Many human traits and diseases have a polygenic architecture, where phenotype is partially determined by variation in many genes. These complex traits or diseases can be highly heritable and genome-wide association studies (GWAS) have been relatively successful in the identification of associated variants. However, these variants typically do not account for most of the heritability and thus, the genetic architecture remains uncertain.

More recently, leaf diseases attributable to virus, other fungi and possibly a bacterium were well recorded as a new disase of the rubber tree. In the state of Amazonas, symptoms of a systemic infection caused by virus (being identified), typified by leaf mosaic, little leaf, malformation and tree stunt were confirmed in clonal nurseries and field plantings in several clones at CNPSD. In tests, the symptoms of virus appeared more distinct at temperatures below 22oC. The new fungal leaf pathogens discovered in the nurseries and fields are Corynespora cassiicola, Periconia manihoticola (previously of minor importance), and an Ascomycete (Dothideaceae) in association with Phyllachora huberi yet to be identified. Also discovered were leaf symptoms in the form of tiny lesions, followed by premature leaf yellowing and fall, suggestive of a bacterial infection. These were reproduced in artificial inoculation with a culture of the bacterium repeatedly isolated from fields of clones Fx 3864, PFB 5, IAN 873 and PA 31. Relating these potentially dangerous new diseases to the welll-being of the rubber industry worldwide, international quarantine measures adopted today against Microcyclus ulei alone must now be reviewed. These should include additional measures for the inspection...

This dissertation in biometeorology has the objective of analyzing the influence of some meteorological elements on the behavior of some infectious diseases in the State of Alagoas, through an observational study, in an attempt to better understand their endemic behaviors and some environmental factors that can trigger them. The research was done in three cities of the State, situated in tree different regions: Maceió (litoral); Palmeira dos Índios (agreste) and Água Branca (sertão). Then a crossing was done between series of data of monthly number of cases of schistosomiasis, dengue and cholera, and meteorological data of precipitation, monthly average temperature, average maximum temperature, and average minimum temperature, during the period from 2002 to 2005. The data used were provided by the Secretaria Estadual de Saude de Alagoas and the Insituto Nacional de Meteorologia / 3o Distrito. The graphs obtained from these series of data were then analyzed, together with statistical techniques, in an attempt to find correlation coefficients (r) that are representative of the influence (or lack of influence) of meteorological factors on the endemic distribution of the diseases in the State of Alagoas. A low incidence of cholera was observed in the State...

This thesis is a continuation of a research project started with a diagnosis of dermatosis mechano-bullosa in a herd of buffaloes from a farm in southern Rio Grande do Sul. After this diagnostic it was created an experimental herd where several congenital defects and / or hereditary disorders have been diagnosed during more than two decades. These diseases were studied by a research group which the author of this thesis is a member. Thus, three papers are presented: the first one is a literature review about what has been diagnosed in Brazil on congenital defects and hereditary diseases in buffalo. It was concluded that undesirable genes are widespread in the population of buffaloes in the country; the second was a study conducted in partnership with UNESP / Jaboticabal, SP and UFCG / CSTR-Patos, PB to identify the mutation in the gene that determines the oculo-cutaneous albinism in Murrah buffalo. Another paper was accomplished to describe the occurrence of malignant melanoma in two albino buffalo. Although it is not a congenital or hereditary disease it was observed only in the buffalo with oculocutaneous albinism and probably the condition is associated with a predisposition of these animals to develop tumors in the skin.; Esta tese d?? continuidade a um projeto de pesquisa iniciado com o diagn??stico de dermatose mecanobolhosa em 1985 em um rebanho de b??falos pertencente a uma propriedade da zona sul do Rio Grande do Sul. This study uses raw data covering over 17,000 people from the 2001 National Socio-Economic Survey (NSES) and 2001 National Household Health Survey (NHHS), including 3621 children under 10 years of age, to investigate the relationship between respiratory diseases and exposure to secondhand cigarette smoke through living in a home where people smoke. An important finding is that children under 10 years of age who live in homes where 30 or more cigarettes are smoked each day are significantly more likely to have various respiratory diseases than children who live in smoke-free homes.

Food provides a link between the population health and climate stabilisation agendas. This paper argues that a broader view of food security for the 21st century in Australia and internationally is needed – one that judges the food system for its nutritional quality, social value and impact on the environment. If done well, climate change mitigation and adaptation policies provide ways to achieve this. This paper focuses on mitigation strategies, and describes how the reduction of greenhouse gas emissions from the agricultural sector through a reduction in consumption of animal source foods can improve food security and reduce the levels of chronic diseases such as cardiovascular diseases and some cancers.

