, p = 0.01) and the human airway epithelial cell line A549 (4.6 ± 1.4, p = 0.0068), and evaluate the functional effects of its overexpression on predicted mRNA target genes in transfected A549 cells. MiR-570-3p upregulated numerous cytokines and chemokines (CCL4, CCL5, TNFα, and IL-6) and also enhanced their induction by TNFα. For other cytokines (CCL2 and IL-8), the microRNA exhibited an inhibitory effect to repress their upregulation by TNFα. These effects were mediated by a complex pattern of both direct and indirect regulation of downstream targets by miR-570-3p. Increasing studies have shown that MicroRNAs (miRNAs) play critical roles in the progression of lung carcinoma. In the present study, the expression and functions of miR-570 were investigated. We found that miR-570 was significantly up-regulated in lung cancer tissues, compared with adjacent non-cancerous tissues. In vitro studies further showed that miR-570 mimics could promote, while its antisense oligos inhibit cell proliferation and invasion. At the molecular level, krüppel-like factor 9 (KLF9), a tumor suppressor gene, was identified as a potential target of miR-570 in lung cancer cells. Indeed, miR-570 could negatively regulate protein levels of KLF9 through targeting its 3’-untranslated region. Taken together, our results suggest a previously unknown miR-570/KLF9 molecular network controlling lung carcinoma progression.

There has been relatively little study of system change among scholars of international relations. Several academics have referred to the subject as the “evaded dimension” of IR. The reasons for the omission are complex but include a normative inclination to model stability during the dangerous years of the Cold War and the related preeminence of political realism as the primary theoretical lens of IR analysis. Much of the research work since the Second World War used a technically suitable but distinctly Westphalian template for examining the international system. This approach was perhaps useful for understanding contemporary western IR but may have provided a skewed data set from which scholars could investigate the dynamics of system change. With the Cold War over and the field in the midst of an intellectual expansion, and in the spirit of those scholars who recently called for an IR approach that isn’t “owned and operated by Europe” (referring specifically to Security Communities), this study seeks to suggest an alternerative historical model of system change - one based less upon the explanatory utility of force or interest, but of ideas. The morphologic abnormalities present in 570 endomyocardial biopsies from 39 cardiac allograft recipients and 16 autopsy hearts are described, and criteria pertinent to the diagnosis of rejection discussed. Entirely normal myocardium was apparent in 16% of the biopsies. Abnormalities related to biopsy of previous biopsy site occurred in 69%, and mononuclear infiltrates of varying intensity were present in 64% of the biopsies. Acute rejection was diagnosed in 17 (43.6%) patients in 32 (5.6%) of the biopsies and five hearts at autopsy. Seven of the biopsies with acute rejection were follow-up biopsies after a previous diagnosis of acute rejection and represented ongoing acute rejection. Similarly, three patients with rejection at autopsy died with ongoing rejection. The overall frequency of acute rejection was thus 0.70 episodes per patient. The most reliable histologic feature in the diagnosis of acute rejection in cyclosporine immunosuppressed recipients was a diffuse mononuclear infiltrate, apparent at low magnification. Myocyte necrosis, said to be critical in the diagnosis of rejection in cyclosporine-treated patients, was not a reliable indicator in the authors' experience.

