The purpose of this study was to describe the composition and abundance of rotifers in different environments: one lotie (the Ivinheima River) and another lentic (the Patos Lake) located in the floodplain of the Upper Paraná River. The influence of limnological variables on the structure and dynamics of rotifers was also examined. Samples were taken monthly from March 1992 through February 1993, in the open water region of the lake and river and in the channel between them and, in the littoral region of the lake and river. Of the 96 species identified, the best represented families were: Brachionidae (20 species), Lecanidae (18 species), and Trichocercidae (15 species). 55 species were widely distributed, 13 were pantropical, 16 neotropical (8 endemic to South America). Trichocerca gracilis was a new record for this floodplain. Dipleuchlanis propatula propatula, Lecane curvicornis, L. leontina, L. bulla, Plationus patulus patulus, Platyias quadricornis quadricornis, Testudinella patina, and T. mucronata hauriensis were consistently present. More species were recorded during the high water period, probably because of the interconnection among the different environments of the floodplain that allows fauna exchange and an increase in available habitats. The interaction mechanism between copper and propionic acid vapors is shown. Exposures were carried out to various pollutant concentrations and times. Cathodic scan, x-ray diffraction (XRD) and scanning electrode microscope (SEM) were used to quantify the degree of deterioration. The results obtained were explained by the following mechanism: A cuprite (Cu2O) layer covers the copper surface immediately; this compound in presence of adsorbed water is oxidized to tenorite (CuO). The hydration of the tenorite layer benefits the formation of a hydroxide compound such as CuO•xH2O. As the pollutant level is increased, insoluble basic propionate is formed (Cu(OH)x(CH3CH2 COO)2-x). Higher concentrations of propionic acid acidify the monolayers and a new compound Cu(CH3CH2COO)2•H 2O begins to precipitate at low pH, this is the main constituent of the green olive patina formed under these conditions. This mechanism agrees, in general terms, with those proposed in previous works to study the interaction between copper and the acetic and formic acids.

Public health laboratories at all capacity levels are facing challenges in exchanging electronic data among themselves and with their partners. In response to this the Association of Public Health Laboratories working collaboratively with CDC launched an innovative portal development project in January 2006. This portal will enable public health laboratories to collaborate in a web-based environment to establish a standardized vocabulary for test identifications and test results, a cornerstone for creating interoperable information systems.

Most gastrointestinal stromal tumors (GISTs) harbor oncogenic mutations in the KIT gene, and the majority of these mutations affect the juxtamembrane domain of the kinase encoded by exon 11. Screening GISTs for KIT gene mutations is important for translational research studies and for providing prognostic information on the likelihood of tumor response to treatment with the kinase inhibitor imatinib mesylate (Gleevec). In a series of GISTs analyzed in our laboratory by a combination of denaturing HPLC and direct DNA sequencing, we identified 19 cases with KIT exon 11 deletions that included from 1 to 14 bp of intron 10 sequence and resulted in loss of the normal splice acceptor site at the beginning of exon 11. Predicted use of the next potential splice-acceptor site was confirmed by cDNA sequencing in 4 cases. Thus, the resulting mutant isoform, deletion KPMYEVQWK 550–558, was the same in all 19 cases. Only two other examples of deletions across the intron 10–exon 11 boundary have been reported, yet among 722 GISTs analyzed in our laboratories these deletions were not uncommon, accounting for 3.9% of exon 11 mutations and 2.6% of all tumors. Oncogenic mutations of the receptor tyrosine kinase KIT contribute to the pathogenesis of gastrointestinal stromal tumors, systemic mastocytosis (SM), and some cases of acute myelogenous leukemia (AML). The D816V substitution in the activation loop of KIT results in relative resistance to the kinase inhibitor imatinib (Gleevec). Because this mutation occurs in 80 to 95% of adult SM, its detection has diagnostic and predictive significance. Unfortunately, the fraction of mutation-positive cells in clinical SM samples is often below the 20 to 30% threshold needed for detection by direct DNA sequencing. We have developed an allele-specific polymerase chain reaction assay using a mutation-specific primer combined with a wild-type blocking oligonucleotide that amplifies D816V at the level of 1% mutant allele in DNA extracted from formalin-fixed, paraffin-embedded tissue. There were no amplifications among 64 KIT wild-type tumors and cell lines, whereas all D816V-mutant samples (eight AML and 11 mast cell disease) were positive. This study compares two cleaning methods, one involving an ammonium carbonate-EDTA mixture and the other involving the sulfate-reducing bacterium Desulfovibrio vulgaris subsp. vulgaris ATCC 29579, for the removal of black crust (containing gypsum) on marble of the Milan Cathedral (Italy). In contrast to the chemical cleaning method, the biological procedure resulted in more homogeneous removal of the surface deposits and preserved the patina noble under the black crust. Whereas both of the treatments converted gypsum to calcite, allowing consolidation, the chemical treatment also formed undesirable sodium sulfate.