In this thesis, I present selected works I conducted in my Master of Philosophy Applied Epidemiology (MAE) placement at the Vaccine Preventable Disease Surveillance (VPDS) Section of the Office of Health Protection (OHP), at the Australian Government Department of Health from March 2013 to November 2014. The works presented comprise my MAE requirements and describe my experiences as an MAE. I discuss my role in the day-the-day activities of the VPDS section, including the surveillance of notifiable diseases, being the secretariat for the rotavirus working group, writing annual reports and editing Communicable Disease Intelligence Journal submissions. I also describe my role as a Watch Officer for the National Incident Room within the OHP. I investigated a foodborne outbreak of gastroenteritis at a Mother’s Day buffet luncheon in the Australian Capital Territory (ACT), where curried prawns and Caesar salad were the suspected cause of the outbreak. The investigation was unable to determine the aetiological cause of illness but highlighted the risk often associated with serving buffet style meals. I present two epidemiological studies. The first is an analysis of notified Legionella infections from 2001 and 2012. Describing the epidemiology of legionellosis in Australia...

In 1992, a mail survey was conducted among South Australians aged 10 years and older to assess the knowledge of prevention of dental caries and gum diseases and to explore its variation by sociodemographic factors. The survey provided 838 completed questionnaires. Questions were asked on perceptions of importance of a number of preventive measures, the main purpose of water fluoridation and sources of information. Respondents rated four myths for preventing dental caries as the most important: 97 per cent rated regular tooth brushing; 87 per cent rated regular dental visits; 85 per cent rated calcium in the diet; and 78 per cent rated eating fibrous foods as definitely or probably important. Only 56 per cent of respondents rated drinking water with fluoride as definitely or probably important for preventing dental caries, and only half (50.2 per cent) identified the main purpose of water fluoridation as the prevention of decay. Respondents rated regular tooth brushing (96 per cent) and regular dental visits (87 per cent) as important for the prevention of gum diseases. However, the myth of massaging the gums was rated as important by 67 per cent. Higher percentages of females, older adults and those with lower educational attainment rated the myths for preventing caries as important. Younger people were less able to specify the main purpose for fluoridation of water supplies. The persistence of myths and the low rating of the importance of scientifically efficacious measures are major challenges for oral health promotion.; Kaye F. Roberts Thomson...

Patients with lysosomal storage diseases (LSDs) have a greatly diminished lifespan and reduced quality of life, particularly those with neurological manifestations. There are few therapeutic options available to treat the neurological signs and symptoms of LSDs. It is, therefore, imperative that efficacious and tolerable treatments are developed. Hematopoietic stem cell transplantation is carried out in some LSDs in which there is neurological involvement. However, this approach is associated with significant morbidity and mortality, and not all patients who receive this treatment exhibit improvements in cognitive signs and symptoms. A growing body of research in animal models of LSDs appears to support the efficacy of repeated delivery of recombinant lysosomal proteins via injection into the cerebrospinal fluid (CSF). Studies in dogs with mucopolysaccharidosis (MPS) Type 1 have shown that this approach enables widespread distribution of the recombinant protein within the brain, leading to a reduction in LSD pathology. Subsequent studies in MPS IIIA mice revealed that this strategy was also effective in ameliorating neuropathology and improving clinical signs in these animals. More recent studies in mice with Krabbe disease or a late infantile form of neuronal ceroid lipofuscinosis have demonstrated that delivery of recombinant proteins into the CSF may be efficacious in reducing disease pathology and neurological signs and symptoms. Whilst there are still important issues that need to be addressed...

