Leukotrienes are reported to be potent proinflammatory mediators that play a role in the development of several inflammatory diseases such as asthma, rheumatoid arthritis and periodontal disease. Leukotrienes have also been associated with protection against infectious diseases. However, the role of leukotrienes in Mycobacterium tuberculosis infection is not understood. To answer this question, we studied the role of leukotrienes in the protective immune response conferred by prime-boost heterologous immunization against tuberculosis. We immunized BALB/c mice (4-11/group) with subcutaneous BCG vaccine (1 x 10(5) M. bovis BCG) (prime) followed by intramuscular DNA-HSP65 vaccine (100 µg) (boost). During the 30 days following the challenge, the animals were treated by gavage daily with MK-886 (5 mg·kg-1·day-1) to inhibit leukotriene synthesis. We showed that MK-886-treated mice were more susceptible to M. tuberculosis infection by counting the number of M. tuberculosis colony-forming units in lungs. As vacinas antimeningocócicas têm se demonstrado efetivas contra os sorogrupos A e C, no entanto ainda não existe vacina contra o sorogrupo B devido à similaridade entre a estrutura capsular do polissacáride B e o ácido polisiálico que faz parte do tecido cerebral humano, podendo levar à autoimunidade. O objetivo deste estudo foi investigar as propriedades adjuvantes dos toxóides Stx1 e Stx2 (STEC) de Escherichia coli, administrados em preparações antigênicas com vesículas de membrana externa nativa (NOMV) de Neisseria meningitidis B, comparando duas vias de imunização prime-boost ou intramuscular, em camundongos BALB/c com idade entre 6-8 semanas. A determinação dos níveis de anticorpos empregando a técnica de ELISA, mostrou elevadas concentrações de anticorpos IgG em soros de animais imunizados pela via intramuscular com Stx1+NOMV, mas não com NOMV, o que sugere que por esta via (intramuscular apenas) Stx1 possa ter atuado como adjuvante. O presente trabalho aborda a análise, o projeto, a implementação e os ensaios de um retificador com correção ativa do fator de potência, o qual permite alimentar cargas CC a partir de uma fonte senoidal, injetando baixo conteúdo harmônico na corrente de alimentação e operando com fator de potência unitário. Serão desenvolvidos os circuitos de potência e o software para o controle e a modulação do conversor boost utilizado para o propósito citado. Purpose To assess outcomes with external beam radiation therapy (EBRT) and a low-dose-rate (LDR) brachytherapy boost without or with androgen deprivation therapy (ADT) for prostate cancer. Materials and Methods From January 2001 through August 2011, 120 intermediate-risk or high-risk prostate cancer patients were treated with EBRT to a total dose of 4,500 cGy in 25 daily fractions and a palladium-103 LDR brachytherapy boost of 10,000 cGy (n = 90) or an iodine-125 LDR brachytherapy boost of 11,000 cGy (n = 30). ADT, consisting of a gonadotropin-releasing hormone agonist ± an anti-androgen, was administered to 29/92 (32%) intermediate-risk patients for a median duration of 4 months and 26/28 (93%) high-risk patients for a median duration of 28 months. Results Median follow-up was 5.2 years (range, 1.1-12.8 years). There was no statistically-significant difference in biochemical disease-free survival (bDFS), distant metastasis-free survival (DMFS), or overall survival (OS) without or with ADT. Immunization of jirds with Bm-alt-2 elicited partial protection against challenge infection with the filarial parasite Brugia malayi. In this study, we initially compared the protective immune responses elicited following immunization with recombinant Bm-ALT-2 protein regimen and Bm-alt-2 DNA regimen. These studies showed that protein vaccination conferred approximately 75% protection compared to DNA vaccination that conferred only 57% protection. Analysis of the protective immune responses showed that the protein immunization promoted a Th2-biased response with an increase in IL-4, IL-5 and IgG1 responses, whereas, the DNA vaccine promoted a Th1-biased response with profound IFN-γ and IgG2a responses. Since protein vaccination gave better results than DNA vaccination, we then wanted to evaluate whether a prime-boost vaccination that combined DNA prime and