New measurements of radial velocities in clusters of galaxies-V

As a part of our galaxy-cluster redshift survey, we present a set of 79 new velocities in the 4 clusters Abell 376, Abell 970, Abell 1356, and Abell 2244, obtained at Haute-Provence observatory. This set now completes our previous analyses, especially for the first two clusters. Data on individual galaxies are presented, and we discuss some cluster properties. For A376, we obtained an improved mean redshift (z) over bar = 0.04750 with a velocity dispersion of sigma(V) = 860 km s(-1). For A970, we have (z) over bar = 0.05875 with sigma(V) = 881 km s(-1). We show that the A1356 cluster is not a member of the ""Leo-Virgo"" supercluster at a mean redshift (z) over bar = 0.112 and should be considered just as a foreground group of galaxies at (z) over bar = 0.0689, as well as A1435 at (z) over bar = 0.062. We obtain (z) over bar = 0.09962 for A2244 with sigma(V) = 965 km s(-1). The relative proximity of clusters A2244 and A2245 ((z) over bar = 0.08738, sigma(V) = 992 km s(-1)) suggests that these could be members of a supercluster that would include A2249; however, from X-ray data there is no indication of interaction between A2244 and A2245.; FAPESP; CNPq

Signal transduction GTP-binding proteins are tightly associated with plasma membrane. In the resting state, the anchorage of the alpha subunit could be indirect by means of the other beta gamma subunits or polydisperse multimers. In the activated state, although the alpha subunit is dissociated from other subunits, it is not released from the membrane and therefore is likely to contain information necessary to remain associated with the plasma membrane. Previous proteolytic experiments suggested that, in contrast to other G proteins alpha subunits, the C-terminal domain of Gs alpha (the G protein involved in adenylate cyclase stimulation) is essential for membrane association of the activated form. To better define the crucial residues involved in membrane attachment, we constructed chimeras between a soluble core and various parts of the Gs alpha C-terminal domain. We first deleted codons 2-6 of Gi1 alpha (the inhibitory G protein of the i1 subtype) to generate a soluble GTP-binding protein, delta N-Gi1 alpha. Molecular genetic analyses of a 376-kDa Golgi complex (GC) membrane protein (giantin) are described. The immunoglobulin G fraction of a human serum containing antibodies against GC antigens as revealed by indirect immunofluorescence microscopy with Hep-2 cells was used to screen a HeLa cDNA expression library, yielding four overlapping cross-hybridizing clones. Additional cDNA clones were retrieved from a lambda gt11 human thyroid cDNA library or generated by reverse transcriptase-mediated PCR from HeLa cell mRNA. Alignment of the clones resulted in a consensus cDNA of 10,300 bp encoding a protein of 376 kDa. The corresponding mRNA with a size of about 10 kb was detected by Northern (RNA) blotting of HeLa, Hep-G2, and Jurkat cell RNA. Sequence analyses of the protein revealed an extraordinarily high content of heptad repeats with the probability of forming coiled coils similar to the proteins of the myosin family. Five overlapping recombinant proteins covering the entire sequence were synthesized and used for antibody production in rabbits and for affinity purification of human and rabbit antibodies. Indirect immunofluorescence experiments also done with brefeldin A-treated Hep-2 and Pt K1 cells revealed an identical GC staining of both the affinity-purified human and rabbit antibodies. Double labeling experiments with antibodies against the GC marker mannosidase II as well as immunoelectron microscopic studies confirmed the localization of the protein within the GC. The activity of DX-619 was evaluated against 376 anaerobic isolates using the reference CLSI agar dilution method. Overall, 90% of the strains were susceptible to DX-619 at ≤1 μg/ml. It was more active than the other four compounds tested except for meropenem, which showed virtually identical overall activity.

