A technical note on the phase transformation in furnace container material after a periodic thermo-chemical treatment

The aim of this work is to investigate the metallurgical changes in the wall of cylindrical containers, fabricated by welding from AISI 310 steel, used in the thermal and chemical treatment of spindle chains for the automotive industry, for an in-service period of over 1000 h. In order to identify the phases originated during this in-service period, several etchants were used in the structural study, together with X-ray diffraction and 57Fe Mössbauer spectroscopy analysis. The metallographic technique demonstrates some limitations in the complete identification of the phases produced in the wall of the containers. The structure of the wall of the containers, after the referred to working period, is composed of an austenitic matrix, whose grain size is about 212 and 238 [mu]m, near the internal and external faces of the wall, respectively. These show numerous precipitates inside the grains and at the grain boundaries. The density of the precipitates decreases from the internal to the external surface. The precipitates are nitrides (Cr, Mo)12 (Fe,Ni)8-x N4-z near the internal surface, and carbides (Cr, Fe, Mo)23C6 on the other zones of the wall.; http://www.sciencedirect.com/science/article/B6TGJ-4502Y49-G/1/4e6ae31d16e053b40c961bebbfda1e0a

Values of binary mutual diffusion (interdiffusion) coefficients, obtained by using the Taylor dispersion method, for aqueous solutions of γ-cyclodextrin in the concentration range from (0.002 to 0.010) mol·dm−3 and temperatures (298.15 and 310.15) K are reported. From these experimental results, the hydrodynamic radius values for the γ-cyclodextrin are estimated. Also, the measured diffusion coefficients are used with both Hartley’s and Gordon’s equations to estimate activity coefficients for aqueous γ-cyclodextrin. These studies are complemented by some density and viscosity measurements, carried out at the same range of concentrations and temperatures. Binary mutual diffusion coefficients measured by the Taylor dispersion method in two different laboratories (University of Naples, Federico II, Italy, and University of Coimbra, Portugal) are reported for aqueous solutions of isoniazid at concentrations from (0.000 to 0.100) mol·dm−3 and at two temperatures (298.15 and 310.15) K. The hydrodynamic radii for the isoniazid in aqueous solution are calculated from the experimental results. In addition, the Hartley equation and the experimental diffusion coefficients are used to estimate activity coefficients for aqueous isoniazid at both temperatures

Several methods exist to retrieve and purify DNA fragments after agarose or polyacrylamide gel electrophoresis for subsequent analyses. However, molecules present in low concentration and molecules similar in size to their neighbors are difficult to purify. Capillary electrophoresis has become popular in molecular diagnostic laboratories because of its automation, excellent resolution, and high sensitivity. In the current study, the ABI Prism 310 Genetic Analyzer was reconfigured into a fraction collector by adapting the standard gel block to accommodate a collection tube at the distal end of capillary. The time to collect the desired peaks was estimated by extrapolating from standard capillary electrophoresis using the original gel block. Fraction collection from a mixture of DNA fragments amplified from wild type and several internal tandem duplication mutations of the FMS-like tyrosine kinase 3 (Flt3) gene yielded highly purified DNA fragments containing internal tandem duplication mutations and predictable electrokinetics using the reconstructed gel block. The reconfigured instrument could successfully isolate DNA amplicons from extremely low-amplitude peaks (110 relative fluorescent units), which were undetectable using polyacrylamide gel electrophoresis. Zinc is an essential trace element for all living organisms and plays pivotal roles in various cellular processes. However, an excess of zinc is extremely deleterious to cells. Bacteria have evolved complex machineries (such as efflux/influx systems) to control the concentration at levels appropriate for the maintenance of zinc homeostasis in cells and adaptation to the environment. The Zur (zinc uptake regulator) protein is one of these functional members involved in the precise control of zinc homeostasis. Here we identified a zur homologue designated 310 from Streptococcus suis serotype 2, strain 05ZYH33, a highly invasive isolate causing streptococcal toxic shock syndrome. Biochemical analysis revealed that the protein product of gene 310 exists as a dimer form and carries zinc ions. An isogenic gene replacement mutant of gene 310, the Δ310 mutant, was obtained by homologous recombination. Physiological tests demonstrated that the Δ310 mutant is specifically sensitive to Zn2+, while functional complementation of the Δ310 mutant can restore its duration capability, suggesting that 310 is a functional member of the Zur family. Two-dimensional electrophoresis indicated that nine proteins in the Δ310 mutant are overexpressed in comparison with those in the wild type. Background and Objective. The aim of this study was to assess the subjective outcome following laparoscopic sacropexy. Methods. We performed a retrospective cohort study among women treated for descensus with laparoscopic sacropexy between January 2000 and December 2007. 310 patients received questionnaires during followup assessing major pre- and postoperative symptoms and overall satisfaction. Results. 214 (69%) patients responded to the questionnaire. Mean followup was 24.5 months. The number of patients with back or lower abdominal pain, foreign body sensation in the vagina and prolapse-related symptoms, urinary symptoms, vaginal and bladder infections, and the need for pessary usage decreased significantly postsurgically. Bowel symptoms increased slightly but not significantly. Two years after surgery, nearly 2 thirds of the women were satisfied or very satisfied with the outcome. Conclusion. Laparoscopic sacropexy is an effective treatment of descensus, with favorable or satisfactory subjective outcomes.