protein boost will significantly increase the protective responses induced by the protein vaccine. Our results suggest that prime-boost vaccination had no added advantage and was comparatively less effective (64% protection) than the Bm-ALT-2 protein alone vaccination. Currently, there are more than 30 million people infected with HIV-1 and thousands more are infected each day. Vaccination is the single most effective mechanism for prevention of viral disease, and after more than 25 years of research, one vaccine has shown somewhat encouraging results in an advanced clinical efficacy trial. A modified intent-to-treat analysis of trial results showed that infection was approximately 30% lower in the vaccine group compared to the placebo group. The vaccine was administered using a heterologous prime-boost regimen in which both target antigens and delivery vehicles were changed during the course of inoculations. Here we examine the complexity of heterologous prime-boost immunizations. We show that the use of different delivery vehicles in prime and boost inoculations can help to avert the inhibitory effects caused by vector-specific immune responses. We also show that the introduction of new antigens into boost inoculations can be advantageous, demonstrating that the effect of ‘original antigenic sin’ is not absolute. Pre-clinical and clinical studies are reviewed, including our own work with a three-vector vaccination regimen using recombinant DNA, virus (Sendai virus or vaccinia virus) and protein. Promising preliminary results suggest that the heterologous prime-boost strategy may possibly provide a foundation for the future prevention of HIV-1 infections in humans.

Most cancer vaccines, thus far, fail to control established tumors. However, their application in preventing tumors is another question that is understudied. In the current study, we investigated the CD8 memory T cell responses of lentivector (lv) immunization and its potential to prevent melanoma using both transplantable B16 tumor and autochthonous melanoma models. We found that lv expressing xenogenic human glycoprotein (hgp100) could induce potent CD8 responses that cross react with mouse gp100. Importantly, the lv primed CD8 response consisted of a high number of memory precursors and could be further increased by recombinant vaccinia virus vector (vv) boost, resulting in enhanced CD8 memory response. These long-lasting CD8 memory T cells played a critical role in immune surveillance and could rapidly respond and expand after sensing B16 tumor cells to prevent tumor establishment. Although CD8 response plays a dominant role after lv immunization, both CD4 and CD8 T cells are responsible for the immune prevention. Adenoviruses show promising potential as vectors for cancer vaccines, however, their high immunogenicity can be problematic when it comes to homologous prime-boost strategies. In the studies presented here we show that heterologous prime-boost vaccinations involving ovalbumin (OVA)-antigen coated microparticles as a prime, and adenovirus encoding OVA (Ad-OVA) as a boost, were equally as effective as homologous Ad-OVA prime-boosts at generating OVA-specific CD8+ T cell responses, which translated into effective tumor protection. OVA-coated biodegradable poly α-hydroxy acid based microparticles of varying chemistries, when used as primes in heterologous prime-boost vaccinations, were comparable in terms of promoting OVA-specific CD8+ T cells as well as providing protection against subsequent tumor challenge. These findings auger well for using poly α-hydroxy acid based microparticles in prime-boost viral vaccination strategies geared toward the safer, and potentially more efficient, generation of anti-tumor immunity.

An effective vaccine usually requires more than one time immunization in the form of prime-boost. Traditionally the same vaccines are given multiple times as homologous boosts. New findings suggested that prime-boost can be done with different types of vaccines containing the same antigens. In many cases such heterologous prime-boost can be more immunogenic than homologous prime-boost. Heterologous prime-boost represents a new way of immunization and will stimulate better understanding on the immunological basis of vaccines.