We showed that the α-CH2 → NH substitution in octanoyl-CoA alters the ground and transition state energies for the binding of the CoA ligands to medium-chain acyl-CoA dehydrogenase (MCAD), and such an effect is caused by a small electrostatic difference between the ligands. To ascertain the extent that the electrostatic contribution of the ligand structure and/or the enzyme site environment modulates the thermodynamics of the enzyme–ligand interaction, we undertook comparative microcalorimetric studies for the binding of 2-azaoctanoyl-CoA (α-CH2 → NH substituted octanoyl-CoA) and octenoyl-CoA to the wild-type and Glu-376 → Gln mutant enzymes. The experimental data revealed that both enthalpy (ΔH°) and heat capacity changes (ΔCp°) for the binding of 2-azaoctanoyl-CoA (ΔH°298 = −21.7 ± 0.8 kcal/mole, ΔCp° = −0.627 ± 0.04 kcal/mole/K) to the wild-type MCAD were more negative than those obtained for the binding of octenoyl-CoA (ΔH°298 = −17.2 ± 1.6 kcal/mole, ΔCp° = −0.526 ± 0.03 kcal/mole/K). Of these, the decrease in the magnitude of ΔCp° for the binding of 2-azaoctanoyl-CoA (vis-à-vis octenoyl-CoA) to the enzyme was unexpected, because the former ligand could be envisaged to be more polar than the latter. Progressive respiratory failure due to Pseudomonas aeruginosa is the leading cause of morbidity and mortality in patients with cystic fibrosis. The pulmonary delivery of antimicrobial agents provides high concentrations of drug directly to the site of infection and attains pharmacokinetic-pharmacodynamic indices exceeding those which can be achieved with systemic dosing. MP-376 is a new formulation of levofloxacin that enables the safe aerosol delivery of high concentrations of drug to pulmonary tissues. In vivo studies were conducted to demonstrate the efficacy of MP-376 in models of mouse pulmonary infection. The superiority of aerosol dosing over systemic dosing was demonstrated in models of both acute and chronic lung infection. In a model of acute lung infection, aerosol treatment with MP-376 once or twice daily reduced the lung bacterial load to a greater extent than aerosol tobramycin or aztreonam did when they were administered at similar or higher doses. The bacterial killing by aerosol MP-376 observed in the lung in the model of acute pulmonary infection translated to improved survival (P < 0.05). In a model of chronic pulmonary infection, aerosol MP-376 had antimicrobial effects superior to those of aztreonam (P < 0.05) and effects similar to those of tobramycin (P > 0.05). In summary...

The aim of this study was to evaluate our treatment of distal tibial physeal injuries retrospectively and explain the relationship between the trauma mechanism, the radiographic injury pattern, the subsequent therapy and the functional outcome, as well as to further deduce and verify prognostic criteria. At the Department of Trauma Surgery, Vienna Medical University, 419 children and adolescent patients with physeal injuries of the distal tibia were treated from 1993 to 2007, of these 376 were included in our study and evaluated retrospectively. Seventy-seven displaced physeal fractures of the distal tibia were reconstructed anatomically by open or closed reduction and produced 95% excellent results. A perfect anatomical reduction, if necessary by open means, should be achieved to prevent a bone bridge with subsequent epiphysiodesis and post-traumatic deformities due to growth inhibition and/or retardation.

Clusterin is a secreted glycoprotein expressed ubiquitously in many tissues that appears to function as a molecular chaperone capable of protecting stressed proteins. It is upregulated in many different forms of neurodegeneration and is thought to represent a defense response against neuronal damage. Clusterin has been found to be a common protein identified in drusen preparations isolated from the retina of donor eyes of patients with age-related macular degeneration (AMD), the leading cause of blindness in the elderly population of developed countries. A retina-specific clusterin-like protein (CLUL1) showing nearly 25% identity to clusterin at the protein level was recently cloned and shown to be expressed specifically in cone photoreceptor cells. For these reasons, we investigated CLUL1 as a candidate gene for AMD. A mutation screen of the entire coding region of the CLUL1 gene in 376 unrelated patients with AMD uncovered three sequence variations, one isocoding change and two intronic changes. One intronic change appears significantly less frequent in patients with the more severe forms of AMD than in control subjects, suggesting that this variant may reduce the risk for AMD or may be linked to a nearby variant that may reduce AMD risk. The pharmacokinetics and tolerability of nebulized MP-376 (levofloxacin inhalation solution [Aeroquin]) were determined in cystic fibrosis (CF) subjects. Ten CF subjects received single 180-mg doses of two formulations of MP-376, followed by a multiple-dose phase of 240 mg once daily for 7 days. Serum and expectorated-sputum samples were assayed for levofloxacin content. Safety was evaluated following the single- and multiple-dose study phases. Nebulized MP-376 produced high concentrations of levofloxacin in sputum. The mean maximum plasma concentration (Cmax) ranged between 2,563 and 2,932 mg/liter for 180-mg doses of the 50- and 100-mg/ml formulations, respectively. After 7 days of dosing, the mean Cmax for the 240-mg dose was 4,691 mg/liter. The mean serum levofloxacin Cmax ranged between 0.95 and 1.28 for the 180-mg doses and was 1.71 for the 240-mg dose. MP-376 was well tolerated. Nebulized MP-376 produces high sputum and low serum levofloxacin concentrations. The pharmacokinetics, safety, and tolerability were similar for the two formulations. MP-376 240 mg (100 mg/ml) is being advanced into late-stage clinical development.