HLA-DM is required for efficient peptide exchange on class II MHC molecules, but its mechanism of action is controversial. We trapped an intermediate state of class II MHC HLA-DR1 by substitution of αF54, resulting in a protein with increased HLA-DM binding affinity, weakened MHC-peptide hydrogen bonding as measured by hydrogen-deuterium exchange mass spectrometry, and increased susceptibility to DM-mediated peptide exchange. Structural analysis revealed a set of concerted conformational alterations at the N-terminal end of the peptide-binding site. These results suggest that interaction with HLA-DM is driven by a conformational change of the MHC II protein in the region of the α-subunit 310 helix and adjacent extended strand region, and provide a model for the mechanism of DM-mediated peptide exchange.

Hepatitis C virus (HCV) is susceptible to cyclosporine (CsA) and other cyclophilin (CypA) inhibitors, but the genetic basis of susceptibility is controversial. Whether genetic variation in NS5A alters cell culture susceptibility of HCV to CypA inhibition is unclear. We constructed replicons containing NS5A chimeras from genotypes 1a, 2a and 4a to test how variation in carboxy terminal regions of NS5A altered the genotype 1b CsA susceptibility. All chimeric replicons including genotype 1b Con1LN-wt replicon exhibited some cell culture sensitivity to CsA with genotype 4a being most sensitive and 1a the least. The CypA binding pattern of truncated NS5A genotypes correlated with the susceptibility of these replicons to CsA. The Con1LN-wt replicon showed increased susceptibility towards CsA when proline at position 310P was mutated to either threonine or alanine. Furthermore, a 15 amino acid long peptide fused N terminally to GFP coding sequences confirmed involvement of proline at 310 in CypA binding. Our findings are consistent with CypA acting on multiple prolines outside of the previously identified CypA binding sites. Background Few prospective data from the Asia-Pacific region are available relating body mass index (BMI) to the risks of stroke and ischaemic heart disease (IHD). Our objective was to assess the age-, sex-, and region-specific associations of BMI with cardiovascular disease using individual participant data from prospective studies in the Asia-Pacific region. Methods Studies were identified from literature searches, proceedings of meetings, and personal communication. All studies had at least 5000 person-years of follow-up. Hazard ratios were calculated from Cox models, stratified by sex and cohort, and adjusted for age at risk and smoking. The first 3 years of follow-up were excluded in order to reduce confounding due to disease at baseline. Results A total of 33 cohort studies, including 310 283 participants, contributed 2 148 354 person-years of follow-up, during which 3332 stroke and 2073 IHD events were observed. There were continuous positive associations between baseline BMI and the risks of ischaemic stroke, haemorrhagic stroke, and IHD, with each 2 kg/m2 lower BMI associated a 12% (95% CI: 9, 15%) lower risk of ischaemic stroke, 8% (95% CI: 4, 12%) lower risk in haemorrhagic stroke, and 11% (95% CI: 9, 13%) lower risk of IHD. Slowly but steadily bibliographic evidence is accumulating that the apparent convergence of the various biomolecular force fields as evidenced from simulations of proteins in the folded state does not hold true for folding simulations. Here we add one more example to the growing list of peptides and proteins for which different force fields show irreconcilable differences in their folding predictions, even at such a fundamental level as that of a peptide's secondary structure. We show that for an undecamer peptide that is known from two independent NMR structure determinations to have a mainly 310-helical structure in solution, three mainstream biomolecular force fields give completely disparate predictions: The CHARMM force field (with the CMAP correction) predicts an outstandingly stable α-helical structure, in disagreement not only with the experimental structures, but also with experimental evidence obtained from circular dichroism. OPLS-AA shows an almost totally disordered peptide with the most frequently observed folded conformation corresponding to a β-hairpin-like structure, again in disagreement with all available experimental evidence. Only the AMBER99SB force field appears to qualitatively agree with not only the general structural characteristics of the peptide (on the account of both NMR- and CD-based experiments)...

Engineering Failure Analysis, Elsevier, Volume 18, Issue 6, setembro 2011. 1435–1444.; In this work a failure analysis was conducted in a mirror plate made of austenitic stainless steel AISI 310. The mirror is part of the inert gas generator, which is a combustion chamber dedicated to produce inert gas atmosphere (N2 + CO2) to be used during the oil transfer in off-shore platforms. The mirror failed in two distinct regions. The first crack was observed in a region where the temperatures reached were sufficiently high to provoke intense sigma phase precipitation. Other cracks were observed in a region subject to seawater refrigeration, named ‘‘cold zone’’. These two failures were mitigated in order to propose solutions to avoid similar failures in the new equipment.