Passive component volume is a perennial concern in power conversion. With new circuit architectures operating at extreme high frequencies it becomes possible to miniaturize the passive components needed for a power converter, and to achieve dramatic improvements in converter transient performance. This thesis focuses on the development of a Very High Frequency (VHF, 30 - 300 MHz) dc-dc boost converter using a MOSFET fabricated from a typical power process. Modeling and design studies reveal the possibility of building VHF dc-dc converters operable over the full automotive input voltage range (8 - 18 V) with transistors in a 50 V power process, through use of newly-developed resonant circuit topologies designed to minimize transistor voltage stress. Based on this, a study of the design of automotive boost converters was undertaken (e.g., for LED headlamp drivers at output voltages in the range of 22 - 33 V.) Two VHF boost converter prototypes using a [Phi]2 resonant boost topology were developed. Neste trabalho é proposta uma modelagem do conversor não-linear CC-CC Boost utilizando a forma generalizada dos sistemas fuzzy Takagi – Sugeno (TS). A modelagem desenvolvida também é chamada de forma exata, a qual permite o projeto de sistemas de controle sem a necessidade da linearização em torno do ponto de operação. Utilizando a modelagem proposta neste trabalho, projeta-se um sistema de rastreamento do sinal de saída para o Boost, operando em modo de condução contínua. A metodologia desenvolvida é descrita em termos de LMIs, o que permite descrever o problema de forma convexa. Este projeto consiste basicamente de duas etapas. Na primeira fase projeta-se um controlador fuzzy de realimentação das variáveis de estados, K(α), objetivando estabilizar o conversor em malha fechada. Este trabalho apresenta uma nova proposta de controle para conversor boost operando como retificador de alto fator de potência, a qual permite priorizar o requisito de alto fator de potência, sob condição normal da rede, ou priorizar a regulação da tensão de saída diante de afundamentos de tensão. O controle implementado consiste na modificação das malhas de realimentação do circuito 3854. O sistema de monitoramento contém o microcontrolador LPC2138, que comanda a alteração das malhas do 3854 em função das condições de operação da rede elétrica. BACKGROUND: There is renewed interest in the development of poxvirus vector-based HIV vaccines due to the protective effect observed with repeated recombinant canarypox priming with gp120 boosting in the recent Thai placebo-controlled trial. This study sought to investigate whether a heterologous prime-boost-boost vaccine regimen in Chinese cynomolgus macaques with a DNA vaccine and recombinant poxviral vectors expressing HIV virus-like particles bearing envelopes derived from the most prevalent clades circulating in sub-Saharan Africa, focused the antibody response to shared neutralising epitopes. METHODS: Three Chinese cynomolgus macaques were immunised via intramuscular injections using a regimen composed of a prime with two DNA vaccines expressing clade A Env/clade B Gag followed by boosting with recombinant fowlpox virus expressing HIV-1 clade D Gag, Env and cholera toxin B subunit followed by the final boost with recombinant modified vaccinia virus Ankara expressing HIV-1 clade C Env, Gag and human complement protein C3d. We measured the macaque serum antibody responses by ELISA, enumerated T cell responses by IFN-γ ELISpot and assessed seroneutralisation of HIV-1 using the TZM-bl β-galactosidase assay with primary isolates of HIV-1. RESULTS: This study shows that large and complex synthetic DNA sequences can be successfully cloned in a single step into two poxvirus vectors: MVA and FPV and the recombinant poxviruses could be grown to high titres. The vaccine candidates showed appropriate expression of recombinant proteins with the formation of authentic HIV virus-like particles seen on transmission electron microscopy. In addition the b12 epitope was shown to be held in common by the vaccine candidates using confocal immunofluorescent microscopy. Several sensors are used to detect and track the missile. These sensors differ from each other in many different aspects. The outputs of radars give range, elevation and azimuth information of the target while space based infrared sensors give elevation and azimuth information. These outputs have to be combined (fused) achieve better position information for the missile. The architecture that is used in this thesis is decision level fusion architecture. This thesis examines four algorithms to fuse the results of radar sensors and space based infrared sensors. An averaging technique, a weighted averaging technique, a Kalman filtering approach and a Bayesian technique are compared. The ballistic missile boost phase segment and the sensors are modeled in MATLAB. The missile vector and dynamics are based upon Newton's laws and