Mutations in phosphoribosyl pyrophosphate synthetase 1 (PRPS1) are associated with a spectrum of non-syndromic to syndromic hearing loss. PRPS1 transcript levels have been shown to be regulated by the microRNA-376 genes. The long primary RNA transcript of the miR-376 RNA cluster members undergo extensive and simultaneous A → I editing at one or both of two specific sites (+4 and +44) in particular human and mouse tissues. The PRPS1 gene, which contains target sites for the edited version of miR-376a-5p within its 3′UTR, has been shown to be repressed in a tissue-specific manner. To investigate whether the transcription of Prps1 is regulated by miR-376 cluster members in the mouse inner ear, we first quantified the expression of the mature miR-376 RNAs by quantitative real-time-PCR. The spatio-temporal patterns of miR-376 expression were assessed by in situ hybridization. Finally, we examined whether A →I editing of pri-miR-376 RNAs occurs in mouse inner ear by direct sequencing. Diagnosis of coeliac disease (CD) relies on a combination of clinical, genetic, serological and duodenal morphological findings. The ESPGHAN suggested that biopsy may not be necessary in all cases. New guidelines include omission of biopsy if the concentration of CD-specific antibodies exceeds 10 times the upper limit of normal (10 ULN) and other criteria are met. We analysed the 10 ULN criterion and investigated multiple antibody-assays. Serum was collected from 1071 children with duodenal biopsy (376 CD patients, 695 disease-controls). IgA-antibodies to tissue transglutaminase (IgA-aTTG), IgG-antibodies to deamidated gliadin peptides (IgG-aDGL) and IgA-endomysium antibodies (IgA-EMA) were measured centrally. We considered 3 outcomes for antibody test procedures utilizing IgA-aTTG and/or IgG-aDGL: positive (≥10 ULN, recommend gluten-free diet), negative (<1 ULN, no gluten-free diet) or unclear (perform biopsy). Positive (PPV) and negative (NPV) predictive values were based on clear test results. We required that they and their lower confidence bounds (LCB) be simultaneously very high (LCB >90% and PPV/NPV >95%). In a prospective study 376 carotid artery plaques (275 symptomatic, 101 asymptomatic) were obtained from endarterectomies (184 unilateral and 96 bilateral) in 280 patients. The gross morphologic features of each plaque were noted at surgery and, together with the patient's clinical history, stored in computer memory. These data were analyzed in order to investigate the relationship of gross morphologic plaque characteristics with both the presence of cerebral symptoms and the degree of stenosis associated with the plaque. Ulceration was the most frequently observed of the five major gross plaque morphologic characteristics (46.0% of all plaques), but only intramural hemorrhage (30.6% of all plaques) was significantly more common in all symptomatic compared with all asymptomatic plaques (p less than 0.02). Hemorrhage was also the only gross characteristic significantly more common in focal symptomatic plaques when compared with either asymptomatic plaques (p less than 0.05) or nonfocal symptomatic plaques (p less than 0.01). We obtained long-slit optical spectra of the nuclear regions of 376 galaxies in the local Universe using the 1.5m Cassini telescope of Bologna Observatory. Of these spectra, 164 were either never taken before by the Sloan Digital Sky Survey (SDSS), or given by the Nasa Extragalactic Database (NED). With these new spectra, we contribute investigating the occurrence of active galactic nuclei (AGNs). Nevertheless, we stress that the present sample is by no means complete, thus, it cannot be used to perform any demographic study. Following the method presented in Gavazzi et al. (2011), we classify the nuclear spectra using a six bin scheme: SEY (Seyfert), sAGN (strong AGN), and wAGN (weak AGN) represent active galactic nuclei of different levels of activity; HII accounts for star-forming nuclei; RET (retired) and PAS (passive) refer to nuclei with poor or no star-formation activity. The spectral classification is performed using the ratio of 6584 {\lambda} [NII] to H{\alpha} lines and the equivalent width (EW) of H{\alpha} versus [NII]/H{\alpha} (WHAN diagnostic introduced by Cid Fernandes and collaborators) after correcting H{\alpha} for underlying absorption. We use deep images acquired with the Advanced Camera for Surveys (ACS) on board of the Hubble Space Telescope (HST) in the filters F555W and F814W to characterize the properties of NGC 376, a young star cluster located in the wing of the Small Magellanic Cloud (SMC). Using isochrone fitting we derive for NGC 376 an age of 28+/-7 Myr, in good agreement with previous studies. The high spatial resolution ACS data allow us to determine the center of gravity of the cluster and to construct extended surface brightness and radial density profiles. Neither of these profiles can be fitted with a theoretical model, suggesting that the cluster is not in virial equilibrium. Considering the young age of the cluster, we speculate that the distortion of the radial profiles may be the result of the rapid gas dispersal that follows the initial phase of star formation. The cluster shows clear evidence of dynamical mass segregation. From the properties of the radial profiles and the present day mass function (PDMF) we conclude that NGC 376 appears to have already lost nearly 90% of its initial stellar mass, probably as a consequence of the sudden gas dispersal that follows the early phase of star formation (SF).; Comment: 17 pages, 10 figures, accepted for publication in the Astrophysical Journal