The MAGIC Cherenkov telescopes observed the Perseus cluster sky region in stereo mode for nearly 90 hr from October 2009 to February 2011. This campaign led to the discovery of very high energy Gamma-ray emission from the central radio galaxy NGC 1275 and the head-tail radio galaxy IC 310. Here we report the results on the most recent discovery of NGC 1275 which was detected at low energies in the 2010/2011 data. We report on the detection with the MAGIC telescopes of very high energy gamma-rays from IC 310, a head-tail radio galaxy in the Perseus galaxy cluster, observed during the interval November 2008 to February 2010. The Fermi satellite has also detected this galaxy. The source is detected by MAGIC at a high statistical significance of 7.6sigma in 20.6 hr of stereo data. The observed spectral energy distribution is flat with a differential spectral index of -2.00 \pm 0.14. The mean flux above 300 GeV, between October 2009 and February 2010, (3.1 \pm 0.5)x10^{-12} cm^{-2} s^{-1}, corresponds to (2.5 \pm 0.4)% of Crab Nebula units. Only an upper limit, of 1.9% of Crab Nebula units above 300 GeV, was obtained with the 2008 data. This, together with strong hints (>3sigma) of flares in the middle of October and November 2009, implies that the emission is variable. The MAGIC results favour a scenario with the very high energy emission originating from the inner jet close to the central engine. More complicated models than a simple one-zone SSC scenario, e.g. multi-zone SSC, external Compton or hadronic, may be required to explain the very flat spectrum and its extension over more than three orders of magnitude in energy.; Comment: 6 pages, 5 figures...

We construct a systematic survey of extragalactic \gamma-ray sky at the energies above 100 GeV using the data of Fermi telescope. Such survey has not been previously done by the ground-based Cherenkov gamma-ray telescopes which have, contrary to Fermi, narrow field of view. We study a map of arrival directions of the highest energy photons detected by Fermi at Galactic latitudes |b| > 10 degrees and search for significant point source like excesses above the diffuse Galactic and extragalactic \gamma-ray backgrounds. We identify eight significant point source like excesses in this map. Seven of the eight sources are known TeV blazars. The previously unknown source is identified with a head-tail radio galaxy IC 310, situated in Perseus cluster of galaxies. The source is detected with significance 6 sigma above 30 GeV. We identify two possible scenaria for gamma-ray emission from this source. One possibility is that emission originates from the base of relativistic outflow from the active nucleus, as in the BL Lacs and FR I type radio galaxies. Otherwise gamma-ray photons could be produced at the bow shock formed in result of fast motion of the galaxy through the intracluster medium. The pp -> pp gamma reaction has been studied at a beam energy of 310 MeV by detecting both final protons at the PROMICE-WASA facility and identifying the photon through the resulting missing-mass peak. The photon angular distribution in the center-of-mass system and those of the proton-proton relative momentum with respect to the beam direction and to that of the recoil photon were determined reliably up to a final pp excitation energy of Epp ~ 30 MeV. Except for very small Epp values, the behavior of these distributions with excitation energy is well reproduced by a new refined model of the hard bremsstrahlung process. The model reproduces absolutely the total cross section and its energy dependence to within the experimental and theoretical uncertainties.; Comment: 12 pages with 11 figures. Differential cross sections for positive pions, protons, and neutrons resulting from inelastic π--p collisions have been measured from 310- to 454-MeV incident-pion kinetic energy. The data were obtained with electronic counter systems, which measured the energy distribution of the final-state particle of interest at a series of fixed angles. The results have been interpreted in terms of the final states π+π-n, π0π0n, and π-π0p. The total cross sections for these three modes as a function of incident pion energy are in qualitative agreement with the predictions by Schnitzer. A preference is shown for his set of π-π scattering lengths; a0=0.65, a1=0.07, and a2=-0.14μ-1. The observed neutron distributions correspond to a strong preference for low c.m.-system neutron energies in both the π+π-n and π0π0n final states. The effect is not present in the observed proton distributions from the π-π0p reaction, which suggests that it is due to a I=0, π-π interaction. The π+ data show the formation of the (3,3) isobar combination of the π--n system in the π+π-n final state. Analysis in terms of an isobar model indicates the predominance of I=1/2 incident state.

The goal of this work was to provide the foundation for a course in networking by developing the hardware and software for a token ring Local Area Network (LAN). Any course work in networking would focus on the emerging International Standards Organization (ISO) Open Systems Interconnections (OSI) model. This model provides a seven layered structure, where each layer performs some logical aspect of the communication process. Thus, the software developed would have to provide the functionality of a multiple layered design, implying multiple tasks communicating by operating system primitives. Also, a simplified design would minimize the time for those students trying to assimilate the basic work prior to embarking on the design of their own work. Within an Intel 310 system running the iRMX operating system, the LAN hardware and software were developed. The package allows reliable transfer of files to the other nodes on a token ring LAN via access by user programs to lower layers. In addition, the user has a reasonable interface to interact with the file system of the home node. Once the physical configuration of the LAN is established, the software is self-initializing and self monitoring for pathological cases. The software runs as an application task on each node...