the simulation uses an earth-centered coordinate system. The aim of the present study was to assess the effect of introducing a priming step with replication-defective viral vectors encoding the capsid proteins of FMDV, followed by a boost with killed virus vaccines, using a suitable BALB/c mice model. Additionally, the immune response to other combined vector immunization regimens was studied. The highest antibody titers were found in the group that received two doses of adjuvanted KV (P < 0.002). Antibody titers were higher in those groups receiving a mixed regimen of vectors, compared to immunization with either vector alone (P < 0.0001). Priming with any of the viral vectors induced a shift of the cytokine balance toward a Th1 type immune response regardless of the delivery system used for boosting. The highest IgG1 titer was induced by two doses of adjuvanted KV (P = 0.0002) and the highest IgG2a titer corresponded to the group primed with Ad and boosted with KV (P = 0.01). Guitar Boost es un videojuego musical que utiliza cualquier guitarra real (eléctrica o electroacústica) conectada al computador sin necesidad de comprar un adaptador adicional. El videojuego evalúa la calidad interpretativa del jugador permitiéndole aprender a tocar guitarra de manera progresiva a través del feedback entregado en tiempo real. Guitar Boost se ofrecerá a los jugadores en dos modalidades: 1.- Un producto para aprender o mejorar en la interpretación en guitarra combinando factores educativos con lo entretenido de un videojuego. 2.- Un servicio una versión web de Guitar Boost con las mismas funcionalidades pero que permita jugar a las personas sin tener que instalar un software. La detección de las notas y acordes que toca el jugador se realiza utilizando un potente y estable algoritmo de procesamiento digital de señales. El algoritmo desarrollado por el equipo emprendedor toma la señal de audio de la guitarra para luego hacer un procesamiento en el dominio del tiempo de las frecuencias y de las wavelets para poder reconocer hasta tres notas tocadas a la vez con bajos tiempos de respuesta lo cual permite una jugabilidad óptima con cualquier guitarra (eléctrica o electroacústica) sin importar marca o modelo. Para la siguiente versión se va a abrir una línea de investigación con el objetivo de lograr un reconocimiento completo de las 6 cuerdas de la guitarra. Modelo de Negocios: El modelo de negocios para cada una de las modalidades es distinto: 1.- Para el producto se lanzarán versiones periódicamente enfocadas tanto gráfica como musicalmente en un grupo o artista determinado. Se venderá en versión física y digital. 2.- El servicio. Versión gratuita y limitada con el fin de generar rápidamente una masa crítica. Canciones de distintos artistas. Se ofrecerá la opción de pagar por una suscripción que dará acceso a más funcionalidades y canciones. Facilidad de expansión a otros países a través de Internet y el pagar online (PayPal). Descripción del Mercado: El mercado objetivo son jóvenes entre 12 y 35 años que tengan interés por aprender a tocar guitarra o bien ya saben tocar. El mercado está dividido en el producto y el servicio ofrecidos siendo el tamaño de estos distintos entre sí. Se hizo un estudio utilizando la herramienta Facebook Adverts que en base a los usuarios de esta red social entrega cifras de las personas con ciertos intereses y edades. 1.- Servicio: El tamaño del mercado es como base 260.000 personas en Chile. Esta cifra toma en cuenta personas cuyos intereses estén relacionados con el arte de tocar guitarra. 2.- Producto: Este mercado ataca a un nicho distinto en cada lanzamiento ya que apunta a seguidores de determinados grupos musicales. Sin embargo crece a medida de que se lancen más versiones. El tamaño de mercado estimado en Chile es de 350.000 personas. Esto considerando las personas que son fieles seguidores de las bandas nacionales más importantes. Cabe destacar que las cifras entregadas corresponden al territorio nacional. Al expandirse a otros países de Latinoamérica la cantidad de personas crece abruptamente. Competencia: Rockband 3 y PowerGig no permiten jugar con cualquier guitarra obligando al usuario a comprarl. Rocksmith aún no se lanza al mercado permitirá el uso de cualquier guitarra pero sólo puede ser usado en Xbox 360 Play Station 3 y PC con Windows dejando de lado Max y Linux. Además cuenta sólo con canciones de artistas de fuera de Latinoamérica. Crecimiento y expansión: Se llevará a cabo captando nuevos clientes a través de la versión gratuita online por otro lado se irá atacando a diferentes nichos correspondientes a los adherentes a artistas o grupos específicos. La rápida masificación de internet ayudará que este proceso. Ya se está en conversaciones con el grupo chileno Los Tres para realizar la primera versión del juego enfocado en bandas y se están haciendo los contactos para contar con otras bandas que la procederán. Una vez que se cuenta con la