The results of the redshift measurements and of the detailed surface photometry in BVR of the compact group ShCG 376 are presented. The radial velocity dispersion, the virial mass, the total luminosity, the M/L ratio, and the crossing time of the group are estimated. The group consists of eight accordant redshift spiral galaxies. Four (or possibly five) of the group members have emission-line spectra. Such morphological content and the number of emission-line galaxies are very atypical for compact galaxy groups. There are signs of interaction between some members of the group. It is suggested that the irregular shape of the brightest galaxy No. 4 is probably due to interaction with other members of the group, particularly, the emission line galaxy No. 6 with a discordant redshift (Delta v = 2600 km/s). Far Ultraviolet Spectroscopic Explorer observations are presented for WD0621-376, a DA white dwarf star in the local interstellar medium (LISM) at a distance of about 78 pc. The data have a signal-to-noise ratio of about 20-40 per 20 km/s resolution element and cover the wavelength range 905-1187 \AA. LISM absorption is detected in the lines of D I, C II, C II*, C III, N I, N II, N III, O I, Ar I, and Fe II. This sight line is partially ionized, with an ionized nitrogen fraction of > 0.23. We determine the ratio $D/O = (3.9 \pm ^{1.3}_{1.0})\times 10^{-2}$ (2$\sigma$). Assuming a standard interstellar oxygen abundance, we derive ${\rm D/H} \approx 1. 3 \times 10^{-5}$. Using the value of N(H I) derived from EUVE data gives a similar D/H ratio. The D I/N I ratio is $(3.3 \pm ^{1.0}_{0.8})\times 10^{-1}$ (2$\sigma$).; Comment: accepted for publication in the ApJS

We present a dynamical analysis of the galaxy cluster Abell 376 based on a set of 73 velocities, most of them measured at Pic du Midi and Haute-Provence observatories and completed with data from the literature. Data on individual galaxies are presented and the accuracy of the determined velocities is discussed as well as some properties of the cluster. We obtained an improved mean redshift value z=0.0478^{+0.005}_{-0.006} and velocity dispersion sigma=852^{+120}_{-76}km/s. Our analysis indicates that inside a radius of 900h_{70}^{-1}kpc (15 arcmin) the cluster is well relaxed without any remarkable feature and the X-ray emission traces fairly well the galaxy distribution. A possible substructure is seen at 20 arcmin from the centre towards the Southwest direction, but is not confirmed by the velocity field. This SW clump is, however, kinematically bound to the main structure of Abell 376. A dense condensation of galaxies is detected at 46 arcmin (projected distance 2.6h_{70}^{-1}Mpc) from the centre towards the Northwest and analysis of the apparent luminosity distribution of its galaxies suggests that this clump is part of the large scale structure of Abell 376. X-ray spectroscopic analysis of ASCA data resulted in a temperature kT = 4.3+/-0.4 keV and metal abundance Z = 0.32+/-0.08 Z_solar. The velocity dispersion corresponding to this temperature using the T_X-sigma scaling relation is in agreement with the measured galaxies velocities.; Comment: 11 pages...

In surface waters, for example, photodegradation may constitute a relevant natural attenuation process for drug residues that have been discharged from sewage treatment facilities. In the present work, the photochemical fate of the prodrug enalapril (376 Da, C(20)H(28)N2O5) and its active metabolite enalaprilat (348 Da, C(18)H(24)N2O5), a hypotensive cardioprotector previously reported to occur in contaminated rivers, was investigated in aqueous media under the influence of irradiation generated by a sunlight simulator. The experiments yielded three detectable photodegradates for enalapril (346 Da, 2 x 207 Da) whereas the photolysis of enalaprilat went hand in hand with the intermittent buildup of one photodegradate (304 Da). Fragmentation patterns of the parent compounds were established on a hybrid quadrupole-linear ion trap-mass spectrometer exploiting its MS3 capabilities.