The pharmacological application of intestinal growth factors has been recognized because of the protective and reparative actions of these factors in the intestinal tract. This review highlights the use of keratinocyte growth factor (KGF), IGF-1 and glucagon-like peptide 2 (GLP-2) in efficacy studies of intestinal damage; the results from these studies support potential clinical applications of these factors in treating intestinal diseases. In particular, GLP-2 has been assessed in preclinical and clinical investigations for its capacity to prevent or treat an increasing number of intestinal diseases, including short bowel syndrome, chemotherapy-induced intestinal mucositis and inflammatory bowel disease.; http://www.ncbi.nlm.nih.gov/pubmed/20336592; R. PURPOSE: This study aims to examine whether specific anxiety disorder comorbidity alters the purported association between depression and specific cardiovascular diseases (CVDs). METHODS: In 4,181 representative German participants of the general population, 12-month prevalence of psychiatric disorders was assessed through the Composite International Diagnostic Interview and CVDs by physician verified diagnosis. Adjusting for conventional risk factors logistic regression analyzed the association between CVDs (peripheral vascular disease (PVD), hypertension, cerebrovascular disease and heart disease) and combinations of comorbidity between depression and anxiety disorder types (panic disorder, specific phobia, social phobia and generalized anxiety). RESULTS: There were 770 cases of hypertension (18.4 %), 763 cases of cerebrovascular disease (18.2 %), 748 cases of PVD (17.9 %), and 1,087 cases of CVD (26.0 %). In adjusted analyses phobia comorbid with depression was associated with cerebrovascular disease (odds ratio (OR) 1.61; 95 % confidence interval (CI) 1.04-2.50) as was panic disorder (OR 2.89; 95 % CI 1.47-5.69). PVD was significantly associated with panic disorder (adjusted OR 2.97; 95 % CI 1.55-5.69). Panic disorder was associated with CVDs (adjusted OR 2.28; 95 % CI 1.09-4.77) as was phobia (adjusted OR 1.35; 95 % CI 1.04-1.78). CONCLUSIONS: Classification of anxiety and depression according to comorbidity groups showed discrete effects for panic disorder and specific phobia with CVDs...

Treatment of neurodegenerative diseases by classical pharmacotherapy is restricted by blood-brain barrier which prevents access to the brain of potentially therapeutic molecules. Recent progress in the knowledge of pathophysiological molecular processes, and in the development of molecular biotechnology have opened the way to new therapeutic interventions for these disorders. This chapter reviews the most recent gene therapy strategies using experimental models for neurodegenerative diseases.

INTRODUCTION: Cardiovascular diseases are the leading cause of death and hospital expenses in the state of Mato Grosso, Brazil. OBJECTIVE: To describe the temporal variation on risk and protective factors for cardiovascular diseases in adults (≥18 years old). METHODS: Data were obtained from population-based telephone surveys carried out between 2006 and 2009 in Cuiabá, the capital of Mato Grosso, Brazil. The prevalence of smoking, alcohol abuse, markers of diet quality consumption, physical activities, leisure, domestic work, and commute was estimated. Statistical significance of temporal variations in these estimators was estimated by logistic regression models, whose dependent variables were the analyzed risk factors (absent = 0; present = 1) and independent variable was the year of the survey. RESULTS: There was an increase in the proportion of men who reported to never have smoked (from 53 to 61%, p = 0.04). The frequency of regular soft drink consumption was reduced by 23% (p < 0.01). Overall, the reference to the consumption of fruits and vegetables increased by 19% (p = 0.02), and among men this increase was clearer (49%; p < 0.01). There was a reduction in domestic work (p < 0.01) and in leisure physical activity among women (from 14 to 10%; p = 0.02). Conversely...

This series evaluates the effectiveness of community-based interventions (CBIs) to prevent and control infectious diseases of poverty (IDoP). Evidence from our reviews suggests that CBIs and school-based delivery platforms are effective in averting risk behaviors and reducing the disease burden. Co-implementation of interventions through existing community-based programs including immunization campaigns, antenatal care and maternal and child health programs have the potential to scale-up interventions for IDoP. Future research should focus on the process of developing and implementing efficient community-based programs through a comprehensive approach, and to gauge the effectiveness of various existing delivery models in order to improve morbidity and mortality outcomes.; Zulfiqar A Bhutta, Rehana A Salam, Jai K Das and Zohra S Lassi; Multilingual abstracts in the six official working languages of the United Nations are online as an additional file.

OBJECTIVE: The purpose of this study was to investigate the prevalence and identify factors associated with simultaneous risk behaviors for chronic noncommunicable diseases in adults in a southern capital in Brazil. METHOD: A cross-sectional, population-based study was carried out with 1720 adults in Florianópolis, Brazil. The simultaneous occurrence of tobacco smoking, abusive drinking, inadequate or unhealthy diet, and physical inactivity during leisure was assessed. The independent variables were demographic and socioeconomic characteristics. RESULTS: Only 8.3% of the respondents did not have any of these factors, whereas the simultaneous occurrence of two or more risk behaviors was 59.4%. The simultaneous presence of four risk behaviors (3.4%) was 220% higher of what would be expected by combining the individual prevalence of these factors (1.5%). The likelihood of individuals having two or more risk behaviors simultaneously was greater in young men, with black skin color, living without a partner, with lower household per capita income, and lower education. CONCLUSION: It is necessary to implement programs that reduce the risk behaviors for chronic noncommunicable diseases among adults in Brazil, especially between young men with low education and income.; Diego A.S.