primera versión del juego enfocada en una banda se pretende realizar un evento de lanzamiento con la participación del grupo. Este modelo se pretende implementar en otros países de Latinoamérica con las bandas que tienen más seguidores en cada país y realizando diferentes versiones del juego. Además en un futuro a mediano plazo se pretende crear una empresa de videojuegos que no sólo cuente con Guitar Boost sino que también cree más productos nuevos y con valor de alto impacto. Así Guitar Boost se convertirá en la primera piedra de lo que será una de las empresas más grandes de videojuegos a nivel nacional. Colaboradores Relevantes: El más relevante es el grupo musical Los Tres . Se está conversando con ellos para que la primera versión por grupos sea del suyo. Además también se está trabajando con sus sellos disqueros (Sony y FeriaMix) para poder lograr esta alianza.; Poner en marcha la operación productiva y comercial del proyecto de desarrollo de video juegos que comenzará con el lanzamiento del video juego musical Guitar Boost. Formal and Computational Properties of the Confidence Boost of Association Rules

Some existing notions of redundancy among association rules allow for a logical-style characterization and lead to irredundant bases of absolutely minimum size. One can push the intuition of redundancy further and find an intuitive notion of interest of an association rule, in terms of its "novelty" with respect to other rules. Namely: an irredundant rule is so because its confidence is higher than what the rest of the rules would suggest; then, one can ask: how much higher? We propose to measure such a sort of "novelty" through the confidence boost of a rule, which encompasses two previous similar notions (confidence width and rule blocking, of which the latter is closely related to the earlier measure "improvement"). Acting as a complement to confidence and support, the confidence boost helps to obtain small and crisp sets of mined association rules, and solves the well-known problem that, in certain cases, rules of negative correlation may pass the confidence bound. We analyze the properties of two versions of the notion of confidence boost, one of them a natural generalization of the other. Fast ABC-Boost for Multi-Class Classification

Abc-boost is a new line of boosting algorithms for multi-class classification, by utilizing the commonly used sum-to-zero constraint. To implement abc-boost, a base class must be identified at each boosting step. Prior studies used a very expensive procedure based on exhaustive search for determining the base class at each boosting step. Good testing performances of abc-boost (implemented as abc-mart and abc-logitboost) on a variety of datasets were reported. For large datasets, however, the exhaustive search strategy adopted in prior abc-boost algorithms can be too prohibitive. To overcome this serious limitation, this paper suggests a heuristic by introducing Gaps when computing the base class during training. That is, we update the choice of the base class only for every $G$ boosting steps (i.e., G=1 in prior studies). We test this idea on large datasets (Covertype and Poker) as well as datasets of moderate sizes. Our preliminary results are very encouraging. On the large datasets, even with G=100 (or larger), there is essentially no loss of test accuracy. On the moderate datasets, no obvious loss of test accuracy is observed when G<= 20~50. Therefore, aided by this heuristic, it is promising that abc-boost will be a practical tool for accurate multi-class classification.

Full calculation of clumpiness boost factors for antimatter cosmic rays in the light of Lambda-CDM N-body simulation results

Anti-proton and positron Galactic cosmic ray spectra are among the key targets for indirect detection of dark matter. The boost factors, corresponding to an enhancement of the signal and linked to the clumpiness properties of the dark matter distribution, have been taken as high as thousands in the past. The dramatic impact of these boost factors for indirect detection of antiparticles, for instance with the PAMELA satellite or the coming AMS-02 experiment, asks for their detailed calculation. We take into account the results of high resolution N-body dark matter simulations to calculate the most likely energy dependent boost factors linked to the cosmic ray propagation properties, for anti-protons and positrons. Starting from the mass and space distributions of sub-halos, the anti-proton and positron propagators are used to calculate the mean value and the variance of the boost factor for the primary fluxes. We take advantage of the statistical method introduced in Lavalle et al. (2007) and cross-check the results with Monte Carlo computations. By spanning some extreme configurations of sub-halo and propagation properties, we find that the average contribution of the clumps is negligible compared to that of the smooth dark matter component.