Este ingreso fue conseguido por nuestro país como corolario de varios años de estudios y recomendaciones por parte de esta organización, en los que se ha intentado identificar la situación económica del país, sus virtudes y deficiencias, y las medidas mediante las cuales debería lograrse un crecimiento y desarrollo económico sustentables en el tiempo comparables al nivel de los países que integran la OECD. De esta manera, una de las importantes deficiencias que ha sido detectada en los “Economic Surveys” sobre el país realizados por esta organización, es justamente la que dice relación con el bajo nivel de inversión privada en Investigación y Desarrollo (I+D), el cual se encuentra muy disminuido en comparación con el estándar de gasto en esta materia por parte de los países miembros de la OECD. The analysing power of the reaction ${\rm n}{\rm p} \to {\rm p}{\rm p} \pi^{-}$ for neutron energies between threshold and 570 MeV has been determined using a transversely polarised neutron beam at PSI. The reaction has been studied in a kinematically complete measurement using a time-of-flight spectrometer with large acceptance. Analysing powers have been determined as a function of the c.m. pion angle in different regions of the proton-proton invariant mass. They are compared to other data from the reactions ${\rm n}{\rm p} \to {\rm p}{\rm p} \pi^{-}$ and ${{\rm p}{\rm p} \to {\rm p}{\rm p} \pi^{0}}$. The reaction ${n} {p} \to {p} {p} \pi^{-}$ has been studied in a kinematically complete measurement with a large acceptance time-of-flight spectrometer for incident neutron energies between threshold and 570 MeV. The proton-proton invariant mass distributions show a strong enhancement due to the pp($^{1}{S}_{0}$) final state interaction. A large anisotropy was found in the pion angular distributions in contrast to the reaction ${p}{p} \to {p}{p} \pi^{0}$. At small energies, a large forward/backward asymmetry has been observed. From the measured integrated cross section $\sigma({n}{p} \to {\rm p}{p} \pi^{-})$, the isoscalar cross section $\sigma_{01}$ has been extracted. Its energy dependence indicates that mainly partial waves with Sp final states contribute. Análisis multicaracter para estimar parámetros genéticos para crecimiento en bovinos de raza Braford en Corrientes

Se ha efectuado un estudio en La Estación Experimental Agropecuaria Corrientes del INTA sobre un rodeo Braford estabilizado 3/8 Brahman x 5/8 Hereford, que tiene como propósito estimar los parámetros genéticos y ambientales para peso al nacer (PNAC), peso ajustado a 210 días (P_AJ_210) y peso ajustado a 570 días (P_AJ_570), mediante el Modelo Animal Multicaracter, utilizando el software MTDFREML. Se procesó una base de datos de 658 registros de producción y genealogía de bovinos de raza Braford, correspondientes al período 1992-2004. Los componentes de varianza y covarianza, y parámetros genéticos y ambientales estimados son: a) Para PNAC: s²a = 6,57, s²m = 2,21, s am = -0,32, s²pe = 0,72, s²e = 7,31, s²p = 16,50, h²a = 0,40, h²m = 0,13, r am = -0,09, c² = 0,04, e² = 0,44; b) Para P_AJ_210: s²a = 153,89, s²m = 62,18, s am = -56,47, s²pe = 10,35, s²e = 223,05, s²p = 393,02, h²a = 0,39, h²m = 0,16, r am = -0,58, c² = 0,02, e² = 0,57; c) Para P_AJ_570: s²a = 292,95, s²e = 419,02, s²p = 711,97, h²a = 0,41, e² = 0,59. Además se calcularon las correlaciones genéticas entre los caracteres analizados: PNAC-P_AJ_210: 0,83, PNAC-P_AJ_570: 0,31, P_AJ_210-P_AJ_570: 0,74. El objetivo de este trabajo fue estimar los parámetros genéticos y ambientales de bovinos de raza Hereford en Corrientes, una provincia subtropical del Nordeste de Argentina. Los datos fueron tomados en el INTA EEA Corrientes entre los años 1074 y 2002. Se emplearon 4092 registros para el carácter peso ajustado a 210 días (I_PESOAJ_210) y 3440 registros para el carácter peso ajustado a 570 días (I_PESOAJ_570). Los datos fueron analizados mediante el Modelo Animal para un único carácter, con efectos maternos para I_PESOAJ_210. Para este carácter, el modelo incluye los efectos fijos grupo contemporáneo (año de nacimiento + sexo), mes de nacimiento, edad de la madre, lactancia previa de la madre y edad en días del animal al destete como covariable. Para I_PESOAJ_570 los efectos fijos fueron grupo contemporáneo, mes de nacimiento y edad en días del individuo al peso final como covariable. El método de estimación fue REML, usando el algoritmo libre de derivadas implementado por el paquete MTDFREML. Los efectos aleatorios fueron los valores genéticos aditivos directos y maternos y el ambiente permanente de las madres para I_PESOAJ_210 y los valores genéticos aditivos directos para I_PESOAJ_570.