P-glycoprotein (Pgp) detoxifies cells by exporting hundreds of chemically unrelated toxins but has been implicated in multidrug resistance in the treatment of cancers. Substrate promiscuity is a hallmark of Pgp activity, thus a structural description of polyspecific drug-binding is important for the rational design of anticancer drugs and MDR inhibitors. The x-ray structure of apo-Pgp at 3.8 Å reveals an internal cavity of ∼6,000 Å3 with a 30 Å separation of the two nucleotide binding domains (NBD). Two additional Pgp structures with cyclic peptide inhibitors demonstrate distinct drug binding sites in the internal cavity capable of stereo-selectivity that is based on hydrophobic and aromatic interactions. Apo- and drug-bound Pgp structures have portals open to the cytoplasm and the inner leaflet of the lipid bilayer for drug entry. The inward-facing conformation represents an initial stage of the transport cycle that is competent for drug binding.

Several fungi present high tolerance to toxic metals and some are able to transform metals into metal–oxalate complexes. In this study, the ability of Beauveria bassiana to produce copper oxalates was evaluated. Growth performance was tested on various copper-containing media. B. bassiana proved highly resistant to copper, tolerating concentrations of up to 20 g L−1, and precipitating copper oxalates on all media tested. Chromatographic analyses showed that this species produced oxalic acid as sole metal chelator. The production of metal–oxalates can be used in the restoration and conservation of archeological and modern metal artifacts. The production of copper oxalates was confirmed directly using metallic pieces (both archeological and modern). The conversion of corrosion products into copper oxalates was demonstrated as well. In order to assess whether the capability of B. bassiana to produce metal–oxalates could be applied to other metals, iron and silver were tested as well. Iron appears to be directly sequestered in the wall of the fungal hyphae forming oxalates. However, the formation of a homogeneous layer on the object is not yet optimal. Following an acute stressor, pre-adolescent rats exhibit a protracted hormonal response compared to adults, while after repeated exposure to the same stressor (i.e., homotypic stress) prepubertal males fail to habituate like adults. Though the neurobehavioral implications of these changes are unknown, studying pubertal shifts in stress reactivity may help elucidate the mechanisms that underlie the increase in stress-related psychological and physiological disorders often observed during adolescence. Here, we investigated hormonal, behavioral, and neural responses of prepubertal (30d) and adult (77d) male rats before, during, or after acute stress (restraint), homotypic stress (repeated restraint) or heterotypic stress (repeated cold exposure followed by restraint). We found that prepubertal males exhibit prolonged corticosterone responses following acute and heterotypic stress, and higher adrenocorticotropic hormone and corticosterone responses after homotypic stress, compared to adults. Despite these significant age-dependent changes in hormonal responsiveness, we found struggling behavior during restraint was similar at both ages, such that both prepubertal and adult animals exposed to homotypic stress struggled less than animals exposed to either acute or heterotypic stress. Depuis quelques temps, on note que les objets d’apparence artisanale symbolisent souvent des qualités écologiques telles que des matériaux naturels et une fabrication éthique. Les visées de l’étude ont été d’explorer les perceptions à l’égard des objets à caractère artisanal et cela, en vue de reconnaître quelles qualités écologiques sont attribuées à ces derniers, tout en tentant de comprendre les raisons qui se cachent derrière ces associations. Une étude auprès d’usagers a permis d’explorer l’ensemble des qualités pouvant être liées à ce type d’objet, en considérant plus précisément le rapport entre les qualités écologiques leur étant accordées et leur durée de vie projetée. Pour ce faire, au cours d’entretiens individuels, des thèmes comme l’appréciation à long terme, la signification et la considération de la diversité culturelle ont été examinés. non-peer-reviewed; This thesis document attempts to explore the idea of decay with regards to buildings; decay in the negative form- the premature demise and death of buildings, the run down nature of certain sites and the unwanted demolition of unadaptable buildings. I also wanted to look at decay in the positive form: mans love affair with ruins, the build up of human patina on a building overtime, uncovering stories and events within its walls. All buildings are capable of such positive decay, which can also be attributed to weathering, however few get the chance to survive this long. In thinking about this, how do we build to last? Looking at infrastructures and they’re nature of survival and durability, I thought about Limerick City and its infrastructural spaces that have with stood the test of time. During construction of the city, Limerick was built a level higher than the original ground plane to allow for the River Shannon to flood underneath without harming the buildings. Thus, a series of culverts, basements and coal bunkers create a secondary subterranean landscape beneath the streets. In thinking about this, I questioned solely building up, and if the entire groundscape is to be constructed, why not take advantage of the spaces that could be uncovered beneath. Thus creating an excavation of subtraction and addition.
